1.Glutamate and early neurological deterioration in ischemic stroke
International Journal of Cerebrovascular Diseases 2013;(2):132-137
The mechanisms underlying early neurological deterioration (END) after ischemic stroke are not clear. There are no reliable predictive factors and effective preventive measures for END.The glutamatemediated excitotoxicity plays a very important role in the cascade reaction of ischemic events.High level of glutamate in plasma is one of the important predictors for END.Studies have shown that the polymorphism in the promoter of the excitatory amino acid transporter-2 gene is a potential cause for individual susceptibility to END.Some therapeutic strategies of interrupting the glutamatergic pathways may be as the strategies of intervention END.

With the development of medical sciences, the concept of prevention and treatment of ischemic stroke is being updated continuously. However, there are still some gaps between the available evidence and clinical practice in its strategies of prevention and treatment. The article performs evidence-based analysis for the available evidence in the prevention and treatment of ischemic stroke in accordance with the scientific levels of evidence classification so as to narrow the gaps between the evidence in this field and clinical practice.

Objective:To establish experimental periodontitis model in Wistar rats by prednisolone acetate injection and ligature of their left second maxillary molars with steel-wire. Methods:Twenty Wistar rats were randomly divided into two groups, control group and experimental group. Muscle injection with saline was regarded as the control group. Prednisolone injection plus ligature of left second maxillary molar with steel-wire was used as the experimental group. The rats were killed after 6 and 8 weeks. Results:The rats in experimental group showed bad appetite and did not want to move after 4 days of injection. These symptoms were agreement with "shen-xu" in traditional Chinese medicine. The osteoporotic changes in alveolar bone, the absorption of alveolar ridge, the formation of periodontal pocket and osteoclastic activity were found in treatment group. None of above pathological changes occurred in periodontal tissue of the control group. Conclusion: Experimental periodontitis model could be successfully established in Wistar rats by ligature with steel-wire plus prednisolone injection, and this animal model offers a good means for the study of periodontitis.

Objective To investigate the correlationship between proliferating cell nuclear antigen (PCNA), intercapillary distance (ICD) and radiosensitivity in uterine cervical cancer. Methods 112 patients who received radiotherapy alone were followed over five years. All PCNA expressions and ICD values were analyzed with immunohistochemistry assay and computer defined capillary images. Results PCNA expression was observed in all patients with the average index of 0.542 ranging from 0.241 to 0.903 um. ICD averaged 180.42 um (ranging from 58.47 to 243.62 um). Conclusions The PCNA expression and ICD value, being closely correlated with radiosensitivity and response to therapy, can be useful as markers for prediction and selection of treatment decision for uterine cervical cancer.

Objective To discuss the relation between level of Hs-CRP and glycometabolic disturbance in patients with acute coronary syndrom. Methods 155 patients with acute coronary syndrome were divided into 3 groups according to the WHO standard of diabetes diagnosis, normal glucose tolerance group(group A,44 cases),impaired glucose tolerance group(group B,60 cases),diabetes group(group C,51 cases). All cases were checked by the 75g glucose tolerance test, blood Glucose, insulin, HbA1c,lipid,hypersensitive C reaction protein(hs-CRP). 95 patients were checked by coronary angiography(CAG). Statistic comparison were measured among three groups including the levels of hs-CRP,insulin,HbA1c,lipid and coronary artery lesions. Results The levels of Hs-CRP,PBG, FINS, PINS,HbA1c,TC,LDL-C in both group B and C were higher than those in group A;The levels of TG,PINS were not found significant differences between group B and C. The level of Hs-CRP was correlated with the levels of PBG,PINS,HbA1c,TC,LDL-C. The coronary artery lesions showed by CAG in group B and C were more severe than those in group A,coronary multivessel changes in group B and C were more frequent than in group A,it had no significant difference between group B and C.Conclusion Compared with the patients of normal glucose tolerance, patients with glycometabolic disturbance have higher levels of hs-CRP,their coronary artery lesions are more severe.

BACKGROUND:Preliminary experiments have reported the influence of serum and nerve growth factor on olfactory ensheathing cells proliferation in vitro, but there are less studies concerning choice of serum concentration and growth time for in vitro culture of olfactory ensheathing cells.
OBJECTIVE:To find out the influence of different blood serum concentration and growth time on olfactory ensheathing cells from the olfactory mucosa of adult rats based on the growth curve of olfactory ensheathing cells.
METHODS:The olfactory ensheathing cells from the olfactory mucosa of adult rats were separated, culture and identified in vitro. Sulforhodamine B and microplate reader were employed to measure absorbance values and plot growth curve of olfactory ensheathing cells.
RESULTS AND CONCLUSION:When cultured for the same time in blood serum of different concentrations, absorbance values, especial y in the groups 10%, 20%, 30%, 40%, tended to increase with time except the 0%group. When cultured in the same serum for different time, absorbance values increased within the first 9 days, then promoted rapidly in the groups 10%, 20%, 30%, 40%at 13 days, entered the plateau phase at 19 days, and decreased at 23 days;meanwhile, in the other groups (50%, 60%, 70%, 80%, 90%) the absorbance values peaked at the 13th day and then decreased gradual y. These findings indicate that different serum concentrations and different growth time in vitro affect cellgrowth and survival of olfactory ensheathing cells significantly, which should be ful y considered when cells are cultured in an in vitro condition.

Objective To study the effects about rhizoma drynariae poly (lactic-co-glycolic acid) (DR-PLGA) /Calcium phosphate cement (CPC) composite scaffold on treating rabbit femoral bone defect. Methods Eight Newzealand rabbits were randomly divided into experiment group(n =4) and control group (n =4).The preparation of rabbit femoral bone defect model, separately implanted DR-PLGA/CPC scaffold and PLGA/CPC scaffold. After 4,8 weeks,we took out the materials, observed with X-ray,gross anatomy, histology observation to evaluate the osteogenetic activity, the effect of accelerating the healing of bone defect.Resuits At the 4th week and 8th week after implantation, the effect of promoting fracture healing and osteogenic activity of the experiment group were greater than those in the control group. Conclusions DR-PLGA combined with CPC could induce new bone formation, promote the healing of rabbit femoral defect.
