4.Effect of Ganle Keli on the Expression of ?-Smooth Muscle Actin in Hepatic Fibrosis Rats

Objective To observe the change of ?-smooth muscle actin (?-SMA) expression in carbon tetrachloride (CCl4)-induced liver fibrosis in rats after treatment with Ganle keli. Methods Liver fibrosis rat model was induced by CCl4 twice a week for 12 weeks. Ganle keli was used daily intragastric administration from the 7th week. Contents of NO, NOS and iNOS were determined by absorption spectrometry. Liver samples were collected after experiments and stained by immunohistochemistry of ?-SMA. Moreover ?-SMAmRNA expression in liver tissue was detected by RT-PCR technology. Results Ganle keli could significantly reduce NO, NOS and iNOS contents, effectively prevent the expression of ?-SMA, and significantly reduce ?-SMA mRNA in liver fibrosis rats. Conclusion Ganle keli can effectively inhibit the expression of ?-SMA in CCl4-induced fibrotic liver in rats.

6.Surgical treatment of subaortic stenosis (Report of 70 cases)

Objective: To summarize the experience of surgical treatment of subaortic stenosis. Methods: 70 patients with subaortic stenosis were treated surgically. The diagnosis was made by echocardiography, left ventricular catheterization and angiography. There were 46 males and 24 females. The mean age at operation was 11.8 years (range 3 to 46 years). 64 patients had discrete stenosis and 6 had tunnel stenosis. For the discrete stenosis cases, simply resection of stenosis membrane was done in 58 cases and plus myoctomy in 6 cases. For the tunnel stenosis cases, left ventricular muscle was resected to relieve obstruction. In 59 patients combined cardiovascular malformation were corrected at the same time. Results: There were two postoperative deaths. Mitral valve injury occurred in 1 patient. The mean systolic gradient of left ventricle to aorta was 7.5 mmHg with a range of 0-30 mmHg in 38 cases after operation. The 1-, 3-, 5-, 10-, 20-year follow-up rate was 83%, 77%, 68%, 45%, and 9%,respectively. All patients were asymptomatic postoperatively. No reoperation was required. Conclusion: Once subaortic stenosis was diagnosed, operation should be done. Preoperative echocardiography and routine exploration of the root of aorta should be done during operation are in diagnosing. The key point of the operation that left ventricular outflow tract should be thoroughly dredged, no injury should be made to the mitral valve, aortic valve and conduction bundle.
7.The ultrastrucural and immunohistochemical study of BBB in streptozocin induced hyperglycemic rats

Objective To investigate the hyperglycemic effect on ultrastructural morphology and intercellular adhension molecule 1(ICAM 1) expression of BBB. Methods Hyperglycemia model was established by intravenous injection of streptozocin in SD rats. A dynamic observation on the changes of the ultrastructure and the ICAM 1 expression on endothelium of BBB in SD rats with hyperglycemia for 3 days to 8 months was carried out by transmission electron microscopy and immunohistochemical method. Results Swelling was slight in endothelium of BBB of rats with hyperglycemia for 3 days, became obvious and occurred in endfoot of astrocyte for 1 to 2 months, and some microthrombi appeared in cavity of capillary with various malformation and apoptosis was found in endothelium of BBB and neuron surrounding abnormal capillary for 3 to 8 months. With immunohistochemical study, ICAM 1 was expressed sparsely on endothelium of BBB in 3 days after injection of streptozocin, became markedly strong in 3 months, and progressed further in 6 to 8 months. There was no changes on BBB in both SD rats and negative controls. Conclusion Long term hyperglycemia could cause obvious ultrastructural changes of BBB, associated with ICAM 1 expression on endothelium of BBB induced by hyperglycemia
8.Study on immune function of left-to-right shunt congenital heart disease infants combined with pneumonia

Objective To explore the relationship between severity of symptom and immune function in left-to-right shunt congenital heart disease (CHD) infants combined with pneumonia. Methods The clinical data of 45 left-to-right shunt CHD combined with pneumonia infants were retrospectively analyzed. The infants were divided into 2 groups according to the modified Ross score:mild group (0-6 scores, 28 cases) and severe group (7-12 scores, 17 cases);then, the infants were divided into 2 groups according to deformities:single deformity group (the infants had a kind of deformity such as ventricular septal defect, atrial septal defect and patent ductus arteriosus, 21 cases) and composite/complex deformity group (the infants combined 2 or more deformities or other malformations, 24 cases). Twelve cases of healthy check-up infants were selected as control group. The levels of lymphocyte subsets (including CD3+, CD4+, CD8+, CD4+/CD8+, NK cell and B lymphocyte) and IgG, IgA, IgM were detected in every group. Results There were no statistical differences in IgG, IgA, IgM, CD8 +, CD4 +/CD8 +, NK cell and B lymphocyte among mild group, severe group and control group or single deformity group, composite/ complex deformity group and control groups (P>0.05). The CD3+ in severe group was significantly lower than that in mild group and control group (0.59 ± 0.10 vs. 0.69 ± 0.13 and 0.69 ± 0.12), the CD4+was significantly lower than that in control group (0.34 ± 0.07 vs. 0.45 ± 0.09), and there were statistical differences (P<0.05). For CD4+, though there was statistically difference among single deformity group, composite/complex deformity group and control groups (P<0.05), no statistically significant difference was in multiple comparison among the 3 groups (P>0.05). The CD4 + in the composite/ complex deformity group was significantly lower than that in the single deformity group and the control group (0.34 ± 0.07 vs. 0.45 ± 0.15 and 0.45 ± 0.09), and there was statistical difference (P<0.05). Conclusions In left-to-right shunt CHD infants combined with pneumonia, the reduction of CD3+and CD4+may be one of the causes of serious illness.
9.Therapeutic effects of inhaled nitric oxide in patients with pulmonary hypertension after mitral valve replacement

10.Progress on the roles of TGF-βSmad signaling pathway in HIV pathogenesis

Abstract: TGF-β/Smad signaling pathway has a wide range of biological activities and plays an important roles in regulating cell growth, adhesion, differentiation, cell dynamic balance, and immune responses. The higher activity of TGF-β/Smad signaling pathway may promote scar formation, organ fibrosis, immunosuppression, and late-stage cancer progression, while low activity may lead to inflammation, dysplasia, poor healing and oncogenesis. The function of the TGF-β/Smad signaling pathway is extremely complex and can exhibit inhibitory or enhancing effects on immunity and inflammation under different circumstances, but immunosuppressive and anti-inflammatory effects are dominant. During HIV infection, the TGF-β/Smad signaling pathway interacts with HIV in a complex manner as HIV proteins tat and gp120 can induce TGF-β expression. Meanwhile, this signaling pathway may also play a role in HIV infection and replication, latent virus reservoir, host immune deficiency and HIV-related inflammation. It is worth noting that even though TGF-β, which mainly exhibits anti-inflammatory effects, is induced by HIV, high levels of TGF-β do not seem to inhibit HIV-related inflammation. So far, the relationship between TGF-β/Smad signaling pathway and HIV infection has not been elucidated, and its role and mechanism in HIV infection and related illnesses need further exploration and validation. This review summarizes the relevant research progress on the TGF-β/Smad signaling pathway and HIV infection, and provides a reference for further understanding of HIV pathogenesis and exploring strategies of AIDS treatment.
