Objective:To investigate the effects of Nano-chitosan rhBMP-2 Gel(NCS/BMP-2 Gel)on the repair of mandibular de-fect.Methods:NCS/BMP-2 Gel and NCS Gel were prepared and respectively injected into the subcutaneous space on both sides of dorsum of 9 rats.3 rats were respectively sacrificed 10,20 and 30 d after injection. The subcutaneous nodules were histologicaly ex-amined.Mandibular defect was made in 54 SD rats and the rats were divided randomly and into 3 groups(n=18):NCS/BMP-2 Gel was implanted into the defects in group 1,NCS Gel in group 2,no injection in group 3.Animals were sacrificed 3,6 and 9 weeks after transplantation.X-ray examination, pathologic observation were conducted. Results:Subcutaneous nodules were found in both sides of the rat dorsum. The residual mandibular defect area in study group 1 was apparently smaller than that in group 2 and 3(P<0.05). More new bone formation was observed in the gel injected area in group 1 than in group 2 and 3.Conclusion:Nano-CS/BMP-2 Gel is biocompatible and can accelerate the healing of mandibular defect.

BACKGROUND:Absorption and atrophy of the alveolar ridge always happen after tooth extraction. There are stil a lot of difficulties to be worked out to prevent the atrophy of the alveolar ridge. OBJECTIVE:To investigate the effect of the nano-chitosan/bone morphogenetic protein hydrogel on preventing absorption of the residual alveolar ridge of the rats. METHODS:Total y 54 Sprague-Dawley rats were divided randomly and equal y into three groups. A model of residual alveolar ridge was made by extraction of the left central incisor. Nano-chitosan bone morphogenetic protein hydrogel was implanted into the rat’s alveolar fossa of the experimental group;the alveolar fossa of the control group was implanted with nano-chitosan hydrogel;the alveolar fossa of the blank group was implanted with nothing. Rat’s mandible was ful y dissected after 3, 6, 9 weeks, respectively. Molybdenum target X-ray examination and pathological observation were evaluated. RESULTS AND CONCLUSION:At weeks 3, 6, 9 postoperation, the residual alveolar ridge was absorbed to different degrees, which was milder in the experimental group than the cotrol group and blank group (P<0.05). After 3 weeks, in the blank group, less new bone formed, but bleeding and a large amount of inflammatory cells were visible; in the control group, a few of osteoid tissues and lymphocytes were found; in the experimental group, new bone formed and a small amount of inflammatory cells were observed. After 6 weeks, new bone formed obviously in the experimental group, a little new bone formed in the control group, but less new bone formed in the blank group. After 9 weeks, a large amount of new bone formed in the experimental group with good osteogenic ability; only a few of new bone formed in the control and blank groups. Nano-chitosan/bone morphogenetic protein hydrogel can be used to prevent the absorption of the residual alveolar ridge after tooth extraction.

Objective: To explore the effect of iodine overdose on cathepsin B and D in thyroidcell line (FRTL). Method: The cells were treated with potassium iodide (0, 1, 5, 10, 50 and 100mmol/L). After treatment 6, 12, 24, 48 and 72 hours, the cells were collected. The activities of cathepsin B (CB) and cathepsin D (CD) were assayed by fluorospectrophotometry and Lowry, respectively. The mRNA levels of CB and CD were determined with real-time PCR. Results: After 12 and 24 hours, the activities of CB and CD, treated by 50mmol/L potassium iodide were decreased significanty. The mRNA levels of CB and CD of 50mmol/L potassium iodide group were decreased significanty after treatment 24 hours. Conclusion: The decreased activities of CB and CD were one of reasons of thyroid colloid retention resulted from iodine excess.

Objective: To detect the damage of excessive iodine on the activities and mRNA of cathepsin B and D and observe the effects of selenium intervention. Method: Thyroid follicular epithelial cell line FRTL was cultured in Kaighn’s modified Ham’s F-12 medium. ① The cells were divided into 8 groups for detection of activities of cathepsin B and D (CB and CD). The medium of NI group did not contain selenium or iodine, while EI group contained 50 mmol/L KI, and IS group contained 50mmol/L KI and different dosages of 0.1,0.2,0.5,1.0,2.0 and 5.0 ?mol/L selenium (sodium selenite). ②The mRNA levels of cathepsin B and D were detected by real-time PCR. The medium of NI group and EI group was same as above, but IS group only contained 0.1?mol/L selenium besides 50mmol/L KI. Results: The activities of CB decreased in EI group (50mmol/L KI treated 12 h), while CD decreased after 24 h and both activities were improved in IS groups with 0.1~0.5?mol/L selenium. The mRNA levels of CB and CD in EI group decreased, while in IS group were improved. Conclusion: Excessive iodine resulted in goiter through decreased levels of activities and mRNA of CB and CD. The intervention of selenium on goiter mayinduce its amelioration effect on decrease of CB and CD activities and the optimal dosage ranges from 0.1?mol/L to 0.5?mol/L.

Chinese Medical Journal 2013;126(16):3097-3102
BACKGROUNDHigh uric acid (UA) levels and metabolic syndrome (MS) are risk factors for atherosclerotic diseases. Brachial-ankle pulse wave velocity (baPWV) is a valid and reproducible measurement by which to assess arterial stiffness and a surrogate marker of atherosclerosis. However, little is known about the relationship between them, especially in elderly Chinese with MS components who are at high risk for atherosclerotic diseases.
METHODSOne thousand and twenty Chinese subjects (159 women) older than 60 years of age (mean age (70.6 ± 5.7) years) with at least one MS component underwent routine laboratory tests, and baPWV measurements were analyzed.
RESULTSParticipants were divided into four groups by MS components. The mean age did not significantly differ among the MS component groups. We found that not only the diagnostic factors (blood pressure, body mass index (BMI), lipids, glucose) of MS but also baPWV, UA, insulin, homeostasis model of assessment for insulin resistence index (HOMAIR) levels increased, and high density lipoprotein (HDL)-C decreased with an increased number of MS components (test for trend P < 0.05). The association between UA and baPWV was observed after adjustment for gender, age, blood pressure, BMI, serum creatinine and high density lipoprotein, and insulin resistance (r = 0.186, P < 0.0001). There were increases in the odds ratios for the association between the number of components of MS, UA and baPWV, even after adjustment for traditional risk factors. However, after adjustment for insulin or HOMA-IR, there were no significant differences in the multivariate odds ratios among the number of MS components for UA.
CONCLUSIONSThe UA level is positively associated with baPWV and MS, but the association between UA and MS is dependent on insulin resistance. Furthermore, baPWV is independently associated with MS in our study population.
