1.Expression and clinical significance of bone morphogenetic protein 3 in hi-lar cholangiocarcinoma tissues

[ ABSTRACT ] AIM: To investigate the expression and clinical significance of bone morphogenetic protein 3 (BMP3) in hilar cholangiocarcinoma tissues.METHODS: Thirty cases of hilar cholangiocarcinoma specimens were col-lected.The expression of BMP3 at mRNA and protein levels in the tumor tissues and paracancerous tissues was detected by real-time PCR and Western blot.The hilar cholangiocarcinoma paraffin-embedded specimens (n=103) were collected. The protein expression of BMP3 was determined by immunohistochemical method, and the relationship of BMP3 protein ex-pression with clinical pathological characteristics was evaluated.RESULTS:In the 30 patients with hilar cholangiocarcino-ma, the expressions of BMP3 protein and mRNA in 22 cases of tumor tissues were significantly decreased compared with the adjacent normal tissues.The results of immunohistochemistry showed that 87 cases were negative and 16 cases were weakly positive in all 103 cases of hilar cholangiocarcinoma.The expression of BMP3 protein was associated with the tumor TNM staging, lymph node metastasis and tumor differentiation (P<0.05).CONCLUSION:BMP3 gene might be inhibited in human hilar cholangiocarcinoma.The down-regulation of BMP3 gene might be associated with the carcinogenesis and devel-opment of hilar cholangiocarcinoma.
2.The preparation of recombinant adenovirus Ad-Rad50-GFP and detection of the optimal multiplicity of infection in CNE1 transfected hv Ad-Rad50-GFP.

OBJECTIVE:
The optimal multiplicity of infection (MOI) of the recombinant adenovirus Ad-Rad50-GFP carrying a mutant Rad50 gene expression region on the cell growth of nasopharyngeal carcinoma and the viral amplification efficiency of CNE1 cell infected by this adenovirus were studied.
METHOD:
The biological titer of Ad-Rad50-GFP was measured by end point dilution method. The impact of recombinant adenoviral vector transfection on the growth of CNE1 cells was observed by cell growth curve. Transfection efficacy of recombinant adenoviral vector was observed and calculated through fluorescence microscope. The expression f mutant Rad50 in the Ad-Rad50-GFP transfected CNE1 cells with optimal MOI was detected by Western Blot after transfection.
RESULT:
The biological titer of Ad-Rad50-GFP was 1.26 x 10¹¹ pfu/ml. CNE1 cell growth was not influenced significantly as they were transfected by recombinant adenoviral vector with MOI less than 50. Transfection efficacy of recombinant adenoviral vector was most salient at 24 hours after transfection, with the high expression of mutant Rad50, and the efficiency still remained about 70% after 72 hours.
CONCLUSION
Recombinant adenoviral vector Ad-Rad50-GFP could transfect CNE1 cells as well as result in the expression of mutant Rad50 in CNE1 cells effectively. MOI = 50 was the optimal multiplicity of infection of CNE1 cells transfected by recombinant adenoviral vector Ad-Rad50-GFP.

3.Evaluation of the effect of health management in patients with metabolic syndrome by family doctors

Objective To explore the effect of the health management service in patients with metabolic syndrome (MS) by family doctors. Methods A total of 417 patients with MS were screened during a health examination and divided into the intervention and control groups (206 and 211 cases, respectively), using a random digital table. Based on routine health follow-up, the intervention group received increased health management intervention by the family doctor, while the control group was only had followed-up with routine health check-ups.After 1 year,the changes in body mass index(BMI),blood pressure, triglyceride (TG) levels, fasting blood glucose (FBG) levels, and high-density lipoprotein cholesterol(HDL-C)in the two groups of patients were observed.The effect of intervention was analyzed using the paired design t-test and chi-square test.Results In the intervention group,the BMI(25.04±2.92)kg/m2, systolic blood pressure(121.5±15.6)mmHg(1 mmHg=0.133 kPa),TG levels(2.02±1.24)mmol/L,and FBGlevels (5.92±1.75)mmol/L after the intervention,were lower than those before the intervention(25.66±2.64)kg/m2, (128.4±16.5)mmHg,(2.43±1.06)mmol/L,(6.34±1.62)mmol/L;respectively,and these differenceswerestatistically significant(t=2.437,3.598,3.536,2.983,respectively;P<0.05).HDL-C levels after the intervention(1.32± 0.26) mmol/L were higher than thosebefore the intervention (1.26 ± 0.32) mmol/L, and the difference was statistically significant(t=-2.218,P<0.05).In the intervention group,blood pressure,BMI,FBGlevels,and HDL-C levels after the intervention were higher than the normal reference values.The proportion of patients with normal reference valuesafter the intervention were 62.6%, 75.2%, 14.6%, and 52.9%, respectively, which were lower than those before the intervention(92.7%, 85.4% 23.3%, and 63.6%, respectively);thesedifferenceswerestatistically significant (χ2=53.795, 6.777, 5.124, and 4.832, respectively;P<0.05). After 1 year,the control group had lower BMI(24.52±2.69)kg/m2than that before the intervention(25.79± 2.85) kg/m2; thisdifference was statistically significant (t=2.421, P<0.05).There were no significant differences in the systolic and diastolic blood pressure, TG levels, HDL-C levels, and FBG levels. Conclusion Health management byfamily doctors can effectively reduce weight, blood pressure, blood lipids,and blood sugar by interfering with the lifestyle of patients with MS.
