1.Downregulation of actin related protein 2/3 complex subunit 5 inhibits proliferation, migration and invasion of lung squamous carcinoma cell line SK-MES-1

Background and purpose:Actin related protein 2/3 complex subunit 5 (ARPC5) is involved in the packaging of actin, and then affects the mobility of cells. It plays an important role in the formation of invadopodia of cancer cells. This study aimed to investigate the effect of ARPC5 gene-silence on cell proliferation, migration and invasion of lung squamous carcinoma cell line SK-MES-1. Methods:Under the induction of LipofectamineTM RNAiMAX, the recombinant of si-ARPC5 was transfected into SK-MES-1. The experiment set up 3 groups:blank (mock) group, negative control (NC) group and si-ARPC5 transfected group. Cell proliferation, migration and invasion assays were investigated by cell counting kit(CCK-8) method, scratch assay and Transwell Chambers respectively. Results:The result of qRT-PCR and Western blot displayed downregulation of ARPC5 in mRNA and protein level of SK-MES-1 cell. Proliferation rate of SK-MES-1 cells in 72 h after si-ARPC5 transfection was lower than those in mock and NC groups (t=7.993, t=8.681, P<0.05). Compared to the mock and NC groups, the scratch repairing rate of the si-ARPC5 group was lower [(43.32±0.23)%vs (73.11±0.43)%and (76.58±0.88)%] (t=7.348, t=10.614, P<0.05);And the number of penetrating the membrane was signiifcantly reduced [(27±6) vs (101±11) and (92±9)] (t=10.229, t=8.391,P<0.05). Conclusion:Downregulation of ARPC5 gene can inhibit the proliferation, migration, and invasion of SK-MES-1 cell.

OBJECTIVE:To study the possible mechanism of Bushenshengsui I(BSSS I),a kind of compound Chinese herbs,in treatment of aplastic anaemiaMETHODS:The animal model of mice with aplastic anaemia was established by 60Co irradiationThe therapeutic effects of BSSS I on aplastic anaemia were observed with the changes of peripheral blood cell count,bone marrow GM-CFU-c and activities of blood IL-1,IL-2 and IL-6RESULTS:After treatment with BSSS I,the peripheral blood cell count,amount of bone marrow karyocyte and GM-CFU-c of bone marrow of the aplastic anaemia mice elevated markedly,and the blood IL-1 and IL-6 activities increased obviouslyThe levels of IL-6 increased to the level of normal and showed positive relationship with IL-1(r=0904)The levels of IL-2 of the treatment group showed a decrease but with no significant differencesCONCLUSION:The results showed that BSSS I may improve the function of hematopoiesis by elevating the peripheral blood cells and the amount of bone marrow karyocytes,accelerating GM-CFU-c of bone marrow,and through moderating the activities of IL-1,IL-2,IL-6

Nicotinic acetylcholine receptors concentrate on the postsynaptic membrane in the neuromuscular junction and participate in the neuromuscular transmission,which is of great importance to normal muscle function. The nicotinic acetylcholine receptor is a kind of pentamer.There are 2 types of nicotinic acetylcholine receptor due to the different subunit:fetal type ?-AChR (? 2???) and adult type ?-AChR(? 2???). This article reviews the difference of 2 types of nAChR in expression period,distribution,regulation mechanism and physiological significance.
4.Thinking of Deepening National Essential Medicine System Reform

OBJECTIVE:To provide reference for improving the national essential medicine system and satisfy the needs of ra-tional use from the public METHODS:The current situation and existing problems in the national essential system were described by literature research and comparative analysis,and recommendations were presented. RESULTS:The implementation of essential medicine system has certain effects in China,but there still remains some problems,involving too much variety of essential medi-cines,high price and others,and limitation of the use of medicine and distribution in primary medical institutions. CONCLU-SIONS:Government should further simplify the essential medicine lists,reduce prices,strengthen policy support for primary medi-cal institutions,deepen the national essential medicine system and protect the public's rational drug use.
5.ApoB/ApoA-I Ratio is a Potential Marker to Assess the Severity of Chronic Hepatitis B Virus Infection

Objective To observe the change of the sera lipid profiles in patients with chronic hepatitis B Virus (HBV)infec-tion.Methods Sera from 254 patients suffered from liver disease were collected and divided into four groups according to the severity of the disease (71 of Cirrhosis;80,46,51 of Light,Moderate and Highly severity of the chronic HBV infection, respectively).Sera of 59 healthy patients were collected as control.The concentrations of their sera lipids (CHOL and TG), lipoprotein (HDL and LDL)and apolipoproteins (ApoA-I and ApoB)were determined and compared.Results Compared to the control group,the levels of the CHOL,TG,HDL,LDL,ApoA-I and ApoB were varied in degree in the chronic HBV in-fection group and the cirrhosis group.For the highly severity group and the cirrhosis group,statistics analysis showed de-creased and significantly different lipid and apolipoprotein results when compared to the control group.For the light and moderate severity group,when compared to the control group,levels of TG,HDL and ApoA-I were decreased with remarka-ble difference.The ApoB/ApoA-I ratio of the chronic HBV infection (Highly severity group)was 2.10±1.44,which was significantly higher than that of any other group (P<0.05).Conclusion The ApoB/ApoA-I ratio can be a potential marker for the evaluation of the severity of the chronic HBV infection.
6.A case of infratemporal fossa abscess cause by facial acupuncture.

The infratemporal fossa abscess is an irregularly shaped cavity, situated below and media to the zygomatic arch. It consists of masseter and nervus vascularis, etc. Comparatively speaking, the low position of the infratemporal fossa has almost freed the patient from dental, nasal,ocular and aural source of pain. So facial acupuncture is more likely to cause the infection. But it is also possible that the patient get cryptogenic infection of maxillofacial cavity, the treatment of which needs further study.

9.Clinical significance of neutrophil and albumin pretreatments in nasopharyngeal carcinoma prognosis

Objective:To investigate the association of pretherapeutic neutrophil, lymphocyte, monocyte, and serum albumin levels with nasopharyngeal carcinoma (NPC) prognosis in 401 patients. Methods:This study involved 401 NPC patients recently diagnosed and admitted in Sun Yat-sen University Cancer Center, China. Laboratory variables, such as immunology-related parameters and albumin level prior to treatment, were analyzed. The overall survival and disease-free survival rates were determined using the Kaplan-Meier method and were then compared using the log-rank test. Univariate and multivariate analyses were performed using the Cox proportional hazards model to identify prognostic factors. Results:Patients with neutrophil counts>5.20×109/L (n=172) had significantly lower overall survival and disease-free survival rates compared with those with neutrophil counts≤5.20×109/L (n=229) (P=0.014, P=0.009). In the 401 NPC cases, patients with high albumin levels (>43.00 g/L;n=253) had significantly higher overall survival (P=0.000) and disease-free survival (P=0.006) rates compared with those with low albumin levels (≤43.00 g/L;n=148). Multivariate analysis showed that the neutrophil level is an in-dependent risk factor for disease-free survival, whereas the albumin level is a strong independent prognostic factor for overall survival. Conclusion:The pretherapeutic neutrophil and albumin levels are independent prognostic factors for survival in NPC patients.
