1.Effects of antidepressant treatment on living quality of patients at stable stage of COPD with depression JIANG

Objective To study the effects of antidepressant treatment on living quality of patients at stable stage of COPD with depression. Methods 43 cases at stable stage of COPD with depression were randomly divided into treatment group( n = 22) and control group( n = 21 ). The control group were given appropriate bronchodilators and disperse phlegm with common;the treatment group were given zoleft(50mg, 1 time/d, 6 weeks and 10 weeks) based on the control group. Lung function, nutritional status and quality of life score before and after inpatient treat-ment were determined. Results There was significant difference on lung function, nutritional status and quality of life score between the treatment group before and after inpatient treatment( P < 0.05 ~ P < 0.01 ). There was no significant difference in the control group (P > 0.05 ). Conclusion Antidepressant treatment on patients at stable stage of COPD with depression is very important for improving the quality of life.
2.Biological characters of islets isolated from rats and purified with Dextran T40

Objective To establish a method of isolating and purifying islets from Wistar rat, and to evaluate the biological characters of the isolated and purified islets. Methods The pancreases were surgically removed from adult Wistar rats, carefully minced with scissors as small as possible prior to incubation in 0.9 mg/ml collagenase (Sigma, type Ⅴ) for 10~17min at 37℃. The digested tissue was filtered through a 60 apertures/cm~2 metal sieve and washed with 10% fetal bovine sera (FBS) in Hank’s solution. Purification was performed using Dextran discontinuous density gradient centrifugation to separate the contents of the postcollagenase pellets. Islet samples were stained with DTZ staining for identification. The viability of the purified islet was determined by acridine orange/propidium iodide (AO/PI) double staining. The in vitro function was assayed by different densities of glucose incubating islets for 45min, respectively. Insulin secretion in the supernatant was assayed by radioimmunoassay. Islets were transplanted under the kidney capsule into recipient SD rats that had been rendered diabetic by injecting streptozotocin 50mg/kg, so as to evaluate the in vivo function of islets. Results The islets were recovered between the 11% and 22% Dextran layers, and between the 11% and 1640 layers. The average number of isolated islets was 2181?14 per rat pancreas. After a discontinuous Dextran density gradient purification, about 1826?24 islets were obtained from per rat pancreas, with an average purity over 95%. Islets showed good response to glucose and high efficiency to invert hyperglycemia 7-10d after transplantation. Conclusion These results showed that the digestion methods and purification procedure which we have developed could provide islets from rats for transplantation studies. The biological characters of islets harvested by our method were very well.
3.Detection of soluble serum transferrin receptor of thalassemia women during pregnancy

Objective Soluble serum transferrin receptor (sTfR) is a valuable indicator to diagnose iron deficiency anemia in pregnant women whose indexes will alter because of gestation. Thus it is worth to compare and analyze the levels of sTfR among the pregnant women with thalassemia, healthy pregnant women, and the pregnant women with iron deficiency anemia. Methods ?-thalassemia and ?-thalassemia were detected employing with the percentage of hemolysis and gene assay in the pregnant women randomly selected from those aged 24~30, 20~24 weeks of gestation, and with no other complications. The ratio of OD413nm of HbA2 to OD413nm of HbA was used to distinguish from the two kinds of thalassemia. Then the genetic mutation or deletion of ?-thalassemia patients and nucleotide mutation of ?-thalassemia patients were detected with thalassemia detection kit. The value of sTfR in serum of different samples was measured with ELISA kit. Results 20 pregnant women were diagnosed as thalassemia by the percentage of hemolysis higher than 63%. Among them, 10, 9 and 1 women were judged as ?-thalassemia, ?-thalassemia and ?,?-thalassemia, respectively. 5.Surgical treatment of chronic pancreatitis and choice of procedure

Objective To analyze the results of surgical treatment on chronic pancreatitis(CP),and investigate how to choose the appropriate surgical procedure.Methods The clinical data of 54 patients with chronic pancreatitis who underwent surgery at the First Affiliated Hospital of Harbin Medical University from January 2007 to December 2011 were retrospectively analyzed.Results All the 54 patients underwent surgery,including 8 decompression and drainage procedure (Partington procedure) ; 13 resections (7 cases of pancreaticoduodenectomy,1 case of distal pancreatectomy,4 cases of distal pancreatectomy combined with splenectomy and 1 case of pancreaticoduodenectomy combined with distal pancreatectomy,respectively); 12hybrids (7 cases of Beger procedure and 5 cases of Frey procedure,respectively) and 21 other procedures (15 cases of pancreatic pseudocyst jejunostomy,4 cases of exploratory laparotomy combined with pancreatic tissue biopsy,2 cases of gastrojejunostomy combined with choledochojejunostomy,respectively).There were 4 cases of post-operative pathologic evidence of cancer.Twelve patients had postoperative complications and were cured with non-operative management.Forty-four patients (81.5%) were followed for 2 to 67 months,36 out of 42(85.7%) patients who suffered from abdominal pain had a persistent remission,there were one case of new on-set diabetes and no steatorrhea was reported.Conclusions For CP patients with surgical indications,the choice of procedure should be individualized for the purpose of preserving the endocrine and exocrine functions of pancreas,and taking effectiveness as well as safety into consideration.
6.Determination of free DNA level in plasma of patients with ovarian cancer

7.Association of interleukin-27 gene polymorphisms with genetic susceptibility to systematic lupus erythematosus in Guangxi Zhuang population

Objective To investigate the association between the single nucleotide polymorphisms of interleukin-27 (IL-27) gene and susceptibility to systematic lupus erythematosus (SLE) in Guangxi Zhuang population. Methods In total, 135 patients with SLE and 150 age- and sex-matched human controls of Zhuang nationality were recruited in this study. PCR-restriction fragment length polymorphism (RFLP) analysis and DNA sequencing were performed to analyze the IL-27 gene -964 A/G and -2905 T/G polymorphisms.Results Significant differences were observed in the distribution of IL-27 gene -964 A/G polymorphism (x2 =9.88, P < 0.01 ). The relative risk for SLE in carriers of G allele at position 964 of IL-27 gene was 1.725 times that in carriers of A allele at this position (OR = 1.725,95% CI: 1.227 - 2.425). A significant increase was observed in the frequency of 964G/2905G alleles of IL-27 gene in patients with SLE compared with the controls (10.7% vs. 5.3%, P < 0.01 ), and the 964G/2905G alleles were associated with a significantly increased risk for SLE (OR = 2.351, 95% CI: 1.228 - 4.501 ). Conclusions The IL-27 gene -964 A/G polymorphism is associated with the development of SLE, and the -964 G allele may increase the genetic susceptibility to SLE.
8.Relationship between pelvic floor dysfunction and serum relaxin H2 and expression of vaginal wall relaxin receptor LGR7 mRBA in late pregnant women

Objective To investigate the relationship between pelvic floor dysfunction and serum relaxin H2 and expression of vaginal wall relaxin receptor LGR7 mRNA in late pregnant women. Methods Before the beginning of delivery,all women were evaluated by pelvic organ prolapse quantitation(POP-Q)scoring.Twelve women with stress urinary incontinence(SUI)and stage II prolapse of anterior vaginal wall were selected as patient group,and another 24 women without SUI and prolapse of pelvic floor were served as control group.Serum relaxin H2 was determined by ELISA.Vaginal wall tissues were taken after vaginal delivery,and the expression of relaxin receptor LGR7 mRNA was detected by RT-PCR.Results The serum level of relaxin H2 and expression of LGR7 mRNA of vaginal tissues in patient group were significantly higher than those in control group(P<0.01,P<0.05). Conclusion The increased level of serum relaxin and expression of vaginal wall relaxin receptor may correlate with the pelvic floor dysfunction in late pregnant women.
9.Effect of morphine on synaptic long-term potentiation in spinal dorsal horn evoked by electric stimulation of sciatic nerve in rats

Objective To evaluate the effect of morphine on synaptic long-term potentiation (LTP) in the spinal dorsal horn evoked by electric stimulation of sciatic nerve in rats. Methods Twenty-seven healthy male SD rats aged 60-90 d weighing 180-200 g were randomly divided into 4 groups: group Ⅰ control (group C, n=7), group Ⅱ morphine (group M, n=7), group Ⅲ naloxone (group N, n=6), and group Ⅳ morphine + naloxone (group MN, n=7). The animals were anesthetized with intraperitoneal 10% urethane 1 g/kg, intubated and then mechanically ventilated. The bipolar insulated stainless steel recording electrode (impedance 0.5-1 MΩ, diameter 0.1 mm) was inserted into the left side of the spinal dorsal horn at T13-L1 to stimulate the left side of the sciatic nerve. Single square pulses (15 V, 0.5 ms, 1/60 Hz for 30 min) was applied to evoke spinal field potentials. Normal saline 10 μl, morphine 10 μl (15 μg/μl), naloxone 10 μl (2.5 μg/μl), and the mixture 10 μl of naloxone 5 μl (2.5 μg/μl) and morphine 5 μl (15 μg/μl) was gradually instilled over 2 rain in the 4 groups respectively. Five minutes later, high-frequency and intensity tetanic stimulation (30-40 V, 0.5 ms, 100 Hz, given in 4 trains of 1-s duration at 10-s intervals) was used to induce LTP. Then single square stimuli (15 V, 5 ms, 1/60 Hz) were applied to the sciatic nerve for 210 min. The amplitude and latency period of the field potential were recorded 30 min before tetanic stimulation, and 0-30, 35-60, 65-120 and 125-210 min after titanic stimulation. Results Compared with group C, the amplitude of the field potential was significantly decreased and the latency period prolonged in group M and MN, but there was no significant difference in the above indices between group N and C. Compared with group M, the amplitude of the field potential was significantly increased and the latency period shortened in group MN. Compared with those 30 min before the tetanic stimulation, the amplitude of the field potential was significantly increased and latency period shorted at the time points after the tetanie stimulation in group C and N, the amplitude of the field potential was significantly decreased and latency period prolonged at the time points after the tetanie stimulation in group M, and the amplitude of the field potential was significantly increased 0-30 and 35-60 min after the tetanic stimulation and latency period shortened 0-30 min after the tetanie stimulation, the amplitude of the field potential was significantly decreased and latency period prolonged 65-120 and 125-210 min after the tetanic stimulation in group MN. Conclusion Morphine can inhibit synaptic LTP in the spinal dorsal horn evoked by electric stimulation of sciatic nerve in rats, and it may be one of the mechanisms of the central sensitization inhibition.
10.Expression of peripheral blood CD54,CD106 and CD62p in patients with multi-infarct dementia

Objective:To investigate the expression of peripheral blood CD54,CD106,CD62p in patients with multi-infarct dementia.Methods:The levels of serum CD54,CD106,CD62P of 82 patients with multi-infarct dementia were measured with ELISA,and were compared with 23 normal controls.Results:The levels of serum CD54,CD106,CD62p in patients with multi-infarct dementia(CD54:469?76.33 ng/ml,CD106:1 103.3?98.96 ng/ml,CD62p:18.22?8.90 ng/ml) were higher than those in normal controls(CD54:196?45.91 ng/ml,CD106:601.0?76.30 ng/ml,CD62p:6.70?3.30 ng/ml). There were significant difference between the two groups.The levels of serum CD54,CD106,CD62p were positively correlated with the degree of dmentia.Conclusion:CD54,CD106,CD62p are closely related to the development of MID and play an important role in pathlogical procedure of cerebral damage after MID.The levels of CD54,CD106,CD62p in patients with MID implies the degree of their neurological function deficit scores and might be an important indicator to observe the changes of disease.
