Objective The human IFN-inducible protein absent in melanoma 2 (AIM2) has been regarded as a tumour sup-pressor, however, the molecular mechanisms of its antitumor activity still remain unclear.This research is to study the molecular mech-anisms of AIM2 inducing breast cancer cell apoptosis. Methods Tet-Off induction model system was established to induce AIM2 ex-pression in great quantities, MCF-7 tTA-AIM2 cells as the experimental group and MCF-7 tTA-Luc cells as control group.Western blotting was used to detect AIM2 expression in breast cancer cell lines and subcellular localization.XTT assay was applied to analyze the effects of AIM2 on cell proliferation.Apoptosis were detected by Annexin V-FITC and propidium iodide staining, and the apoptosis mechanism were investigated by west blot. Results The established Tet-off guidance system could combine tTA to TRE, thereby promoting AIM2 gene transcription and inducing great expression of AIM2 protein.4 days after induction, the expression of AIM2 could be detected in cytoplasm and nucleus, and AIM2 expression increased with the increasing days.XTT assay detected the growth speed of MCF-7 tTA-AIM2 cell lines slowed down 6 days after induction.After abundant expression of induced AIM2, the percentage of FITC fluorescence apoptosis increased significantly (2.36%vs 14.45%, P<0.01) .Increased AIM2 expression inhibited the expression of the anti-apoptotic protein Bcl-xL, increased the expression of the apoptosis proteins Bad and Bax, and activated caspases, resulting in the cleavage of DNA repair protein PARP. Conclusion In breast cancer Tet-off induction system, AIM2 will express in cytoplasm and nucleus, influence cell proliferation, and stimulate the mitochondria to cell apoptosis.

Objective:To explore C-terminal tensin-like(CTEN) gene in breast cancer tissue and its significance. Methods:Explore the prevalence and clinical significance of CTEN expression in invasive breast cancer cases ( n = 1409 ) using immunohistochemistry and tissue microarray. Results: The staining pattern for CTEN in breast tumour cells was homogenous cytoplasmic staining. Moderate to strong cytoplasmic immunoreactivity for CTEN was observed in 90% of the studied cases(1268 cases) . CTEN expression was significantly associated with poor prognostic variables including larger tumour size(χ2=4. 254,P=0. 044), higher histological grade(χ2=7. 555,P=0. 019),axillary nodal involvement(χ2=5. 842,P=0. 035),poor Nottingham Prognostic Index (χ2=7. 578,P=0. 016),Local recurrence(χ2=5. 211,P=0. 039),Regional recurrence(χ2=4. 778,P=0. 042),Distant metastases (χ2=4. 780,P=0. 041) and Histological type(χ2=7. 634,P=0. 012). Significant associations were observed between increased CTEN expression and up-regulation of phosphorylated-Akt(P-Akt)(χ2=27. 23,P<0. 001),Phosphoinositide 3 kinase(PIK3)(χ2=37. 22,P<0. 001) and N-cadherin proteins(χ2=26. 91,P<0. 001). Kaplan-Meier survival analysis demonstrated that patients with high CTEN ex-pression had significantly shorter Breast Cancer Specific Survival(P=0. 004) and Metastasis-Free Survival(P=0. 041) than those with low-CTEN expression. Multivariate analysis showed that CTEN was not an independent prognostic marker in breast cancer. Conclusion:CTEN expression increase is a cancer gene,its association with poor prognostic parameters,and the expression of CTEN associated with poor prognosis of breast cancer parameters.

Objective To investigate whether the interleukin-10 (IL-10) gene therapy has the effect of anti liver fibrosis in mice and its mechanism.Methods Liver fibrosis was induced by long-term thioacetamide administration in mice.Human IL-10 expression plasmid was delivered via electroporation after liver fibrosis was established.The immunohistochemistry was used to study the expression of IL-10 in liver.Sircol collagen determination method was used to detect the contents of collagen in the liver.The reverse transcription polymerase chain reaction (RT-PCR) was used to detect the expressions of liver fibrosis-related genes,including transforming growth factor-β1 (TGF-β1),collagen α1,tumor necrosis factor-α (TNF-α),intercellular adhesion molecule-1 (ICAM-1),fibronectin,vascular cell adhesion molecule 1 (VCAM-1),and matrix metalloproteinase-inhibiting factor (TIMP-1,TIMP-2).Results Immunohistochemical results showed IL-10 gene therapy reversed hepatic fibrosis.Sircol collagen assay showed that IL-10 gene therapy reduced the content of collagen fibers(P ＜ 0.05).RT-PCR revealed IL-10 gene therapy reduced liver TGF-β1,TNF-α,collagen α1,cell adhesion molecule,and TIMPs mRNA upregulation.Conclusions Electroporative IL-10 gene therapy might be an effective therapeutic reagent for liver fibrosis with potential future clinical applications.

As a cavola marker protein, caveolin-1 participates in many pathophysiological processes through its scaffolding domain oligomering many celular signal transduction molecules, and also regulates inflammation after cerebral ischemia through different pathways. This article reviews advances in caveolin-1 and inflammation after ischemic stroke in recent years, mainly focusing on its mechanism in regulating inflammation.

Objective To evaluate the irreversible ischemia injury in patients with acute coronary syndrome using Cardiac MRI. Methods We have studied 35 patients with acute coronary syndromes who received standard medical therapy. Coronary angiogram confirmed that all the patients had suffered from multi-coronary artery stenosis without total occlusion. Cardiac MRI (CMRI) examinations were performed 6-12 weeks after the coronary angiography and revascularization. IR-GRE was used in the CMRI protocal to detect myocardial delayed enhancement (viability). The definition of hyperenhancement was that the signal of the region was two times that of the remote myocardium. The hyper-enhanced volume was presented as a percentage of LV mass (%LV), i.e. The total number of hyperenhanced pixels divided by the total number of pixels of the LV myocardium and multiplied by 100. Results CMRI revealed that all the patients had hyperenhanced region, of whom 30 (85.7%) had non-transmural enhancement and only 5 (14.3%) had transmural enhancement. The average size of the enhancement of non-transmural was significantly smaller than that of transmural hyperenhancement (45.7?7.3%). Conclusion The CMRI can demonstrate that infarction has occurred in patients with multi-coronary arteries stenosis.

Objective To study the cytotoxicity of the membrane material of heart assist pump. Methods The membrane materials tested incladed polyurethane (group A) and medical polychloroethylene (group B),and a blank control group with none of the material. L-929 cells (the mouse fibroblasts) were cultured on either material in groups A and B,and 6 cell cultures were without any material. After 24,48 and 72 hours,the growth of cell in groups A,B and the control was observed,and the number was counted. Educts of two materials used in groups A and B was respectively added to culture medium,then the OD was measured every 24h and proliferation rate was determined by MTT test. Results L-929 cells grew very well in groups A and B with less death. The cell counts showed no difference between 2 groups at each time. Objective:To investigate the mechanism responsible for lost sensibility of tyrosine aminotransferase(TAT)to dexam- ethasone(Dex)in human hepatoma cell line SMMC-7721 through examining the cDNA sequence of TAT and the status of glucocorticoid receptor(GR)pathway.Methods:The TAT cDNA fragment containing the full length of coding sequence was amplified by reverse transcription-polymerase chain reaction(RT-PCR)and was sequenced.The expression of TAT mRNA was determined by real-time quantitative PCR to observe the influence of Dex on expression of TAT mRNA in SMMC-7721 cells.The experiement with HepG2 cells was performed as the control.Reporter genes(GRE-tk-LUC and GRE-MMTV-CAT)were transiently transfected into SMMC-7721 cells by electroporation.The induction efficiencies of LUC and CAT genes expression by Dex were examined and compared between SMMC-7721 cells and HepG2 cells.Results:The results showed that there was a same-sense mutation(Gln576Gln)in TAT cDNA se- quence.TAT mRNA could be induced by Dex,with the maximal induction level being 2.22-folds in SMMC-7721 cells,which was signifi- cantly lower than that in HepG2 cells(15.1-fold increase,P

Objective: To observe the oral care effects of finger toothbrush. Methods: The finger toothbrush worn on the index finger was made of silica gel .It was used to brush the in vitro healthy teeth pulled out on the same day and the standard hard plaster tooth model and to brush the in vivo healthy teeth in adults; the bacteria in the finger toothbrush, The dentofacial pluque and the abrasion on the tooth model were tested respectively. Results: After in vivo use the bacteria remained in the finger toothbrush were fewer than those in the commonly used toothbrush(1 683.24?1 355.59 and 2 353.76?1 582.06 respectively). The dentofacial plaque was decreased in the same extent by the two kinds of toothbrushes. The zero rate of abrasion on the tooth model was 93.3%(28/30) by finger toothbrush and 23.3%(14/30) by the commonly used toothbrush. Conclusion: The finger toothbrush worn on finger can remove dentofacial plaque effectively and produce a lower abrasion on tooth.
