Objective To establish a matrine delivery system in vitreous is very important for the dynamic treatment of proliferative vitreoretinopathy(PVR) . Present study was to evaluate the efficacy of matrine polyactic acid microsphere(MAT-PLA-MS) in prevention of PVR. Methods The suspension of cultured fibroblasts was injected into vitreous cavity of 30 healthy adult New Zealand albino rabbits to induce PVR. Then the experimental rabbits were divided into 3 groups and 10 rabbits for each. The animals received intravitreal injection of 0.3 mL MAT-PLA-MS(4 mg) matrine in MAT-PLA-MS group. Free matrine normal sodium solution 0.3 mL(containing 2mg matrine) was injected in vitreous cavity in free matrine group. 0. 3 mL normal saline solution was injected into the vitreous of the left eyes and the equivalent volume of blank polyaetic acid microsphere(blank-PLA-MS) into the right eyes in control group. The changes of cornea, aqueous humor, lens, vitreous and fundus were examined and recorded by slit lamp biomicroscope, indirect ophthalmoscope, fundus color camera and B ultrasonogram on the 1st, 3rd, 7th, 14th, 21st, 28th and 35th day following injection of drug. The inhibition effect of matrine on PVR was evaluated according to Ryan' s grading criteria of PVR. Results On the 14th days after implantation of MAT-PLA-MS, the rate of retinal detachment was 60%, 10%, 5% and 60% in normal saline group, free matrine group, MAT-PLA-MS group and blank-PLA-MS group respectively. Statistically significant difference was found among normal saline group, blank-PLA-MS group, MAT-PLA-MS group and free matrine group(P <0. 05). On the 21st day after injection of fibroblasts, the morbidity of retinal detachment was 80%, 30%, 10% and 80% in normal saline group, free matrine group, MAT-PLA-MS group and blank-PLA-MS group respectively, showing a significant difference among different groups. On the 28th day, the incidence rate of retinal detachment was 90%, 50%, 15% and 90% respectively, presenting statistical difference among various groups (P < 0. 05) as well as between free matrine group and MAT-PLA-MS group (P<0. 05). On the 35th day, considerably difference also was seen in the morbidity of retinal detachment among various groups (90%, 60%, 15% and 90% respectively) (P<0.05). Conclusion Implantation of MAT-PLA-M S into vitreous cavity can effectively inhibit the development of PVR induced by fibroblasts in rabbit model.
3.A comparison between arthroscopic and open retrograde intramedullary interlocking nails in the treatment of femoral supracondylar fracture

Objective To compare the curative effect between arthroscopic and open retrograde intramedullary interlocking nails in the treatment of femoral supracondylar fracture. Methods A total of 46 patients with femoral supracondylar fracture underwent the treatment by either arthroscopic (19 patients, Group A) or open (27 patients, Group B) retrograde intramedullary interlocking nails. Results The operation time, surgical blood loss and bone union time in the Group A were less than those in the Group B ( t =-4 193, -21 517 and -4 055, respectively; P =0 000). Follow-up for (10 3?3 6) months in the 46 patients revealed the results of therapy were, according to the Kolmert criteria, excellent in 15, good in 3, fair in 1 in the Group A while excellent in 7, good in 14, fair in 4 and poor in 2 in the Group B, without statistically significances between the two groups in the “good” or “excellent” rates (94 7% vs. 77 8%; ? 2 =1 345, P =0 246). Conclusions Use of arthroscopic retrograde intramedullary interlocking nails has advantages of shorter operating time, less blood loss and quicker bone union, benefiting the recovery of knee joint functions.
4.Protective effects of NADH on cardiac fibroblasts apoptosis induced by hydrazine

Objective To explore the protection effects of the reduced form of nicotinamide-adenine denucleotid(NADH) on cardiac fibroblasts apoptosis induced by hydrazine in vitro.Methods Cardiac fibroblasts isolated from neonate rats were divided into four groups.In NADH pretreatment before hydrazine treatment group,the cardiac fibroblasts were cultured first in the medium containing 400?g/ml NADH for 2 hours,and then cultured in the medium containing 400?g/ml NADH plus 2mmol/L hydrazine for 72 hours;in hydrazine treatment group,the cardiac fibroblasts were cultured first in normal medium containing 400?g/ml NADH for 2 hours and then cultured in the medium containing 2mmol/L hydrazine for 72 hours;in control group,the cardiac fibroblasts were cultured only in normal medium for 72 hours;and in NADH pretreatment control group,the cardiac fibroblasts were cultured in medium containing 400?g/ml NADH for 72 hours.After the treatments mentioned above,the changes in nuclear of apoptotic cardiac fibroblasts as stained by Hoechst33258 were observed under fluorescence microscope,the apoptotic and necrosis rates of cardiac fibroblasts stained by AnnexinⅤ/PI were assayed by flow cytometry,the mitochondria membrane potential of cells stained by rhodamine123 was also assayed by flow cytometry.Results Hydrazine could induce apoptosis of the cardiac fibroblasts.After NADH pretreatment,the number of apoptotic cardiac fibroblasts with pyknosis of nuclear was lowered,the rates of apoptosis and necrosis decreased,and the mitochondria membrane potential of cells was elevated.Conclusion NADH pretreatment could reduce the apoptosis induced by hydrazine by improving the function of mitochondria.
5.Effect of salidroside on Akt/GSK-3βof myocardium with ischemia-reperfusion injury in rats

Objective Ischemic preconditioning and postconditioning can provide certain protection for myocardium .The ar-ticle was designed to observe the protective effect of salidroside on myocardial ischemia reperfusion injury ( MIRI) and explore its mech-anisms. Methods SD rats were randomly divided into 6 groups with 6 rats for each: sham operation group (S group), ischemia-reperfusion group(I/R group), salidroside preventive group(salidroside treatment followed by ischemia-reperfusion), salidroside treat-ment group (ischemia-reperfusion followed by salidroside treatment ), salidroside preventive+LY group(LY294002 preventive group) and salidroside treatment+LY group(LY294002 treatment group).Salidroside was administered once a day for three days before mod-elling in both salidroside preventive group and LY 294002 preventive group;while salidroside was given immediately after the reperfu-sion in both salidroside treatment group and LY 294002 treatment group .The same volume of NS was administered only to the rats in S group and I/R group.The PI3K inhibitor(LY294002) was given additionally 35 mins before LAD ligation in both LY294002 preventive group and LY294002 treatment group .All injections were given intraperitoneally .Akt, p-Akt, GSK-3βand p-GSK-3βin myocardium were examined with immunocytochemical method in all groups .The protein expression and phosphorrylation status of Akt /GSK-3βwere determined by western blot. Results The levels of Akt/GSK-3βin myocardium of S group(0.246 ±0.002), I/R group(0.457 ± 0.012), LY294002 preventive group(0.303 ±0.005), LY294002 treatment group(0.361 ±0.019) decreased significantly in com-parison to those of salidroside preventive group (0.857 ±0.014) and salidroside treatment group(0.683 ±0.009)(P<0.05).The levels of p-GSK-3β/GSK-3βin myocardium of S group (0.137 ±0.004), I/R group(0.380 ±0.026), LY294002 preventive group(0.148 ± 0.013), LY294002 treatment group(0.267 ±0.050) also decreased significantly in comparison to those of salidroside preventive group (0.944 ±0.045) and salidroside treatment group (0.895 ±0.043) (P<0.05).As to the comparison between salidroside preventive group and salidroside treatment group , there was no significant difference (P>0.05). Conclusion The result indicates that salidro-side protects myocardium against MIRI in rats .The cardioprotective effect might be associated with the increased protein expression and the phosphorylation rate of Akt/GSK-3β.
6.The value of MSCT in diagnosing bowel ischemia of small-bowel obstruction

Objective To investigate the value of MSCT in assessing bowel ischemia of small-bowel obstruction .Methods CT images and electronic medical records of 40 patients with small-bowel obstruction were retrospectively evaluated.Patients were treated by surgery.The CT signs of bowel ischemia were recorded.Relationship between CT signs and bowel ischemia were tested by Fisher exact andχ2 test.Sensitivity and specificity of MSCT for ischemia were also assessed.Results Bowel ischemia was confirmed at surgery and/or pathological examination in 21 of 40 patients.Diminished enhancement after contrast agent injection was the most common sign in bowel ischemia patients.The signs of increased bowel-wall attenuation on non enhanced images and diminished enhancement after contrast agent injection were significantly associated with ischemia (P <0.000 1).The signs of increased bowel-wall attenuation on non enhanced images and pneumatosis intestinalis had high sensitivity(100%).The sign of diminished enhancement after contrast agent injection had high sensitivity(95.2%)and specificity(94.7%).Conclusion MSCT is accurate in assessing bowel ischemia of small-bowel obstruction, and improves the timeliness and diagnosis for this disease.
7.The Value Of MR Dualecho Imaging in Differential Diagnosing Liver Benign,Malignant Lesions

Objective To evaluate MR Dual echo imaging in the differential diagnosing liver lesions.Methods:MR Dual echo features in 80 patients with different liver lesions,including 40 cases of benign lesions and 40 cases of malignant lesions,were analyzed,and comparison with MR Dual echo imaging was made.Results MR Dual echo sensitivity and specificity were 85% and 95% in the benign lesions,80% and 94% in the malignant lesions respectively.Conclusion MR Dualecho imaging is a useful MR technique,which is helpful in differentiating benign from malignant lesions.
8.Protective effect of nicorandil on myocardial ischemia reperfusion injury in patients undergoing cardiac valve replacement

AIM:To observe the protective effect of nicorandil on myocardial ischemia reperfusion injury in patients undergoing valve replacement.METHODS:Sixty patients undergoing valve repalacement were randomly divided into 3 groups:control(Ⅰ)group,nicorandil delayed precondictioning(Ⅱ)group and nicorandil preconditioning(Ⅲ)group.20 mg nicorandil was given i.v.24 h before operation in group Ⅱ,whereas in group Ⅲ,20 mg nicorandil was given i.v.after induction of anesthesia.Blood samples were taken from coronary venous for determination of cTnI,TNF-?,IL-6 levels at aortic clamping(T0),15 min(T1),30 min(T2),60 min(T3),90 min(T4)after aortic declamping.Right atria myocardium tissue were taken at T0 and T2 to observe the histopathological changes with electron microscopy.RESULTS:The levels of cTnI,TNF-?and IL-6 in group Ⅱand Ⅲ were significantly lower than those in group I.Myocardium injury was obviously lighter in groupⅡand Ⅲ than that in group Ⅰ,whereas the levels of cTnI,TNF-?,IL-6 and myocardium injury were lower in group Ⅱ than those in group Ⅲ.CONCLUSION:Nicorandil is effcetive in decreasing myocardial ischemia repefusion injury in patients undergoing valve replacement,The protective effect of nicorandil delayed preconditioning is more obvious than precondictioning.
9.Changes of resting heart rate in patients with essential hypertension and its clinical significance

Objective To study the relationship between resting heart rate(RHR)and essential hypertension(EH).Methods 215 patients with EH and 82 healthy subjects underwent RHR detection.The relationship between RHR and the state of EH was analyzed.Results RHR in EH group was significantly higher than that in normal group[(78.9±9.1)bpm vs(69.5±10.9)bpm,P<0.01)],RHR in different stages of EH was significantly different(F=21.280,P<0.01,respectively),and increased with grades(P<0.05 orP<0.01).RHR in EH patients complicated with left ventricle hypertrophy(LVH)was higher than those with no LVH[(83.9±9.1)bpm vs (75.9±8.9)bpm,P<0.01].There was positive correlation between RHR and LVH(r=0.8097,P<0.01).Conclusion RHR is related to the development and progress of EH,and it may be one of the index for estimating the state of EH.
10.Tumorigenicity of rat bone marrow-derived liver stem cells

BACKGROUND: Mobilizing autologous or extraneous bone marrow-derived liver stem cells may promote liver regeneration, however, its safety before the large scale clinical application needs further evaluation.OBJECTIVE: Bone marrow-derived liver stem cells (BDLSCs) were induced by culturing the rat bone marrow mesenchymal cells in the medium containing 5% cholestatic sera, and then were implanted into nude mice to observe the tumorigenicity. METHODS: Rat bone marrow mesenchymal cells (BMSCs) were isolated and incubated in the medium containing 5% cholestatic sera. Immunofluorescent stain was used to detect the expression of albumin, alpha-fetoprotein and cytokeratin18 by the cultured cells. Glycogen and urea synthesis by these cells were analyzed, respectively. BDLSCs following 14 days of culture were incubated in the skin of nude mice to observe neoplasia in local site. RESULTS AND CONCLUSION: Rat BMSCs survived in the medium containing 5% cholestatic serum and formed into small colonies on the fourth day after culture. Seven days later, the colonies expanded and there appeared some polygonal cells in the peripheral area. About 14 days later, these polygonal cells were confluent and presented the shape of cobblestone. Immunofluorescent stain showed that these cells expressed cytokeratin18, albumin and alpha-fetoprotein. Staining for glycogen displayed that glycogen granules were seen in cells. From 12 to15 days after culture, urea nitrogen concentrations in the medium were gradually increased. Rat BDLSCs were incubated in the skin of nude mice. Thirty days later, no neoplasia was found in the local site, and the tissue structure was normal. This result indicated that rat BDLSCs induced with the medium containing 5% cholestatic serum might have not tumorigenicity.
