1.Prevalence of white coat hypertension and its effect on left ventricular hypertrophy

Objectives To investigate the frequency of white coat hypertension (WCH) in all patients with hypertension (HT) and its effect on left ventricular hypertrophy (LVH).Methods A total of 224 patients (with an average age of 51 years, 155 males) with seated diastolic blood pressure of (95~114) mm Hg in clinic were recruited into this study. Their blood pressure was monitored dynamically for 24 h with non-invasive ambulatory method at the same day, and M-mode bi-dimensional echocardiography, electrocardiograph, determinations of serum levels of lipid and glucose were also performed for them. 2.Bone marrow mesenchymal stem cells differentiate into neuron-like cells induced by combination of two cytokines

BACKGROUND:Bone marrow mesenchymal stem cells (BMSCs) can be induced to differentiate into neuron-like cells directional y. Accordingly, BMSCs can be used as seed cells theoretical y in constructing tissue-engineered peripheral nerves.
OBJECTIVE:Using combination of two cytokines to induce BMSCs differentiating into neuron-like cells directional y, and further to discusse its application in peripheral nerve injury.
METHODS:BMSCs were isolated and purificated from the bone marrow of Wistar rats by using the differential adherence method. Basic fibroblast growth factor and epidermal growth factor were used to induce the BMSCs differentiating into neuron-like cells. The morphological change was observed and the neuronal specific markers were detected by immunohistochemistry technique. The morphological and immunohistological changes were also studied after the induce agent were removed.
RESULTS AND CONCLUSION:With presence of morphological and immunohistochemical features of nerve cells induced by neurotrophic factors, BMSCs exhibited two or more processes that were interconnected as a meshwork;cellnucleus and nucleus could be observed with strong light refraction of cytoplasm. After immunohistochemical staining, neuroln specific enolase, neurofilament protein and synaptophysin protein positive cells were detected. A great amount of cells reversed to their original fibroblast-like morphology, and the expression of the three above-mentioned proteins decreased as the induce agent withdrawn. Our study showed that BMSCs can be induced to differentiate into neuron-like cells, but the transdifferentiation is a short-time reversible phenomenon.
3.Values of D-dimer,cTnT and myocardial enzymogram detections in the diagnosis of acute myocardial infarction

Objective To investigate the value of D‐dimer (D‐D) ,cardiac troponin T(cTnT) and myocardial enzymogram detec‐tions in clinical diagnosis of acute myocardial infarction (AMI) .Methods 36 cases of AMI patients (AMI group) and 20 cases of healthy people (control group) were selected as study subjects .The plasma levels of D‐D ,cTnT and myocardial enzymogram of two groups were detected ,and the sensibility and specificity of these indexes were analyzed .Results The levels of D‐D ,cTnT ,CK ,CK‐MB ,AST and LDH in AMI group were significantly higher than those of control group (P<0 .05) .The positive rates of cTnT ,D‐D and myocardial enzymogram in AMI patients were 91 .67% ,69 .44% ,and 66 .67% ,respectively .The positive rate of cTnT was significantly higher than those of D‐D and myocardial enzymogram (P<0 .05) .Both of the diagnosis sensibility and specificity of plasma cTnT were significantly higher than those of D‐D and myocardial enzymogram (P<0 .05) .Conclusion Comparing with D‐D and myocardial enzymogram ,plasma cTnT has better diagnosis sensibility and specificity .Joint detection of kinds of indexes could improve the accuracy .
4.Expresson of the N-terminus truncated phosphotase D in Escherichia coli and characterization of its anti inflammatory activity

To investigate the immunological activities of the recombinant human phosphatase D2 (rhPLD2) in vitro and in vivo, especially its ability to reduce inflammatory reactions, the cDNA fragment encoding rhPLD2 was cloned into prokaryotic expression vector pET30a by RT-PCR and the recombinant protein rhPLD2 expressed in E.coli was purified from the inclusion bodies, while the anti inflammatory activity of rhPLD2 was determined by the amount of eosinophils in bronchoalveolar fluid(BALF) and blood and the expression of IL-5 and MMP-9 in lung tissues of guinea pig model of chronic asthma. It was found that the rhPLD2 recombinant protein was obtained from human Daudi cells by cloning to E.coli, which contained no membrane-binding site and signal peptide. The cDNA sequence encoded 631 amino acid residues (GenBank Accession Number: AY178289). The purity of the rhPLD2 approached up to 76% with a bioactivity of 50.9745 units/L (0.9212 g/L). In addition, the anti inflammatory effect of rhPLD2 protein could be demonstrated in the guinea pig model of chronic asthma after treatment with rhPLD2 protein, such as down regulation in the expression of the inflammatory cytokine IL-5. It is concluded that the anti-inflammator activity of the recombinant human truncated PLD2 protein produced from the E.coli plasmid can be demonstrated both in vitro and in vivo.
5.Apoptosis and the changes of Na+ and K+ contents in retina of rats with diabetes mellitus

BACKGROUND: Diabetic retinopathy is a commonly chronic-vascular complication in a course of diabetes mellitus (DM), and its relation with apoptosis and changes of ion level needs to be proved.OBJECTIVE: To observe apoptosis and changes of Na+ and K+ contents in retinal tissue of DM rats.DESIGN: Randomized controlled study.SETTING: Department of Ophthalmology, Liuzhou Municipal People's Hospital.MATERIALS: The experiment was completed at the Department of Ophthalmology, Liuzhou Municipal People's Hospital. A total of forty adult male Wistar rats, aged 20 months, weighing 320-350 g, were provided by Animal Center of Medical College Affiliated to Sun Yat-sen University.METHODS: Forth Wistar rats were randomly divided into control group and experimental group with 20 in each group. Streptozotocin was used to induce DM in the experimental group. Apoptosis and changes of Na+ and K+ contents in retinal tissue were measured on the 4th, 6th, 12th and 16th weeks after DM onset. Rats in the control group were injected only with the same volume of citrate buffer solution. Then, correlations on the aspect of fasting blood glucose (FBG), hemoglobin glycosylation (HbAlc), Na+ and K+ contents and DM course were analyzed between the two groups.MAIN OUTCOME MEASURES: ① Na+ and K+ contents; ② FBG concentra tion and HbAlc level; ③ changes of apoptosis; ④ correlations among markers.RESULTS: Apoptosis could be detected in retinal tissue in the experimental group at 4 weeks after DM onset, and with course elongating, level of apoptosis was aggravated gradually. Na+ concentration was increased in retinal tissue, but K+ concentration was decreased (P < 0.01 or 0.05). Levels of apoptosis in retinal tissue in the experimental group were positive correlation with FBG concentration, HbAlc level, Na+ content and DM course (r=7.584, 7.844, 7.369, 6.246; P < 0.01); however, they were negative correlation with K+ content in retinal tissue (r=7.658, P < 0.01).CONCLUSION: There are apoptosis and abnormal Na+ and K+ contents in retinal tissue of DM rats. Moreover, these changes may be one of pathological bases of diabetic retinopathy.
6.Radiosensitization of gemcitabine in human cholangiocarcinoma cell line

Objective To evaluate the radiosensitization effect of gemcitabine in human cholangiocarcinoma cells(QBC939)in vitro. Methods The IC10,IC50 and IC90 of gemcitabine in QBC939 cells were determined by clonogenic assay,which were adopted in the following experiments.The cells were divided into five groups:control group,chemotherapy group,radiation group,radiation after chemotherapy(C+R)group and radiation before chemotherapy(R+C)group.The radiation dose ranged from 1 Gy to 10 Gy.The cell survival Curves were fit using two-component equation and the sensitive enhancement ratio(SER)was calculated.Results The IC10,IC50 and IC90 of gemcitabine in QBC939 cells were 0.1 nmol/L(low concentration),11.0 nmol/L(moderate concentration)and 21.5 nmol/L(high concertration),resDectively.Gemcitabine in low concentration radiosensitized the cells at low irradiation dose(≤2 Gy)in R +C group(SERDq=1.52).Gemcitabine in moderate concentration had radiosensitization effect at all irradiation doses in C+R group(SERD0=1.27,SERDq=116.93),and at low irradiation dose(≤2 Gy)in R+C group.Gemcitabine in high concentration had radiosensitization effect at all irradiation doses in both R+C and C+R groups,which was most markedly at low irradiation dose(≤2 Gy)in R+C group(SERDq =323.30). Conclusions Gemeitabine has radiosensitization effect in QBC939 cells,which is based on the optimal scheduling and concentration.
7.Theoretical research and progress of basal body and bio-coatings in biomaterial field

BACKGROUND: In the research of biomaterial, especially bio-coatings field, how to find a good biological and mechanical properties basal body, and to prepare coatings with uniform and compact surface and strong bonding force, is the key factor in clinical Application of artificial prosthesis. OBJECTIVE: To review the progress of bio-coatings.METHODS: Databases of CNKI, VIP, and Science Direct were retrieved with key words of "Biocoatings, HAp, Titanium, Carbon/carbon composites" to search papers published from January 1998 to October 2009. PApers underlying bio-coatings were selected. Totally 216 literatures were initial searched, according to inclusive criteria, 31 literature were summarized. RESULTS AND CONCLUSION: By contrasting the basal bodies made of titanium or carbon/carbon and different coating preparation, we found that the basal body composed of carbon/carbon composites exhibited good biocompatibility, which had similar strength to natural bone. Using hydrothermal method, the composites coating prepared by taking carbon/carbon as basal body and hydroxyApatite, chitosan, or polymethyl methacrylate as coatings provided a new idea for increasing the combing strength between coatings and basal body.
8.Protective effect of mycophenolate mofetil to renal tissures in diabetic rats

Objective To assess the renal protective effects of mycophenolate mofetil(MMF) in diabetic model rats and to explore its mechanism. Methods SD rats were divided randomly into there groups: control group,diabetic model group and diabetic group treated with MMF. In 12 weeks, blood glucose(BG), blood urea nitrogen(BUN), serum creatinine(Scr), index number of kidney hypertrophy(kidey weight to body weight, KW/BW),creatinine clearance(Ccr) and 24- hour urinary protein (24Upro) were detected. Kidney tissure were examined by microscopy. The protein exprssion of intercellular adhesion molecule-1 (ICAM-1) and proliferating cell nuclear antigen(PCNA) in renal tissure were determined by immunohistochemical technique. The mRNA expression of ICAM-1 in kidney tissure were semi-quantitatively determined with reverse transcription-polymerase chain reaction(RT-PCR). Objective To detect the change of discharge phase of guinea pig hippocampal CA1 pyramidal cells during visual discriminative task with an effective and convenient program we designed. Methods Five guinea pigs were performed by extracellular single unit recording in vivo when they were performing visual discriminative task. Discharge signals of individual pyramidal cells were extracted from different frequency signals by wavelet transform (WT), which made it feasible to calculate discharge phase of pyramidal cells in terms of time correlation between discharge and ? rhythm. Results The discharge phase of CA1 pyramidal cells in the 1 to 5s interval before visual discriminative task (172??1.8?) was obviously earlier than that in the 6 to 10s interval after visual discriminative task (189??3.7?) ( P0.01). Conclusion The program we designed is capable of detecting discharge phase of pyramidal cells. Regular shift of discharge phase of hippocampal CA1 pyramidal cells emerges before and after performing visual discriminative task.
10.Effect of CTLA-4Ig on BAEP and SEP in rats of experimental autoimmune encephalomyelitis

Objective To explore the curative effect and safety of CTLA-4Ig transgene for experimental autoimmune encephalomyelitis (EAE). Methods The adenovirus loaded CTLA-4Ig was injected into Wistar rat EAE models through the lateral cerebral ventricle. The clinical symptomatic and electrophysiological changes were observed to judge the efficiency of CTLA-4Ig. Results After injection of AdCTLA-4Ig into the lateral cerebral ventricle of Wistar rat EAE models, the time of EAE onset was delayed and the incidence rate was significantly reduced. And the transductive function tested by brain-stem auditory evoked potential (BAEP) and somatosensory evoked potential (SEP) was obviously improved. Conclusion CTLA-4Ig is effective in managing EAE.
