1.The effects of siRNA-mediated Plk1 depletion on COS-7 cells

Objective:To investigate the effects of siRNA-mediated Plk1 depletion on COS-7 cells and the therapeutic potential of Plk1 in cancer treatment.Methods:Small interfering RNA(siRNA) were used to specifically deplete Plk1 in COS-7 cells.RT-PCR and immol/Lunoblotting analysis were used to verify the inhibitory effect of siRNA against Plk1.Trypan blue exclusion assay and FACS analysis were conducted to determine the effects of Plk1 depletion on the growth and cell cycle profile of COS-7 cells.Results:Synthetic siRNA duplexes against Plk1 were introduced into COS-7 cells,which subsequently resulted in a significant inhibition in Plk1 expression in the cells.Cell growth assay indicated that siRNA-mediated Plk1 depletion significantly inhibited the proliferation of COS-7 cells.FACS analysis revealed that Plk1 depletion caused cell cycle arrest,resulting into large population of sub-G1 cells.Conclusion:siRNA-mediated Plk1 depletion significantly inhibited the proliferation of COS-7 cells suggesting the therapeutic potential of Plk1 in cancer treatment.

4.Depth of counseling process:exploring of a three-dimension model

Objective:To test Jiang Guangrong's(2005)three-dimension model on the depth of counseling process.Methods:In this process-outcome research,28 sessions in a university counseling center were videotaped and coded in terms of the model.Structure Equation Model(SEM)was used to examine the relations between the coding and counselors and clients' evaluation of the session depth.Results:(1)Clients and counselors evaluated differently on the session depth,and clients' evaluation was more positive than counselors' [(4.95±1.12)vs.(4.23±0.94),P<0.05];(2)The model was significantly correlated with counselors' session depth evaluation(path coefficient = 0.55,P<0.01),but not with the clients';(3)The session curves describing each dimension changing over time had different patterns in sessions of different depth level,through which more about the counseling process could be known.Conclusion:The three-dimension model on the depth of counseling process has got some empirical evidences.

Acute promyelocytic leukemia (APL) is a subtype of acute myeloid leukemia,which is rare in children.The treatment of APL mainly includes anthracycline-based chemotherapy,all-trans retinoic acid,and arsenic trioxide,which allow for complete remission,improve the long time survival rate and reduce related toxicity as well.About the optimized treatment,quick breakthroughs have already reached,not only at home in the 80's at the end of the 90's,but also abroad,like National Comprehensive Cancer Network of the United States in 2006-2013.This review summarizes the progress in diagnosis and treatment of children with APL.
7.Measurement of CA125 in serum and ovarian cyst fluid of patients with endometriosis and its clinical significance

Objective To study the relationship between CA125 and endometriosis.Methods We tested the level of CA125 in serum and fluid of ovarian cysts of patients with endometriosis and non-endometriosis,respectively.Results The mean CA125 concentration in the fluid of endometrial cysts of the ovary(ECO) was remarkably higher than that of the serum in ECO group(P0.05).Conclusion The level of CA125 has bearing on the occurrence,development and prognosis of endometriosis,CA125 level is positively correlated with the severity of the disease.Therefore,it is valuable to test serum CA125 level for diagnosing endometriosis.

Objective:To study VEGF-C expression in colon cancer and the relationship of VEGF-C expression with clinicopathological features and prognosis.Methods:VEGF-C expression was examined by immnohistochemical staining in 44 cases of primary colon cancers.To analyze the relation of VEGF-C expression with clinicopathological features and prognosis.Results:Of the 44 cases of colon cancers,19 cases of carcinoma (43.2%) were evaluated to be VEGF-C positive expression and 25 cases of carcinoma (56.8%) were determined to be VEGF-C negative expression.VEGF-C expression was associated with tumor differentiation (P=0.003),nodal status (P=0.002) ,Dukes stage (P=0.001).The median survival time of the patients with positive VEGF-C expression was lower than that of patients with negative VEGF-C expression (P=0.0225).Metastatic status,the nodal status and VEGF-C were found to be significant independent prognostic factors (P=0.021,P=0.0004,P=0.0469,respectively).Conclusion:VEGF-C expression is upregulated in colon cancer.VEGF-C expression plays a important role in nodal metastasis.VEGF-C appears to be a new prognostic factor for the prediction of outcome in patients with colon cancer.
10.Clinical features of sinonasal inverted papilloma with recurrence and malignant transformation.

OBJECTIVE:
To investigate the clinical characteristics, treatment and the effectiveness of sinonasal inverting papilloma with recurrence and malignant transformation.
METHOD:
Retrospective comparative analysis of sinonasal inverting papilloma with recurrence and malignant transformation treated from Jan. 2008 to Oct. 2012 in our hospital.
RESULT:
All the 24 patients with recurrence had the history of sinonasal surgery. Among them, 17 patients recurred once,6 recurred twice and 1 recurred three times. The recurrence time was from 1 month to 14 years after operation. Four patients relapsed within half year after 5 months to 4 years follow-up and the recurrence ratio was 16.7% (4/24). All the 4 patients with recurrence had an operation once again and one of them was given chemoradiotherapy after surgery whose pathological results showed canceration. There was no recurrence among the all of 24 patients followed up until now. Among 5 patients with malignant transformation, 3 patients of them were given radiochemotherapy after surgery. Except 1 patient died of extensive recurrence, 1 received other operation after relapsed twice and 1 lost follow up,all the others were alive without recurrence during 10 months to 4 years.
CONCLUSION
The surgical treatment with completely and positively is the firse choice and an effective management for sinonasal inverting papilloma. To the patients with recurrence on many times and malignant tumors, post-operative radiation or chemotherapy should be considered when needed.
