Objective To explore the imaging characteristics of intrathoracic extramedullary hematopoiesis(EMH).Methods Clinical and imaging findings of 6 cases with EMH were retrospectively analyzed, and the imaging characteristics, diagnosis and differential diagnosis were discussed. Results Among 6 the cases of EMH,3 lesions were located in mediastinum, and the other 3 lesions in mediastinum and adjacent chest wall.Five cases appeared as globular shadows which protruded toward the lung fields on X-ray films,on plain CT scan, all the 6 lesions appeared of smooth margin and homogeneous soft-tissue density, and the CT values ranged from 38 to 45 HU.Two lesions showed slight homogeneous enhancement on postcontrast CT scan using the common CT scanner, and the CT values ranged from 61 to 65 HU.Four lesions showed significant homogeneous enhancement on postcontrast CT images using the 16-slices CT,and the CT values ranged from 72 to 83 HU.On MRI images,4 lesions showed the same signal intensity as compared to adjacent muscles on T_1WI and T_2WI,and there was slight enhancement of the lesions after intravenous contrast administration. Conclusion EMH has specific imaging findings, and accurate diagnosis can be made by combining X-ray,CT and MR imaging findings with clinical history.

A fragment sized 400bp of White spot syndrome virus(WS SV,formerly de signated NOSV),recovered from recombinant plasmid pAFD, was labeled with Digox igenin as a probe to detect dynamic distribution of WSSV within 120h and 72h in crawfishes(Cambarus proclarkii) inoculated WSSV by oral taking and injecti on r espectively. Stomach epithelium, intestine epithelium, heart, gill, haemolymph, muscle, hepatopancreas, hypoderm, connective tissue and ovary of infected crawfi shes were examined for WSSV. In both groups, WSSV was first detected in heamoly mph at 12h p.i. and then disappeared. Again it was detected at 96h p.i. only in oral infection group and maintained till 120h p.i., but it didn't appear at 72h p.i. in injection group. WSSV in heart, muscle was detected at 36h p.i. in oral infection group and 24h p.i. in injection group respectively, and then increased generally. In addition, WSSV in intestine epithelium, connective tissue, ovary of oral infection group and intestine epithelium, hypoderm, ovary of injection g roup could also be detected. In dead crawfishes after 120h and 72h p.i. in two groups, WSSV could be detected in all the examined tissues and it demonstrated t hat systemic infection occurred in the animales. The tissue containing more amo unts of WSSV was hypoderm in oral infection group, while intestine epithelium, g ill, hypoderm, ovary in injection infection group. It deduced that WSSV first a ppears in haemolymph and then goes into heart, muscle and other tissues and prol iferates in them. Once again, WSSV is released into heamolymph resulting in syst emic infection till crawfishes' death.

Objective To construct an expression system to produce the virus-like particles containing a part of the sequence of PSA mRNA, which are ribonuclease-resistant due to the encapsulation of the mRNA by bacteriophage MS2 coat proteins. Methods The PCR products of PSA cDNA fragments were cloned to TA vector pBS-T, then the targeted segments could be obtained when the pBS-T-PSA were digested by restriction endonuclease Hind Ⅲ and cloned to prokaryocytic expression vector pNCCL1. The recombinant plasmids named PNCCL1-PSA were transfected into E. Coli BL21-DE3 and induced to express with IPTG. Results The recombinant plasmids were successfully constructed. The bacteriophage MS2 coat protein which expressed in BL21 can self- assemble to form ribonuclease resistant virus-like particles and the PSA mRNA was encapsulated into virus-like particles. Conclusions The virus-like particle containing PSA mRNA can be expressed in prokaryocyte and it can be used as standard and control in detecting PSA mRNA. It provides a new, stable and ribonuclease-resistant RNA standard in RNA detection.
