1.Growth inhibition and apoptosis induced by oridonin combined with valproic acid in HL-60 cells

Objective To investigate the apoptosis-inducing effect of oridonin combined with valproic acid( VPA)on leukemic cell line HL-60,and study the feasibility of oridonin combined with VPA to be used in clinical practice. Methods Oridonin of 6-12 μmol/L combined with VPA of 0. 5-1 mmol/L were added in exponential growth HL-60 cells respectively. Cell count assays were used to measure the growth inhibitory effect of oridonin combined with VPA or alone. Flow cytometry was used to evaluate apoptosis with Annexin V and propidium iodide (PI) double staining. Results Combined use of oridonin and VPA could synergistically inactivate HL-60 cells,and inhibit the cell proliferation and induce apoptosis in a dose-and time-dependent manner (P < 0.05 ). Conclusion Oridonin has a synergistic effect combined with VPA. Oridonin has a promising prospect in clinical use of leukemia.

3.Current status of bariatric surgery for type 2 diabetes management

Although weight loss is beneficial for obese patients with diabetes, current medical treatment for obesity is difficult. Bariatric surgery leads to substantial and durable weight reduction.Nearly 30% of patients who undergo bariatric surgery have type 2 diabetes.For many of them, diabetes resolves following bariatric surgery (83% to 93% for bypass procedures and 55% to 84% for restrictive procedures).This review discusses the effects of various bariatric procedures on type 2 diabetes mellitus and their possible mechanisms.
5.Construction of pDsVEGF165Red1-N1 and pIRES2-BMP2-EGFP and their co-expression using RFP and EGFP as reporter gene in HEK 293-T cells

BACKGROUND: It has been proved that vascular endothelial growth factor 165(VEGF165) and bone morphogenetic protein 2(BMP2) can accelerate the vascularization synergistically.OBJECTIVE:To construct the vectors, pDsVEGF165Red1-N1 and pIRES2-BMP2-enhanced green fluorescent protein(EGFP) followed by co-transfected into HEK 293-T cells,and study their expression and location of VEGF165and BMP2 in the cells.DESIGN: A randomized and controlled experiment.SETTING: National Key Laboratory,Institute of Surgery Research,Daping Hospital,Third Military Medical University.MATERIALS: The experiment was conducted at National Key Laboratory,Institute of Surgery Research,Daping Hospital,Third Military Medical University from September 2002 to March 2004. pcDNA3.1\BMP2 ( gift of Dr.Bostrom, UCLA School of Mediine, Los Angeles,USA).pDsRed1-N1(gift of Pro. Roger Y.Tisen,University of California,San Diego,USA). pUC18/VEGF165,293-T cells(preserved by our Laboratory).METHODS: According to the nucleotide sequence of hVEGF165, the primers were designed.The hVEGF165 gene without stop codon was amplified by polymerase chain reaction (PCR).The fragment digested was cloned into the expression vector pDsRed1-N1.Meantime,the pIRES2-BMP2-EGFP expression vector was constructed.The two plasmids were co-transfected into HEK 293-T cells.The co-expression and distribution of the VEGF165and BMP2 were observed with confocal laser microscopy (CLSM) and detected by reverse transcription-polymerase chain reaction (RT-PCR) and Western Blotting.MAIN OUTCOME MEASURES: Identification of the recombinant plasmids and the expressing mRNA and protein in 293-T cells.RESULTS: The recombinant plasmids were verified correct construction by restriction enzyme analysis, PCR and sequencing. The two genes which were co-transfected could express in HEK 293-T cells.The co-expression of the report genes,RFP and EGFP, were found over the cytoplasm and in the nuclei by CLSM.CONCLUSION: Two report gene expression vectors contain VEGF165 and BMP2 have been constructed successfully, which can be co-expressed in HEK 293-T cells. Thus, they can provide important and convenient tool to study intracellular interaction of VEGF165 and BMP2.
6.Determination of five triterpenic acids from Salvia chinensis of different parts by RP-HPLC-PDA.

OBJECTIVETo establish a RP-HPLC-PDA method for determination of five triterpenic acids (pomolic acid, hawthorn acid, corosolic acid, oleanolic acid and ursolic acid) in different parts of Salvia chinensis.
METHODThe isocratic elution and separation was achieved on a Kromasil C18 column (4.6 mm x 250 mm, 5 microm), using acetonitrile-water (90:10) as the mobile phase at a flow rate of 0.8 mL x min(-1). The detection wavelength and column temperature were set at 205 nm and 28 degrees C, respectively.
RESULTThe calibration curves of pomolic acid, hawthorn acid, corosolic acid, oleanolic acid and ursolic acid were linear over the ranges of 0.096 0-2.400, 0.1230-3.075, 0.2420-6.050, 0.2830-7.075 and 0.2730-6.825 microg (r = 0.9998, 0.9997, 0.9999, 0.9995, 0.9999), respectively. The average recoveries were 98.43%, 98.13%, 100.6%, 98.19% and 99.15%%, respectively, with RSD (n=6) being 1.3%, 0.67%, 1.2%, 0.87% and 0.43%.
CONCLUSIONThe proposed method is so simple and highly reproducible that it promises to be applicable for determination of major triterpenic acids in S. chinensis.

7.Regional radical resection combined with vessel resection and reconstruction (Fortner type Ⅰ、Ⅱ) for the treatment of pancreatic cancer

ObjectiveTo evaluate the effect of regional radical pancraticoduodenectomy (PD) combined with resection and reconstruction of invaded blood vessel (Fortner type Ⅰ?Ⅱ procedure) for pancreatic cancer.MethodsResult of 5 Fortner Ⅰ cases undergoing PD and resection of superior mesenteric-portal vein (SMPV),and 1 FortnerⅡ case with resection of SMPV,superior mesenteric artery(SMA)and hepatic artery(HA) was reviewed. ResultThe case undergoing Fortner Ⅱ surgery survived for 13 months.The 5 Fortner Ⅰ cases were followed-up for 5~34 months,with patent graft as identified by CT and ultrasonography and with no recurrence. ConclusionRegional pancreatectomy combined with resection of invaded blood vessel can be carried out in carefully selected patients of pancreatic cancer with favourable long term-result.
8.Correlation between polymorphism in the promoter of DNA methyltransferase-3B gene and susceptibility of colorectal cancer

Objective To observe and analyze the correlation between the DNA methyltransferase-3B (DNMT3B) gene single nucleotide polymorphisms (SNP) rs 1569686 and the susceptibility to colorectal cancer (CRC) in Tianjin Han population.Methods A case-control study was performed.89 CRC patients and 94 normal controls were examined,and Sequenom MassArray system was applied to detect the genotype of rs1569686 in DNMT3B gene polymorphisms.The association results were analyzed of the SNP and CRC.Results The frequencies of genotypes TT,GT and GG at SNP rs1569686 were 74.2%,24.7% and 1.1% in CRC patients respectively,which there were no significant differences from that in controls(73.4%,23.4% and 3.2%; P>0.05).Conclusion It suggests that the SNP rs1569686 of DNMT3B gene may not be related to susceptibility of CRC in Tianjin Han population.
10.Effects of angiostatic agents on the endometriosis-like lesions in a chicken chorioallantoic membrane

Objective A prospective study was performed to determine the effects of angiostatic agents on endometriosis. Methods Eutopic endometrium from endometriosis was transplanted to a non-vascular region of a chick embryo chorioallantocic membrane (CAM), and angiostatic compounds including recombinant human endostatin, thalidomide, monoclonal antibody of integrin ?v?3 and VEGF were given, their effects on the angiogenesis and on endometriosis-like lesion formation in CAM model were examined. Results Transplantation of endometrium onto the CAM led to a strong angiogenic response in the chick embryonic tissue; a number of newly formed vessels were significantly decreased in the groups using angiostatic agents compared with the control group. Endometriosis-like lesion formation was significantly impaired after treatment with angiostatic agents, and it was associated with decreased vessel density in the surrounding chorioallantoic membrane, accomponied by various degree of necrosis in the endometriosis-like lesions under microscopic observation. Conclusion Endometrium induced angiogenic response in the ectopic environment is critical for implantation and subsequent survival of the tissue. Angiostatic agents can inhibit the angiogenesis induced by eutopic endometrium and has a remarkable anti-endometriosis effect.
