1.Clinical study of Yiqi-Wenshen decoction combined with salmon calcitonin injection for the patients with bone atrophy after calcaneal fracture

Objective To evaluation of curative effect of Yiqi-Wenshen decoction combined with salmon calcitonin subcutaneous injection for the patients with postoperative bone atrophy after calcaneal fracture.Methods According to the inclusion criteria, 120 patients with calcaneal fracture were randomly divided into 2 groups, 60 in each group. The control group received the subcutaneous injection of salmon calcitonin, and the combined group received the control group treatment plus Yiqi-Wenshen decoction. Both groups were treated for 6 weeks. The X-ray was used to detecte the local bone density of limbs, and the foot function scores of ankle function and the changes of clinical symptoms and signs before and after treatment were observed.Results After treatment, joint pain (2.8 ± 1.1vs. 3.6 ± 1.1,t=4.129), tenderness (1.0 ± 0.5vs. 1.4 ± 0.7,t=3.297), the color change of (1.5 ± 0.8vs. 2.0 ± 0.6,t=3.117) score in the combined group were significantly lower than those in the control group (P<0.05). The degree of swelling (112.5 ± 11.8 cm3vs. 122.5 ± 13.6 cm3,t=3.816) in the combined group was significantly lower than that in control group (P<0.05). The bone mineral density (0.762 ± 0.020 g/cm2vs. 0.722 ± 0.023 g/cm2,t=3.803,P<0.05) in the combined group was significantly higher than that in the control group. The effect rate of the combined group was 90% (54/60), and the control group was 70%. There was significant difference between 2 groups (χ2=7.500,P=0.006). The effect rate of ankle joint function of the combined group was 83.3% (50/60), and the control group was 70% (42/60). There was significant difference between 2 groups (χ2=4.444,P=0.035).Conclusions The Yiqi-Wenshen decoction combined with subcutaneous injection of salmon calcitonin could relieve the pain, swelling and other symptoms of the patients with bone atrophy after calcaneal fractures, and could benefit the recovery of ankle joint function.
2.Induction of cytotoxic Tlymphocytes from the peripheral blood of a hepatocellular carcinoma patient using a MAGE-1 peptide (NYKCRFPEI) in vitro

ObjectiveTo use MAGE-1 antigen as tumor vaccine for the treatment of hepatocellular carcinoma (HCC).MethodsIrradiated peripheral blood mononuclear cells (PBMCs) pulsed with a MAGE-1 peptide (NYKCRFPEI) were used as antigen presenting cells (APC). The PBMCs from the same patient were stimulated with APCs on every 7th day for 4 times to elicit cytotoxic T lymphocytes (CTLs). The cytotoxicity of CTLs to various kinds of target cells was detected with the method of lactate dehydrogenase (LDH) releasing assay. Results The number of PBMCs increased by 32 folds during 28 days of culture. When effective cells: target cells (E∶T) was 10∶1, CTLs exhibited 62.5% cytotoxicity against autogenous lymphoblasts pulsed with the peptide of MAGE-1 antigen (NYKCRFPEI), 40.25% cytotoxicity against cells of BEL7405, a HCC cell line expressing both MAGE-1 and HLA-A24, compared with 17.88% cytolysis observed against autogenous lymphoblasts, 19.55% against cells of HLE, a HCC cell line expressing MAGE-1, negatively expressing HLA-A24. The cytotoxicity against cells of QGY7701, a HCC cell line expressing neither MAGE-1 nor HLA-A24 was much lower (1.6%). When E∶T was 3.3∶1, the cytotoxicity of CTL against peptide pulsed lymphoblasts was 53.6%, while against autogenous lymphoblasts, cells of HLE and cells of QGY7701 was much lower, 15.6%, 13% and 1% respectively. After 4h culture, most cells of BEL7405 were adhered with several CTL, few cells of QGY7701 were adhere with CTL. ConclusionsMAGE-1 peptide NYKCRFPEI in vitro successfully induced CTL with the ability of specifically killing target cells expressing both MAGE-1 and HLA-A24.

Objective To investigate the protein expressions of E-cadherin and ?-,?-catenin in breast cancers and their lymph-node metastatic tumors to evaluate their possible role in breast cancer invasion and metastasis. Methods High sensitive S-P immunohistochemical method was used to detect the protein expressions of E-cadherin and ?-,?-catenin in 60 breast cancers(31cases with lymphatic metastasis) and their lymph-node metastatic tumors. Objective Onto investigate the significance of the pleth variability index (PVI) in predicting hypotension after epidural anesthesia for cesarean delivery. Methods Sixity seven pregnant women, with ASAⅠ~Ⅱ, aged 19 ~ 34 years, were enrolled for elective cesarean delivery. SBP, BDP, MAP, HR, PI and PVI were measured at 5 min after the parturients arrived in the operation room. SBP, BDP, MAP and HR were measured every 3 min after epidural anesthesia. According to the change rate of SDP or MAP was higher than 30% or not, the parturients were divided into two groups (the hypotension group and the non-hypotension group). Results No significant differences were found in parturients'age, body height, weight, BMI, gestational weeks, SDP, DBP, MAP and HR before anesthesia between two groups. The PVI in the hypotension group was significantly higher than that in the non-hypotension group (P<0.05). A receiver operator characteristic curve analysis showed the area under curve was 0.888 when PVI was used for boundary value. When PVI over 17.35 was used as the occurrence of hypotension, the sensitivity of PVI was 0.727, and specificity was 0.895. Conclusion PVI can be used to predict the occurrence of hypotension after epidural anesthesia for cesarean delivery.
5.Evaluation of skin barrier function in children with atopic dermatitis from two communities in Shanghai

Objective To evaluate skin barrier function in children with atopic dermatitis (AD) as well as healthy children from two communities in Shanghai and to assess the relationship between skin barrier function and AD severity.Methods Totally,169 children with AD and 142 healthy children aged 3-12 years were recruited from two communities (Changning Xining community and Jiading Juyuan community) in Shanghai,China.Transepidermal water loss (TEWL) and stratum corneum hydration were measured in normal appearing nonlesional skin at four body sites (dorsal and volar forearm,cheek and anterior shin) of the patients,as well as in normal skin at the same sites of the controls.AD severity was evaluated by using the severity scoring of atopic dermatitis (SCORAD) index.Results Compared with the healthy children,the patients with AD showed higher TEWL value at all of the four body sites (all P < 0.05),but lower water content in stratum corneum at dorsal forearm and anterior shin (both P < 0.05).In patients with AD,the SCORAD index was positively correlated with mean TEWL value,but negatively correlated with the mean water content in stratum corneum.Conclusion Skin barrier function may serve as an index for evaluating the severity of AD.
6.Correlation between serum 25-hydroxy vitamin D3 concentration level and children with bronchial asthma

Objective To investigate the relationship between serum 1 ,25‐hydroxy vitamin D3 concentration level and children with bronchial asthma .Methods 222 children with bronchial asthma treated in paediatrics of Zhongshan Boai hospital from 2012 Jan 2014 Dec were chosen as asthma group(A) ,while 223 cases of healthy children were selected as control group(C) .Electro‐chemical luminescence method was used to detect the serum level ,and its effect on asthma was disscussed .Results The concen‐tration of serum 1 ,25‐hydroxy vitamin D3 of asthma group (86 .30 ± 27 .70 nmol/L) was low than the control group ( 93 .39 ± 27 . 15nmol/)L .Serum levels of asthma group in different age groups (A1 ,A2 and A3) were 106 .87 ± 28 .28 nmol/L ,88 .98 ± 20 .65 nmol/L ,68 .38 ± 19 .23 nmol/L respectively ,While those of control groups were (110 .00 ± 22 .30) nmol/L ,(87 .36 ± 24 .42) nmol/L ,(21 .23 ± 16 .42) nmol/L respectively .Male and female serum level in asthma group were (88 .26 ± 27 .73)nmol/L and (81 .94 ± 26 .91)nmol/L respectively .While those of control group were(91 .91 ± 26 .57)nmol/L and (85 .69 ± 28 .04) nmol/L re‐spectively .Conclusion There is significant correlation between serum 1 ,25‐hydroxy vitamin D3 level and children with bronchial asthma .,yet no correlation between serum level and gender .1 ,25‐hydroxy vitamin D3 duficiency is one of the risk factors of chil‐dren asthma .
7.Recent technical research hot spots and development progresses in medical whole-body positron emission tomography.

Medical whole-body positron emission tomography (PET), one of the most successful molecular imaging technologies, has been widely used in the fields of cancer diagnosis, cardiovascular disease diagnosis and cranial nerve study. But, on the other hand, the sensitivity, spatial resolution and signal-noise-ratio of the commercial medical whole-body PET systems still have some shortcomings and a great room for improvement. The sensitivity, spatial resolution and signal-noise-ratio of PET system are largely affected by the performances of the scintillators and the photo detectors. The design of a PET system is usually a trade-off in cost and performance. A better image quality can be achieved by optimizing and balancing the key components which affect the system performance the most without dramatically increases in cost. With the development of the scintillator, photo-detector and high speed electronic system, the performance of medical whole-body PET system would be dramatically improved. In this paper, we report current progresses and discuss future directions of the developments of technologies in medical whole-body PET system.

8.Establishment of delay-trace conditioning procedure of classical eyeblink conditioning in Kunming mice and analysis of behavioral characteristic parameters

ObjectiveTo investigate the characteristic parameters and variation of acoustic startle reflex (ASR) and conditioning response (CR)during the acquisition of delay-trace eyeblink conditioning.Methods15healthy Kunming mice were randomly divided into 3 groups:single-task DEBC group ( n =5 ) ; single-task TEBC group ( n =5) ; dual delay/trace group ( n =5).Three groups received paired training of tone conditioned stimulus(CS)in different audio frequency (TEBC groups:2KHz; DEBC group:1KHz) and a 100 ms comeal oxygenpuff unconditioned stimulus(US).Then observed the characteristic parameters of ASR and CR.ResultsAfter pairing training for 10 days,ASR rate( ASR% ) of single-task groups ( 10th day (38.00 ± 8.64) % and (38.55 ±12.41 ) % respectively,time effect:F =4.574,P =0.008 ; group effect:F =2.021,P =0.193 ) and dual delaytrace group ( 10th day (47.95 ± 14.23 ) % and (62.01 ± 9.03 ) % respectively,time effect:F =5,547 P =0.013 ;group effect:F =0.738,P =0.415) changed significantly with the following training but between single-task groups or dual delay-trace group.CR rate ( CR% ) of single-task groups ( 10th day were (45.4 ± 5.39) % and (65.6 ± 6.77) % respectively,time effect:F =9.558,P =0.000 ; group effect:F =5.117,P =0.054 ) and dual delay/trace group ( 10th day (57.66 ± 4.34) % and (77.35 ± 7.36) % respectively,time effect:F =7.750,P =0.002 ;group effect:F=1.449,P=0.263 ) showed obvious change with the following training but between singletask groups or dual delay-trace group.Peak amplitude of single-task DEBC group ( time effect F =2.679,P =0.017 ) and dual delay-trace group ( F=3.452,P=0.034 ) had increased significantly with the following training.Peak latency showed no obvious change.ConclusionDual delay-trace conditioning procedure of classical eyeblink conditioning and single-task can be successfully established in Kunming mice.Although there are some variations,those can be controled and be weaken effect of them by taking targeted measures.
9.Effects of glycyrrhizin on the expressions of glucocorticoid receptor and NF-κB in lung of rat with acute lung injury

Objective To investigate the modulatory effects of glycyrrhizin (GL) on the expressions of nu-clear factor kappa B (NF-κB), glucocorticoid receptor (GR) and cytokines in serum, and to explore the protective mechanism of GL in acute lung injury (ALI) induced by lipopolysaecharide (LPS). Method Twenty-four male Sprague-Dawley rats were randomly(random number) divided into three groups (n = 8 in each) : normal group, ALI group and GL treatment group. Rats in the ALI group and GL treatment group were administered with LPS (5 mg/kg) intravenously. In GL treatment group, rats were treated with GL (20 mg/kg) one hour before LPS injec-tion. The animals were sacrificed 4 hours after injection of LPS, and then the lung wewt/dry ratio and PaO_2 were measured, the histopathology of lung injury was observed under light microscope, the expressions of NF-κB mRNA and GR mRNA in lung tissues were detected by using RT-PCR, and the levels of NF-κB protein and GR protein were determined by using Western blot. The levels of TNF-α and IL-10 in serum were observed by using ELISA.Data were analyzed with SPSS version 13.0 software, and means were compared with analysis of variance and Stu-dent-Newman-Keuls test. Results (1) TNF-α levels in normal group, ALI group and GL treatment group were (43.96±7.57), (153.68±20.42), and (87.23±7.52) ng/L, respectively, and IL-10 levels were (24.72±8.03), (42.48±6.81) and (58.33±9.62) ng/L, respectively (F = 183.70, all P <0.01). (2) NF-κB mRNA expressions in normal group, ALI group and GL treatment group were (0.432±0.085), (3.414±0.521) and (1.894±0.272), respectively, and NF-κB protein levels were (45.6±7.3), (254.7±16.4)and (133.5 ±11.7) ng/L, respectively, and comparison between groups showed statistical significant (F = 187.82 and 1466.53, ALL P < 0.01). GR mRNA expressions in normal group, ALI group and GL treatment group were (0.434±0.013), (0.152±0.025) and (0.308±0.033), respectively, and GR protein levels were(54.6±6.5), (11.5±2.3)and (28.2±5.6) ng/L, respectively (F = 246.00 and 260.92, all P < 0.01). (3) Com-pared with normal group, infiltration of PMNs, capillary congestion and swelling were found in ALI group, and treatment with GL could attenuate the lung injury. Conclusions Glycyrrhizin has a protective effects on rats with ALI induced by LPS maybe through down-regulating the expressions of NF-κB mRNA and TNF-α mRNA, and up-regulating the expression of GR mRNA and level of IL-10 protein.
10.Significance of expression of thrombospondin-1 and receptor-CD36 in hepatocellular carcinoma

AIM: To study the expression of thrombospondin-1 (TSP-1) and receptor-CD36, and investigate the relationship between tumor invasive capability and microvessel density and thrombospondin-1. METHODS: 43 hepatocellular carcinoma (HCC) cases were under investigation. Tissues from tumor, corresponding adjacent non-HCC tissue were stained with CD34 to show the MVD. TSP-1 and CD36 were examined by immunohistochemistry (SP) and RT-PCR. Relationship between clinical pathological features and above parameters was analyed. RESULTS: The staining of TSP-1 in HCC tissue is significantly lower than that in corresponding adjacent non-HCC tissue. Expression of TSP-1 was correlated to tumor thrombi, capsule, tumor invasive capability and CD36. CD36 was also correlated to tumor thrombi and tumor invasive capability. MVD was significantly higher in TSP-1, CD36 positive group than that in negative group. CONCLUSION: TSP-1 inhibits the growth, invasion and angiogenesis in HCC. TSP-1 may take effect through CD36.
