1.Effect of Astragalus membranaceus on antioxidant capacity and liver and kidney function in patients with chronic schizophrenia and metabolic syndrome

Objective To investigate the effect of Astragalus membranaceus on the antioxidant capacity and liver and kidney function in patients with chronic schizophrenia and metabolic syndrome.Methods 110 cases of patients with chronic schizophrenia and metabolic syndrome were selected as subjects.They were randomly divided into the control group and the observation group by random number table method.Both groups were treated with routine antipsychotic drugs, and the observation group were additionally treated with Astragalus injection, once per day.10~14 d as a course of treatment, they were treated for a total of 6 courses.The antioxidant capacity and the liver and kidney function were compared between the two groups.The mental symptoms was evaluated with the positive and negative symptoms scale(PANSS).Results After treatment, the antioxidant capacity of the two groups was significantly increased while MDA was significantly decreased, and the changes were more obvious in observation group than the control group.After treatment, indexes of liver and kidney function were significantly increased.Except BUN and SUA, the other indicators were better in the observation group than the control group(P<0.05).After treatment, PANSS score (61.4±7.8)in the control group was significantly higher than that of the observation group(54.5±8.2)(P<0.05).Conclusion Astragalus membranaceus can be used as an assisted drug for treating chronic schizophrenia and metabolic syndrome.It can effectively improve the antioxidant capacity, reduce the damage to liver and kidney function and improve symptoms of mental illness, so as to improve the quality of life.
2.Research on the effect of morphine hydrochloride sustained-release tablets given by different administration routes on patients with cancer pain

Objective:To analyze the effect of morphine hydrochloride sustained-release tablets given by different administration routes on patients with cancer pain.Methods: Chosen patients with cancer pain as research subjects, underwent morphine hydrochloride sustained-release tablets for treatment of pain, randomized to receive oral administration as control group and rectally administration as observation group, observed pain relief degree, onset time of analgesia and adverse reaction rates.Results:1)After treatment, the observation group patients’ overall response rate was 94.05%,no significant difference compared with the control group; 2) After treatment, the observation group patients had a mean onset time (0.68 ±0.17) h, significantly shorter than the control group,0.5h, 1h, 2h, 4h of onset percentages of analgesia were higher than control group; 3)After treatment, the observation group patients’ incidence of adverse reactions was 2.38%, significantly lower than the control group.Conclusion: The morphine hydrochloride sustained release tablets rectal administration route can significantly alleviate cancer pain, shorten onset time of analgesia, and don’t increase the incidence of adverse reactions.
3.The case control study on the relation between level of Hs-CRP and glycometabolic disturbance in patients with acute coronary syndrome

Objective To discuss the relation between level of Hs-CRP and glycometabolic disturbance in patients with acute coronary syndrom. Methods 155 patients with acute coronary syndrome were divided into 3 groups according to the WHO standard of diabetes diagnosis,normal glucose tolerance group(group A,44 cases),impaired glucose tolerance group(group B,60 cases),diabetes group(group C,51 cases). All cases were checked by the 75g glucose tolerance test, blood Glucose, insulin, HbA1c,lipid,hypersensitive C reaction protein(hs-CRP). 95 patients were checked by coronary angiography(CAG). Statistic comparison were measured among three groups including the levels of hs-CRP,insulin,HbA1c,lipid and coronary artery lesions. Results The levels of Hs-CRP,PBG, FINS, PINS,HbA1c,TC,LDL-C in both group B and C were higher than those in group A;The levels of TG,PINS were not found significant differences between group B and C. The level of Hs-CRP was correlated with the levels of PBG,PINS,HbA1c,TC,LDL-C. The coronary artery lesions showed by CAG in group B and C were more severe than those in group A,coronary multivessel changes in group B and C were more frequent than in group A,it had no significant difference between group B and C.Conclusion Compared with the patients of normal glucose tolerance, patients with glycometabolic disturbance have higher levels of hs-CRP,their coronary artery lesions are more severe.
4.Pharmacokinetic studies on curcumin in Curcuma phaeocaulis in rats in vivo

Objective To study pharmacokinetics of curcumin in Curcuma phaeocaulis in rats in vivo.Methods HPLC method was used to determine the curcumin in rat plasma.The conditions were column: Lichrospher-5-C_(18)(250 mm?4.6 mm, 5 ?m); column temperature: 25 ℃;mobile phase: CH_3CN-5% HAc water solution(45∶55);flow: 1 mL/min;detection wavelength: 420 nm.Results The calibration curve was liner(r=0.999 5) at the range of 6.5—104 ?g/mL.The average recovery was 98.5%.RSD was 2.41%(n=5).The pharmacokinetic parameters of curcumin were as follows: k_a was 0.53/h,k_e was 0.10/h,t_(1/2ka) was 1.32 h,t_(1/2k) was 6.89 h,t_(peak) was 3.89 h,C_(max) was 93.15 ng/mL,AUC was(1 369.38) ng/mL.Conclusion This method is stable,simple,and reliable,which can be applied for the determination of curcumin in plasma and pharmacokinetic studies.
5.Study of anti-diabetic nephropathy efficacy of berberine analogue Y53 in STZ-induced diabetic C57 BL/6 J mice

Aim To investigate the ameliorative effects pseudoberberine(Y53), a berberine(BBR) analogue, on diabetic nephropathy( DN) in streptozotocin( STZ)-induced diabetic mice. Methods Diabetes mellitus ( DM) of the C57BL/6J mice was induced by intraper-itoneal injection of STZ at 120 mg·kg-1 . The diabetic animals were divided into 4 groups, which were orally treated with saline, 50 mg · kg-1 of BBR, 50 mg · kg-1 of Y53 or 5 mg · kg-1 of rosiglitazone ( ROSI ) , respectively. During and after the experiment, the u-rine, blood, serum and kidney of the animals were harvested for determination of relevant parameters by kits. Kidney tissues of the mice were subjected to pathological examination by hematoxylin & eosin( HE) staining;mRNA and protein expression levels of target genes in the kidney were determined by quantitative re-al-time reverse transcriptase-polymerase chain reaction ( qRT-PCR) and Western blot, respectively. Results Y53 greatly reduced the fasting blood glucose ( FBG ) and glycosylated hemoglobin( GHb) , improved diabet-ic symptoms such as polyphagia and polyuria in the di-abetic mice( P<0. 01 vs DM control group) . Y53 po-tently reduced the blood urea nitrogen ( BUN ) , serum creatinine( Scr) , 24 h urinary protein, kidney index, serum and kidney advanced glycation end-products ( AGEs) and nitric oxide( NO) , as well as kidney cho-lesterol( CHO ) and triglyceride ( TG ) contents ( P <0. 05 or P<0. 01 vs DM control group) . In the patho-logical examination, Y53 greatly restored kidney mor-phology and suppressed glomerular sclerosis( P<0. 001 vs DM control group). In addition, Y53 significantly reduced the renal expression of fibrosis-related genes, such as the transforming growth factor-β1 ( TGF-β1 ) and smad2(P<0. 01 vs DM control group). The reno-protective efficacies of Y53 were superior to those of BBR and ROSI in our study ( P<0. 05 or P<0. 01 ) . Conclusions The BBR analogue Y53 has potent ac-tivities in ameliorating renal injury and restoring renal function in STZ-induced diabetic mice. Y53 may be developed as a novel kind of agent for the treatment of DN in the future.
6.Construction and application of pharmacophore model of benzoylurea derivatives as beta-tubulin inhibitors.

Ten pharmacophore models of beta-tubulin inhibitors were established from the training set of seventeen beta-tubulin inhibitors (two categories) with comformer analysis by using the Catalyst software. The optimal pharmacophore model with two hydrophobic units and two hydrogen bond acceptor units were confirmed (RMS = 0.43, Correl = 0.98, Weight = 2.06, Config = 15.97). This pharmacophore model is able to predict the activity of known beta-tubulin inhibitors and can be further used to identify structurally diverse compounds with higher activity.
7.Study on the results in patients treated posterior correction and fusion for degenerative lumbar scoliosis

Objective To investigate the surgical strategy for degenerative lumbar scoliosis (DIS)and evaluate the clinical results of the posterior correction and fusion technique. Methods From May 2003 to May 2009,27 patients with DIS were treated posterior operation. All the patients were performed posterior laminectomy,spinal fusion and correction with pedicle screws system. Among the patients, 18 cases with posterolateral bone grafting,9 cases complicated spondylolishesis with PLIF intervertebral combined with posterolateral fusion. Results All the cases were followed up for a minimum of 12 months and 88.9% (24/27) of them had excellent or good outcome, Oswestry score was(21.54 ± 1.95 )%;22 cases were followed up for 24 months and showed a rate for excellent or good results of 86.4%( 19/22),and Oswestry score was (24.26 ± 2.32)%. Oswestry scores of the postoperative 12,24 months were lower than that before operation (57.83 ± 2.71 )%. There was statistical difference in the cases preoperation and postoperation. Correction of scoliosis of the coronal plane was 47.2%. Preoperative lumbar lordosis was augmented from 23.6°± 8.3° to 43.8°± 9.1°. One case complicated with wound infection during perioperation period, and 2 eases with heart diseases. The screw of 1 case was loosening at 3 months postoperation. Conclusion Decompressing thoroughly and improving lumbar lordosis, the operation of posterior correction and bone graft fusion shows good results for degenerative lumbar scoliosis.
8.Treatment of primary liver cancer with preoperative radiotherapy and postoperative brachy radiotherapy

Objective To evaluate the effect of preoperative radiotherapy and postoperative brachy radiotherapy (POBRT) on patients with primary liver cancer(PLC). Methods 50 patients with PLC were randomly divided into 2 groups:(1)Radiotherapy group, 25 patients who underwent preoperative radiotherapy 14~17 days before hepatectomy and POBRT 3~10 days after hepatectomy ;(2)Control group,25 cases who underwent hepatectomy only. In radiotherapy group, before heptectomy, the single-dose 6Gy per time and 3 sessions were given to each patient. 3~6 ductus were placed for POBRT during operation,and 10Gy of POBRT per time and 2~4 sessions were given postoperatively. 9.The clinical application of modified rhytidectomy incision in superficial parotid tumor surgery.

OBJECTIVE:
To evaluate the modified rhytidectomy incision in superficial parotidectomy.
METHOD:
Thirty-five patients with superficial parotid tumor were included in this study. A modified rhytidectomy incision often used in facial plastic surgery was used for superficial parotidectomy and subtotal superficial parotidectomy with preservation of facial nerve and great auricular nerve. The follow-up study included the exposed region, the cosmetic effect of this approach and the rate of complication.
RESULT:
All patients healed without salivary fistula, and were satisfied with this modified approach. Temporary paralysis of the marginal mandibular branch of facial nerve were found in five patients, and six patients felt insensible around earlobe after operation. They all recovered in 1 to 3 months after surgery, no recurrence was happened during follow-up in 36 to 60 months (median follow-up period was 48 months).
CONCLUSION
The modified rhytidectomy incision provided good exposure, had less complication and better cosmetic outcome.

10.Monitoring tumor response to antiangiogenic treatment by integrating of dynamic contrast enhanced MRI, diffusion weighted imaging and optical imaging in animal model

Objective To evaluate the response of the lung tumor xenografts in nude mice to antiangiogenic treatment from perspectives of anatomic,vessel function,cellular and molecular level using the multimodality imaging techniques including optical imaging,dynamic contrast enhanced MRI(DCE-MRI)and diffusion weighted imaging(DWI).Methods The green fluorescent protein(GFP)was transplanted labeled using GFP-expressing NCI-H460 cells.After the transfection of GFP,NCI-H460 cells were implanted subcutaneously into nude mice.Ten days after implantion,12 nude mice whose tumor xenografts grew to 0.5-1.0 cm in the maximum diameter were randomly divided into 2 groups,and injected with phosphate-buffered saline and recombinant human endostatin respectively.Then the nude mice in the two groups underwent optical imaging,DCE-MRI and DWI.The volumes,photon counts,the quantitative MR vessel functional parameters including volume transfer constant(Ktrans),rate constant(Kep),volume of extravascular extracellular space(Ve)and maximum area under the enhancement curve(iAUC),and apparent diffusion coefficient(ADC)values of the tumors were recorded.Then tumors were collected and observed using the transmission electron microscopy and pathology examination,including HE staining,microvessel density(MVD)and the expressions of vascular endothelial cell growth factor(VEGF).The Kep and VEGF expressions in experimental group and control group were compared with x2 text,and other values were compared with t test.The Pearson and Spearman test were used for analyzing the correlation of values in the two groups.Results Seven days after inoculation,the fluorescence signals were detected and grew with the growth of the tumors.On the 7 day after starting therapy,the photon counts of experimental group and control group were(2.51 ± 2.43)× 1010(photon/sec)and(5.77 ± 3.25)× 1010(photon/sec),respectively with no significant differences(t =1.964,P >0.05).Two sample t test showed that the tumor volumes in experimental group were smaller than those in control group[(365 ± 56)vs(987 ± 265)mm3,t =0.001,P < 0.01].There was a positive correlation(r =0.673,P < 0.05)between the photon counts and the volumes of the tumors.The mean Ktrans,Kep,Ve and iAUC of experimental group were:(0.055 ±0.012)min-1,0.335(0.184—0.894)min-1,0.297 ± 0.041 and 7.334 ± 3.930,and those for control group were:(0.117 ± 0.027)rin-1,0.417(0.324-1.736)min-1,0.326:±:0.062 and 13.280 ± 4.245.There were significant differences of Krans and iAUC(t =5.155,2.518,P < 0.05)between experimental group and control group.And there was a positive correlation(r =0.715,P < 0.0 1)between the values of iAUC and MVD,but not the expressions of VEGF(r =0.484,P > 0.05).The values of ADC in experimental group were higher than that in control group[(791 ± 38)× 10-6 vs(737 ± 43)×10-6 mm2/s],and there were significant differences(t =-2.299,P < 0.05).Two sample t test showed that the MVD in experimental group were lower than that in control group[(11.9 ± 4.8)vs(19.2 ±4.3)item/hpf,t =2.774,P < 0.05].The VEGF expressions in experimental group were lower than that in control group(x2 =4.000,P > 0.05).It was observed that some cells in experimental group had degenerated and apoptotic signs by the electron microscopy.Conclusions Evaluating the response of lung tumor xenografts to antiangiogenic treatment at anatomical,vessel functional,cellular and molecular level using the multimedality imagings is applicable.And it will be in favour of evaluating the therapeutic effect promptly.
