1.Study on Microwave Irradiation of Berberine from Rhizoma Coptidis

Objective Ethanol, acid and alkali were used in the traditional Bererine extracting which required quite a few extracting time and polluted the environment. Water instead of ethanol was used. Microwave irradiation was chosen as the extracting method. Methods The best parameters, microwave power, extracting time, solid-liquid ratio were got in the experiment. According to the orthogonal experimental design, the optimum extracting condition was determined. Results Microwave irradiation, compared with the traditional extracting techniques, was with short extracting time. Conclusion The method was practicable and the product possessed the virtue of high purity, safe quality and free pollution.
2.The content of thromboxane and prostacyclin in optic nerves after forehead impact injury in rabbits

Objective To observe the content of thromboxane (TXA 2) and prostacyclin (PGI 2) in optic nerves after forehead impact injury. Methods The right forehead zones of 32 rabbits were struck by biology impact machine. Tweenty four rabbits that had afferent papillary defect after injury were chosen, and randomly divided into four groups: 1 day, 2, 4, and 7 days group. Right eyes were in the experimental group and left eyes were in the control group. Flash visual evoked potentials were examined before and after the traumatic injury. The rabbits′ eyes were removed, the optic nerves were pathologically examined, and the content of TXB 2 and 6 Keto PGF 1? which were the products of TXA 2 and PGI 2 were assayed 1, 2, 4, and 7 days after traumatic injury respectively. Results Histopathological examination revealed the findings of injuries of optic nerves of all the 24 rabbits. The latency of wave P 1 was significantly delayed after traumatic injury ( P
4.Perioperative management and impact of preoperative renal dysfunction on short-term survival for patients undergoing valve replacement

Objective To review the experience of perioperative management and effect of valve replacement combined with renal dysfunction.Methods 536 cases of valve replacement.According to preoperative glomerular filtration rate(GFR),renal dysfunction was classified as normal in 322 cases,mild in 162,moderate in 40 and severe in 12.Logistic regression analysis was used to assess the effect of preoperative renal dysfunction on operative mortality and morbidity.Results Patients with a lower GFR were older and more likely to have hypertention.They also tended to have larger left ventricular end-diastolic diameter and worse left ventricular ejection factor.Operative mortality rose inversely with declining renal function,from 2% for those with normal renal function to 17% for patients with severe renal dysfunction.Renal dysfunction was significantly associated with a large blood transfusions,re-intubation,longer mechanical ventilation,intensive care unit stay and hospital stay.Conclusion Preoperative renal dysfunction is an important factor of postoperative mortality and morbidity for valve replacement.We must pay more attention to perioperative management.
5.A case report of combined liver pancreas transplantation

Objective To probe the indication, technique and effect of combined liver pancreas transplantation. Methods Donor organs were harvested by the technique of total abdominal evisceration. Simultaneous orthotopic liver and heterotopic pancreas duodenum transplantation was performed in one patient diagnosed as having chronic hepatitis B, hepatocirrhosis, hepatic cellular cancer, and insulin dependent diabetes. Liver and pancreas graft function was monitored after transplantation. Results The recipient recovered with excellent allograft function, and insulin independence was achieved without any surgical complication. Conclusion End stage liver disease with concomitant insulin dependent diabetes is the indication for combined liver pancreas transplantation.
6.The expression of chondroitin sulfate proteoglycans after focal cerebral infarction in hypertensive rats

Objective To examine the expression of inhibitory chondroitin sulfate proteoglycans (CSPGs) and in-vestigate their potential effects on neural plasticity in the peri-infarct cortex and ipsilateral thalamus after focal cerebral infarction in hypertensive rats. Methods Twenty-four adult renovascular hypertensive Sprague-Dawley rats per group were subjected to permanent right middle cerebral artery occlusion(MCAO) or sham operation. Twelve rats which were se-lected randomly from per group at each time point were decapitated and their brains were removed and cut into coronal sections at 7 and 14 days post MCAO. The expression of CSPGs, NG2 and Neurocan was examined using immunostaining and western blot. Results All rats displayed neurological deficits to varying degrees and the expression of CSPGs, NG2 and full length Neurocan was increased in the peri-infarct cortex and ipsilateral thalamus at 7 and 14 days (P<0.05). However, there were no significant difference in either expression of CSPGs, NG2 and full-length Neurocan between 7 and 14 days or the expression of C-terminal fragment Neurocan at 7 and 14 days (all P>0.05). Conclusions CSPGs may play a negative role in neural plasticity through induction of inhibitory environment in the peri-infarct cortex and ipsilat-eral thalamus following focal cerebral infarction in hypertensive rats.
7.Prostate cancer with homologous recombination repair gene mutations and PARP inhibitors: clinical progress

DNA damage repair gene mutations are prevalent in advanced prostate cancer. Among these, mutations in homologous recombination repair genes could impair the ability of cell to restore the DNA double-strand break, which can be exploited by Poly-ADP-ribose polymerase (PARP) inhibitors through synthetic lethality and result in cell death. The phase Ⅲ study " PROfound" showed that the PAPR inhibitor Olaparib could significantly improve the survival of patients with homologous recombination repair gene mutations compared with novel hormone agents, starting the era of targeted, precise and individualized treatment based on genetic profile detection for prostate cancer treatment.
9.Expression and suppressive function of CD39+ regulatory T cells in kidney transplant recipients

Objective To investigate expression and suppressive function of CD39 + regulatory T cells (Treg) in kidney transplant recipients.Method Thirty recipients of first kidney transplants were treated with tacrolimus,mycophenolate mofetil and prednisone.Within 28 days posttransplantation,there were 14 patients subject to acute rejection (AR group),and the rest 16 patients had no episodes of acute rejections (NR group).Twelve healthy volunteers served as healthy controls (HC group).We collected peripheral blood from the three groups and separated PBMC by density gradient centrifugation,and sorted Tresp,CD39-Treg and CD39+ Treg by flow cytometry.We next analyzed the ratio of CD39 + Treg/CD4+ T cells.ELISA was used to determine the suppressive ability of CD39-Treg and CD39+ Treg on secretion of IFN-γ and IL-17 by Tresp.Results The ratio of CD39 + Treg/CD4 + T cells in AR group was significantly reduced as compared with HC group and NR group (P<0.05).In HC group and NR group,the secretion of IFN-γ and IL-17 by Tresp was suppressed significantly (P<0.05) by CD39+ Treg.CD39Treg could suppress secretion of IFN-γ but not IL-17 production by Tresp.CD39+ Treg in AR group AR could suppress the secretion of IFN-γ significantly (P<0.01),but not to IL-17 production.Conclusion CD39+ Treg have important immunoregulation function.The relative amount of CD39+ Treg was reduced and their regulatory function was impaired in patients with acute rejection.
10.Determination of nitrite additives in marketed milk powder

Objective To detect and analyse the levels of nitrite in marketed milk powder.Methods 6 brands of maketed milk powder were selected in this study.Interferents,such as protein,were removed from milk powder preliminarily by using potassium ferrocynide and zinc acetate.The levels of nitrite were detected by using fluorospectrophotometry method,and compared with na-tional standard(2 mg/kg).Results The levels of nitrite in the 6 brands of maketed milk powder were lower than the national stand-ard limit,had statistically significant differences(P <0.05).Conclusion The levels of nitrite of 6 brands of milk powders do not ex-ceed the national standard.
