OBJECTIVE: To investigate the application value of computed tomograph( CT) guided percutaneous lung biopsy in the diagnosis of occupational pneumoconiosis( the following is referred to as “pneumoconiosis”). METHODS: A total of63 cases of CT-guided percutaneous lung puncture biopsy from patients with pneumoconiosis, were analyzed for the successful rate of percutaneous lung puncture, the positive rate of pneumoconiosis pathological results and diagnostic accuracy. RESULTS: The successful rate of puncture was 100. 0% in these 63 cases. Pathological examination revealed60 cases of pneumoconiosis like change, with a positive rate of 95. 2%. Among them,56 cases were diffuse fibrosis,48 cases were dust deposition, and 32 cases were dusty nodules. There were 5 cases of tuberculosis granuloma,1 case of tumor,1 case of macrophage pneumonia and 1 case of infection. According to the pathological examination results,54 cases of simple pneumoconiosis,1 case of occupational hard metal lung disease,1 case of pneumoconiosis combined with tumor,4 cases of pneumoconiosis combined with tuberculosis and 2 cases of simple tuberculosis were confirmed. The diagnostic accuracy was 98. 4%. CONCLUSION: CT guided percutaneous lung biopsy is an ideal clinical method for obtaining pathological specimens of lung tissue. The positive rate of detecting pneumoconiosis pathological changes is high. The accuracy of pneumoconiosis diagnosis is high. It has high application value in the diagnosis and differential diagnosis of pneumoconiosis.

Objective To establish a rapid detection method for zika virus based on direct amplification re-al-time fluorescent quantitative reverse transcription polymerase chain reaction(RT-PCR)technique.Methods A direct amplification RT-PCR technique for the rapid detection of zika virus in 5 samples(whole blood, serum, saliva, throat swab and urine)was established by using a special function DNA polymerase and a preferred PCR enhancer. Results The detection limits of the 5 samples were 103 PFU/mL in serum,102 PFU/mL in urine, throat swab, and saliva, and 104 PFU/mL in whole blood. The coefficient of goodness-fit of stand-ard curves was above 0.98,and the amplification efficiency was 90%-110%.Zika virus nucleic acid was suc-cessfully amplified, but non-zika virus nucleic acid was not amplified.Based on the repeatable detection of sam-ples from urine, whole blood, and saliva, the variation coefficient of 6 repeated Ct values at 106 PFU/mL and 102 PFU/mL concentrations were all<5%.The zika virus detection method established by the direct amplifi-cation RT-PCR technique was consistent with the detection results of conventional RT-PCR technique.Only two serum samples were detected in eight zika virus samples, and the remaining 62 non-zika virus samples and 12 negative samples were not amplified. Conclusion A rapid detection method for zika virus based on direct ampli-fication RT-PCR technique is successfully established. The method is simple, rapid, sensitive and specific.

Objective:
To investigate the effect of PM2.5 exposure on the thyroid function in male SD rats.
Methods:
Adult Sprague-Dawley rats were divided into " filtered air" (FA) and " PM2.5" groups. These rats exposed to the 2 kinds of air for 12 weeks were killed and their plasma and thyroid tissues were taken for investigation. The morphological status and pathological changes of thyroid tissue were observed after H&E staining. The levels of triiodothyronine (T3), free triiodothyronine (FT3), thyroxine(T4), free thyroxine (FT4), anti-thyroglobulin antibody (TGAb), anti-thyroid peroxidase antibody (TPOAb), thyrotropin (TSH), tumor necrosis factor α (TNF-α), and interleukin-6 (IL-6) in rats were measured by ELISA.
Results:
The results showed that there was no significant difference in body weight between two groups after 12 weeks of exposure to either filtered air or PM2.5. No pathological changes and significant differences were observed in the thyroid tissues of two groups after HE staining. There were no significant differences in plasma levels of T4, T3, FT3, TNF-α, and IL-6 between two groups. However, compared with rats exposed to filtered air, rats exposed to PM2.5 had significantly lower plasma FT4[(3.81±1.23)
