2.Eph receptor tyrosine kinase and its relation to cancer

Eph,the largest subfamily of tyrosine kinase receptor, could be divided into two groups,EphA and EphB.Ephs play a role in the embryonic development and differentiation of the nervous and vascular systems. In recent years,a large number of ephrins and Ephs had been found to be overexpressed in numerous human tumors,and have relation to development and prognosis of cancers.Down-regulated of EphA3 and EphA7 in cancers,was attributed to the hypermethylaiton of CpG island in promoter region.
3.Down-regulation of EphA7 by aberrant methylation in colorectal cancer

Objective:Ephs have been found to be over-expressed in numerous human tumors,and relate to the development and prognosis of cancers.The expression of EphA7 in colon cancer cell lines and primary colorectal cancers was detected.The role of EphA7 in the development of colorectal cancer was investigated. Methods:Quantitative reverse transcription-polymerase chain reaction was used to detect the expression of EphA7.Methylation specific PCR and bisulfite sequencing were used to examine the methylation status of EphA7.pEGFP-EphA7 plasmid was constructed and was transfected into HCT116 which lost the expression of EphA7.Gene chip was analyzed for the expression of genes between transfectant and mock cell. Results:Down-regulation of EphA7 was observed in cell lines of primary colorectal cancers,and the evidence of aberrant methylation was found in CpG island of EphA7 promoter.The methylation status of EphA7 gene was related to sex,differentiation and location of colorectal cancers.Some genes were up or down regulated in pEGFP-EphA7 transfectant. Conclusion: EphA7 gene may be involved in the carcinogenesis of colorectal cancer.The hypermethylation of CpG island is an important mechanism leading to the down regulation of EphA7 in colorectal cancer.
4.Rare type of hepatic benign tumor and tumor-like lesions

Objective To summarize the experience in diagnosis and treatment of rare type of hepatic benign tumor and tumor-like lesions.Methods The clinical data of 58 cases of uncommon benign hepatic tumor and tumor-like lesions admitted to Changhai Hospital from January 1980 to June 2000 were retrospectively reviewed. Results Of the 58 cases, 26 were inflammatory pseudotumors, 8 focal hyperplastic lesions, 7 hepatocellular adenomas, 7 lipomatous tumors, 5 cystadenomas, 3 tuberculosis, 1 harmatoma and 1 neurofibroma. 82.8% of the cases had symptoms, 17.2% without obvious symptoms. Single lesion was found in 93.1%, multiple lesions in 6.9%. Operative procedures as follows: 30 local resections of the lesion, 14 hepatic segmentectomies, 9 hepatic lobectomies, 2 hepatic artery ligations, 1 hepatic artery ligation plus drainage of cyst, 2 liver biopsies. No operative mortality and no serious morbidity were noted. Conclusions With popularization of advanced imaging techniques, the incidences of the benign tumor and tumor-like lesions of the liver have been increased in recent years. However, current imaging studies lack specificity, and differential dignosis of these lesions remains difficult. So we should be familiar with the clinical behavior, presentation of imagings and management of the diseases mentioned above.
5.Influence of different blood serum concentration and growth time on olfactory ensheathing cells from the olfactory mucosa of adult rats

BACKGROUND:Preliminary experiments have reported the influence of serum and nerve growth factor on olfactory ensheathing cells proliferation in vitro, but there are less studies concerning choice of serum concentration and growth time for in vitro culture of olfactory ensheathing cells.
OBJECTIVE:To find out the influence of different blood serum concentration and growth time on olfactory ensheathing cells from the olfactory mucosa of adult rats based on the growth curve of olfactory ensheathing cells.
METHODS:The olfactory ensheathing cells from the olfactory mucosa of adult rats were separated, culture and identified in vitro. Sulforhodamine B and microplate reader were employed to measure absorbance values and plot growth curve of olfactory ensheathing cells.
RESULTS AND CONCLUSION:When cultured for the same time in blood serum of different concentrations, absorbance values, especial y in the groups 10%, 20%, 30%, 40%, tended to increase with time except the 0%group. When cultured in the same serum for different time, absorbance values increased within the first 9 days, then promoted rapidly in the groups 10%, 20%, 30%, 40%at 13 days, entered the plateau phase at 19 days, and decreased at 23 days;meanwhile, in the other groups (50%, 60%, 70%, 80%, 90%) the absorbance values peaked at the 13th day and then decreased gradual y. These findings indicate that different serum concentrations and different growth time in vitro affect cellgrowth and survival of olfactory ensheathing cells significantly, which should be ful y considered when cells are cultured in an in vitro condition.
6.Construction of a lentiviral expression vector carrying EphB4 and its expression in colon cancer cells

Objective Studies show that the abnormal expression of EphB4 plays an important role in the development and progression of colon cancer .The present study aims to provide some experimental evidence for the gene therapy of colon cancer by con -structing a lentiviral expression vector carrying the homo EphB4 gene and further establishing colon cancer cell lines with stable overex-pression of EphB4. Methods A series of oligonucleotides (oligo) encoding the homo EphB4 gene were ligated together by PCR and then cloned into a lentiviral expression vector pLenti 6.3-MCS-IRES2-EGFP.After confirmed by sequencing , the vector pLenti6.3-EphB4-IRES2-EGFP and its helper vectors were mixed and co-transfected into 293 T cells to obtain recombinant virus containing the EphB4 gene.The lentiviral titer was detected and the resulting recombinant lentiviruses carrying EphB4 or control viruses only carrying green fluorescence protein (GFP) were used to infect the human colon cancer cell lines .The expression of GFP was determined under the inverted fluorescence microscope and the level of EphB 4 mRNA in the infected cells detected by qPCR . Results The lentiviral expression vector pLenti6.3-EphB4-IRES2-EGFP carrying correct homo EphB4 gene sequence was successfully constructed .The titer of the recombinant EphB4 lentiviral supernatant Lenti6.3-EphB4 was 1 ×108 TU/mL.The expression of GFP was observed in the trans-duced cells under the fluorescence microscope , and that of EphB4 mRNA in the transfected SW480 and Coca-2 cells was significantly up-regulated as compared with the control and blank groups . Conclusion The homo EphB4 gene was successfully amplified and cloned.A lentiviral expression vector was successfully constructed , and so were colon cancer cell lines stably overexpressing EphB 4, which may shed light on the lentivirus-mediated genetic therapy for colon cancer .
7.Different prognosis in patients of peritoneal dialysis with high peritoneal solute transport

To investigate association between prognosis of patients of peritoneal dialysis with their initial high peritoneal solute transport and complications. Two hundred and two patients with end-stage renal disease who began peritoneal dialysis during September 1,2006 to September 1, 2008 at Peking University Third Hospital, Beijing were recruited in the study. They were undergone peritoneal equilibration test within six weeks after initiating peritoneal dialysis, and their types of peritoneal solute transport, complications,nutrition status and outcomes were all recorded until August 31,2010. Their survival rate was estimated by Kaplan-Meier method. Compare to those with other types of solute transport, patients with high peritoneal solute transport showed higher proportion of complications ( P < 0. 05 ) during dialysis, but less ultrafiltration function ( P < 0. 05 ), worse nutrition status ( P < 0. 05 ) and lower cumulative survival rate ( P < 0. 01 ).Among patients with high peritoneal solute transport, cumulative survival rate in those with complication was lower than those without it ( P = 0. 031 ). Prognosis of patients with high peritoneal solute transport possibly varies with their complications in dialysis.
8.Anti-infection value of combined determination of serum levels of procalcitonin, C-reactive protein,prealbumin, white blook cell in the elderly with bacterial infection

Objective To evaluate the clinical significance of the combined determination of serum procalcitonin (PCT),C reactive protein(CRP),prealbumin(PA)and white blood cell(WBC) in curative effect and prognosis of elderly patients with bacterial infection.Methods Serum PCT,CRP,PA and WBC levels and their dynamic changes of 70 patients with bacterial infection from intensive care unit(ICU)were measured.The patients with bacterial infection were classified into serious infection group and local infection group.And the comparison with virus infection group was made.Results The levels of PCT(18.00±3.20)μg/L and CRP(86.17±10.64)mg/L in serious infection group and local infection group[PCT(2.03±0.46)μg/L,CRP(50.24±7.45)mg/L] were increased.The contents of PA were reduced[(3.214±0.88)mg/L,(12.89± 1.03)mg/L].The levels of WBC were increased mildly[(8.89±1.23) × 109/L,(6.54±0.87) × 109/L].The differences were statistically significant between bacterial infection groups(serious infection group and local infection group) and virus infection group [PCT (0.21 ±0.06)μg/L,CRP (5.21 ±0.84) mg/L,PA (20.14 ±2.57)mg/L,WBC (5.30 ± 0.93) × 109/L] (P< 0.01).Serum PCT,CRP.PA and WBC showed significant differences between serious infection group and local infection group(P<0.01).After antibiotic treatment,the levels of PCT (0.5 ± 0.08) μg/L,CRP (10.32 ± 1.65) mg/L and WBC (6.30± 0.91) × 109/L in bacterial infection group were reduced obviously and the concentration of PA (18.19±2.66)mg/L was elevated compared with before treatment [PCT(11.61±8.27)μg/L,CRP (71.80±20.09)mg/L,WBC(7.95± 1.59) × 109/L,PA(7.08±4.87)mg/L] (P<0.01).Correlative analysis was performed between PCT and others indicators before and after antibiotic treatment in bacterial infection group and virus infection group.There was positive correlation between PCT and CRP,WBC before(r=0.586,0.445) and after treatment (r=0.688,0.463) in bacterial infection group.PCT and WBC(r=0.432) were positively correlated after treatment,while no correlation existed between PCT and others indexes in virus infection group(r<0.306).When comparing sensitivity and specificity of PCT and CRP for diagnosis of bacterial infection disease,the sensitivity (95.7%) of PCT was similar to that of CRP.The specificity of PCT(86.7%) was remarkably higher than that of CRP(73.3%).Conclusions The combination of serum PCT,CRP.PA and WBC is valuable for early diagnosis of bacterial infection disease.Monitoring PCT level dynamically would assist an evaluation of curative effect and disease outcome of elderly patients.
9.Development of Puncture-drainage System for Intracranial Hematoma

Objective To introduce a puncture-drainage system for intracranial hematoma, which can be used in both peacetime and wartime. Methods The inserting-style connections were taken to design the catheter needle and driller. In the design of the combined T-piece, silica membrane was used to be the main sealing end. The fixed division between the main sealing end and the catheter needle took thread connections. The angle between the side-tube of the T-piece and the catheter needle was 120 degree; the side-tube of the T-piece and the Y-tube also took thread connections. The another two sides of Y-tube linked to different drainage bag and syringe, and all of the hoses got the individual and portable switches. Results This system was more efficient, rapid, safe and convenient. What's more, the injury was minimal when doctor performed an intracranial hematoma puncture surgery by using the puncture-drainage system. Conclusion It is very easy to operate and suitable to puncture drainage for intracranial hematoma surgery in both peacetime and wartime.
