Academic Journal of Second Military Medical University 2016;37(6):690-693
Objective To study the pharmacokinetics and bioequivalence of asparaginase (Asp) hyaluronic acid-graft-poly (ethylene glycol)/dimethyl-β-cyclodextrin (DCD) nanocapsules (AHDPs) in male Sprague-Dawley (SD) rats. Methods AHDPs were observed under the transmission electron microscope. The size, zeta potential and entrapment efficiency of AHDPs were examined. Asp activities were assayed after intravenous injection of AHDPs or free Asp in rats. Pharmacokinetic parameters were calculated by software DAS 2.1.1. Then the bioequivalence of AHDPs and free Asp were evaluated. Results The average particle size of AHDPs was (439.63±8.49) nm, zeta potential was (-20.43±2.20) mV, and entrapment efficiency was (55.75±4.11)% (n=3). AUC(0-48 h) of AHDPs and free Asp were (138.93±0.89) U•mL-1•h and (46.38±1.98) U•mL-1•h, the AUC0-∞ were (175.22±13.59) U•mL-1•h and (51.44±3.01) U•mL-1•h, and t1/2 was (4.46±1.04) h and (1.86±0.38) h, respectively. Compared with free Asp, the AUC0-48 h, AUC0-∞ and t1/2 of AHDPs were increased by 3.00, 3.40 and 2.40 times, respectively. The 90% confidential intervals of AUC0-48 h, AUC0-∞ and Cmax were 76.9%-78.3%, 76.9%-78.3% and 92.8%-94.4%, respectively. Conclusion AHDPs can prolong the biological half-life and improve the bioavailability of Asp in rats. AHDPs and free Asp are not bioequivalent.

Academic Journal of Second Military Medical University 2016;37(9):1076-1081
Objective To prepare hyaluronic acid-nricase liposomes (UHLP) and to investigate the activity and stability of UHLP and uricase (UC) in vitro. Methods UHLP was prepared using reverse-phase evaporation and observed by transmission electron microscopy. The entrapment efficiency, particle size and zeta potential of UHLP were detected. The physical and chemical properties of UC in free UC and UHLP-including the optimum pi I and temperature-thermal stability-storage stability, pi I stability, stability to trypsinase, and stability to metal ions and organic compounds, were examined. In addition, the mechanism by which UHLP promotes UC activity was also explored. Results The mean entrapment efficiency of UIILP was (57. 2 ± 3. 93)% (n = 3), mean particle size was (322. 6 ± 8. 2) nm (n=3), and mean zeta potential was (–19.4 ± 1.7) mV (n = 3). UHLP was round or oval in shape and was evenly distributed under transmission electron microscopy. The optimum temperature of UC in UHLP and free UC was 40°C -and the optimum pH values of UC in UHLP and free UC was 8. 0 and 8. 5, respectively. Thermal stability, storage stability, pi I stability, stability to trypsin and stability to metal ions and organic compounds of UC in UHLP were better than those in free UC. The envelopment of UC with UHLP increased the activity of UC by reversing conformation and exposing active center of UC. Conclusion UHLP can not only enhance the activity but also improve the stability of UC in vitro.

Objective To optimize the extraction process of volatile oil and inclusion preparation of hydroxypropyl-β-cyclodextrin from compoundJinling Sini Siwu Shixiaosan Granules. Methods The orthogonal approach of L9 (34) was applied to investigate the volume of volatile oil extraction by 3 factors to obtain the optimum extraction conditions. With inclusion rate of volatile oil and yield of inclusion compound as comprehensive evaluation indexes, the optimum preparation conditions were discussed. The characterization of this prepared inclusion was analyzed by TLC.Results The optimum extraction conditions were as follows:the crude medicine with medicinal powder was shattered into 20 meshes;8 times the amount of water was added and was extracted for 8 h. The best inclusion conditions were as follows:the proportion of volatile oil to HP-β-CD was 1∶6, with inclusion temperature of 30℃, grinding time of 2 h. Analysis results of TLC showed that stable inclusion compound was formed from volatile oil and HP-β-CD.Conclusion The optimized extraction process and inclusion preparation are with stable quality and high feasibility.

Objective To estimate curative effect of reconstruction of rabbit knee joint cartilage defect with the homogeneitic tissue engineered cartilages. Methods The chondrocytes were isolated and collected from articular cartilages of eight New Zealand white rabbits. The tissue engineered cartilages after culturing chondrocytes and atelocollogen for two days.Cartilage defects were created in both keen joint of twenty-six rab- bits.Complexes of chodrocytes and atelocollagen was grafted into the defect of left knee joint at once as experi- mental group, and no implantation were served as control.General and histological examination were respec- tively performed in both group at four weeks and eight weeks after surgery. Results After implantation, the defects were filled with cartilaginous tissue in experiment group, while there were only tissue in control group. Histologically, defective areas were filled with chondrocytes in experiment group, but only fibroblast in control group. Conclusion The implantation of the tissue engineered cartilages contenting with chondrocytes and atelocollogen can effectively improve reconstruction of rabbit knee joint.

This study was aimed to investigate the comprehensive ecological factors of Hippophae rhamnoides L. and their regional suitability in China. Based on field survey, specimen examination and literature investigation, ecologi-cal factors and appropriate production areas were analyzed by Traditional Chinese Medicine Geographic Information System (TCMGIS-II). The results showed that the proper region (with similarity of 95%~100%) of H. rhamnoides L. accounts for 737 994.71 km2, including 15 provinces/municipalities and 387 counties/cities. The largest area among them is Tibet autonomous region with area of 313 857.73 km2 (42.53%), followed by Sichuan province (223 987.02 km2, 30.35%), Gansu province (66 314.43 km2, 8.99%) and Shanxi province (4 237.79 km2, 0.57%). There are also certain appropriate production areas distributed in Liaoning province, Beijing, Chongqing and Hubei province. It was concluded that this system is much valuable to the recognition of the formation of the producing area, the division of adaptive area, introduction and acclimatization of medicinal materials. It also provided a scientific reference for the introduction and cultivation of H. rhamnoides L. Through further field study and experiments, these new areas have the potential to be developed into suitable production region of H. rhamnoides L. in the future.

Objective To develop a new isotope dilution gas chromatography mass spectrometry method (ID/GC/MS) for the measurement of serum cholesterol. Methods Serum was mixed with an isotope labeled internal standard ([3,4-~(13)C]-cholesterol) and treated with alcoholic sodium hydroxide to hydrolyze cholesterol ester to cholesterol.Cholesterol and internal standard was extracted and derived by N, O-Bis(trimethylsilyl) trifluoroacetamide to trimethylsilyl ethers. The derivation products were analyzed by capillary column GC combined with electron impact MS using scan and selected ion monitor (SIM) modes. Signals of cholesterol internal standard were corrected for the contributions from cholesterol and the signal ratio of cholesterol to internal standard for the calibrators were linearly regressed against cholesterol concentrations. The resulted regression equation was used for the calculation of serum cholesterol concentrations. Results The new ID/GC/MS method showed a mean within-run coefficient variance (CV) of 0.04%-0.81%.Comparison with two levels of standard reference material (SRM1951a) of National Institute of Standards and Technology (NIST) displayed a bias of 0.19% and 0.90% respectively. Conclusion A time-gaining ID/MS method has been established that is highly precise and accurate and can be used for the measurement of serum cholesterol.
