1.Study on present situation and countermeasures of Chongqing Traditional Chinese Medicine

Objective To investigate the status of TCM development in Chongqing city,and to put forward countermeasures and suggestions of the development of Chinese medicine in view of the existing problems,so as to provide reference for the government and relevant departments.Methods The relevant questionnaires were issued and recycled in 20 districts and counties of Chongqing city.Part of the data culled from Chinese management office yearbooks of Chongqing.Finally,comprehensive analysis was per-formed.Results There′s enormous upside potential for the development of Chongqing traditional Chinese medicine (TCM),but there were some problems and deficiencies.The main problems were as follows:the TCM characteristics not brought into full play in traditional Chinese Medicine hospitals,lack of following talents,unsound organization management of traditional Chinese medi-cine,the government′s insufficient investment and attention for traditional Chinese medicine (hospitals,schools,“hand-to-hand”ap-proach),not enough national policy support for TCM,TCM hospitals and folk medicines of ethnic minorities,and insufficient exca-vation in ethnic minority medicine.Conclusion In order to adapt to the development of Chinese medicine and to meet the growing requirements of Chinese medicine,some countermeasures should be taken as follows:giving full play to the characteristics of tradi-tional Chinese medicine,strengthening personnel training in Chinese medicine,reforming the management mechanism,increasing the government investment,strengthening policy support for Chinese medicine and folk medicines of ethnic minorities,accelerating the process of TCM standardization,scientific and internationalization.It is suggested that the miscellaneous diseases &incurable dis-ease medicine research office and slow disease traditional Chinese medicine health care office should be established under the TCM management office.Specialized agencies are advised to mine and collect herbal remedies,recipes in ethnic minority area and so on.
3.Effects of different concentrations of propofol on apoptosis in hippocampal neurons in neonatal rats in vitro

Objective To investigate the effects of different concentrations of propofol on the apoptosis in hippocampal neurons in neonatal rats in vitro.Methods Primary hippocampal neurons were prepared from newborn Sprague-Dawley rats (aged 7 days) and cultured for 7 days.The neurons were randomly divided into 7 groups (n =8 each):control group (group C),propofol 4,8 and 12 μg/ml groups (groups P1-3),and fat emulsion 4,8and 12 μg/ml groups (groups F1-3).The cells were cultured for 24 h in the culture medium containing propofol 4,8 and 12μg/ml in groups P1-3,respectively.The cells were cultured for 24 h in the culture medium containing fat emulsion 4,8 and 12 μg/ml in groups F1 3,respectively.The cell morphology was examined by microscopy after 24 h culture.The expression of caspase-3 (by immuno-histochemistry) and neuronal apoptosis were detected.The neuronal apoptosis rate was calculated.Results Compared with group C,the neuronal apoptosis rate and caspase3 expression were significantly increased in groups P1-3 (P < 0.05 or 0.01).The neuronal apoptosis rat and caspase-3 expression were increased in a concentration-dependent manner in groups P1-3 (P < 0.05).The neuronal apoptosis rate and caspase-3 expression were significantly lower in groups F1-3 than in groups P1-3 (P < 0.05).There was no significant difference in the neuronal apoptosis rate and caspase-3 expression between groups F1-3 (P > 0.05).The damage to neurons was induced in groups P1-3 and most severe in group P3.Conclusion Propofol can promote the apoptosis in hippocampal neurons in neonatal rats in vitro in a concentration-dependent manner.
4.The preliminary study on commitment differentiation of embryonic stem cells induced by the medium of cultured retinal neurons of SD rats

Objective To investigate the possibility of commitment differentiation of embryonic stem cells induced by the medium of cultured retinal neurons of SD rats. Methods The medium from cultured retinal neurons of SD rats were collected, sterilized and mixed with DMEM medium according to 2∶3 proportion, ES cells were cultured with these mixed medium and were observed under the phase contrast microscope daily, the induced cells were identified by NF immunohistochemistry methods. Results The ES cells cultured with these mixed medium can differentiate into neuron like structure, and the induced cells were positive in NF immunofluorescence staining. Conclusion The medium from cultured retinal neurons of SD rats can induce ES cells commitment differentiation into neuron like structure.
5.Robot-assisted laparoscopic radical prostatectomy for prostate cancer: report of 34 cases.

OBJECTIVETo assess the feasibility and safety of robot-assisted laparoscopic radical prostatectomy (RLRP) in the treatment of prostate cancer.
METHODSUsing the da Vinci robot surgical system, we performed RLRP for 34 patients with localized prostate cancer and analyzed the intraoperative and follow-up data.
RESULTSThe procedures were performed successfully in all the patients, with the mean operation time of 198 min (range 135-340 min), average blood loss of 257 ml (range 50-700 ml), and 1 case of blood transfusion, but no postoperative complications. Three cases had positive surgical margins. Postoperative examination at 4 weeks showed PSA > 0.2 microg/L in 2 cases, suggestive of residual tumor, for which maximal androgen block therapy was administered. The other 32 patients were followed up for 3-10 (mean 7.5) months, during which the average level of serum tPSA remained < 0.2 microg/L. Urinary continence was found in 94% (32/34) and 97% (33/34) of the patients at 3 and 6 months, respectively, of whom 77% (26/34) and 88% (30/34) had no urinary leakage (0 pad per day).
CONCLUSIONRLRP, with its advantages of less perioperative blood loss, low rate of positive margin, and good urinary continence, is a safe and effective surgical option for the treatment of prostate cancer.

6.Intervention Mechanism of Extracts from Radix Ginseng, Radix Notoginseng and Rhizoma Chuanxiong on Adventitia of Senescent Rats.

OBJECTIVETo observe the reconstruction features of adventitia in senescent rats, and to explore the intervention mechanism of Chinese herbs (CH, extracts from Radix Ginseng, Radix Notoginseng, and Rhizoma Chuanxiong).
METHODSTotally 85 20-month senescent rats were randomly divided into 5 groups according to body weight, i.e., the aging model group, the high dose CH group, the middle dose CH group, the low dose CH group, the Losartan group, 17 in each group. Another 14 2-month old Wistar rats were selected as a young group. Extracts of CH at the daily dose of 1493. 4, 746. 7, and 373. 4 mg/kg were administered to rats in the 3 CH groups respectively by gastrogavage. Losartan suspension at the daily dose of 10 mg/kg was administered to rats in the Losartan group by gastrogavage. Equal volume of distilled water was administered to rats in the aging model group and the young group. All medication was performed once daily. After 15-week intervention, morphological changes of thoracic aorta were observed by HE staining. The types, distribution, and contents of vessel wall collagens were determined using picric acid picrosirius red staining. The plasma renin activity (PRA) , the concentration of rennin angiotensin II (Ang II), and the content of Ang II in adventitia were detected by radioimmunoassay. The content of hydroxyproline ( Hyp) was detected by biochemical analysis. mRNA contents and protein expressions of angiotensin II receptor 1 (AT1R) and angiotensin II receptor 2 (AT2R) were detected by real-time PCR (RT-PCR) and Western blot.
RESULTSCompared with the young group, thickened adventitia, increased adventitia thickness/caliber, accumulated collagen fiber, increased area of type I collagen, decreased area of type III collagen, decreased type III/I collagen area ratio (P <0. 05), decreased plasma PRA and Ang II (P < 0.01, P < 0.05), increased contents of Ang II and Hyp in adventitia, down-regulated mRNA and protein expressions of AT1R, and up-regulated mRNA and protein expression of AT2R could be seen in the aging model group (P < 0.05). Compared with the aging model group, morphological changes could be improved in the 3 CH groups. Adventitia thickness/caliber was reduced in middle and high dose CH groups, as well as the Losartan group. The area of type I collagen was reduced and the area of type III collagen was enlarged, type III/I collagen area ratio obviously increased, contents of adventitia Hyp was obviously lowered in the high dose CH groups and the Losartan group (P < 0.05, P < 0.01). Ang II levels in adventitia decreased in middle and high dose CH groups and the Losartan group (P < 0.05, P < 0.01). There was no statistical difference in PAR among all groups (P > 0.05). Compared with the aging model group, mRNA expression of AT1R all increased in each treatment group (P < 0.01); mRNA expression of AT2R also increased in middle and high dose CH groups (P < 0.05). Protein expression of AT1R increased in the high dose CH group and the Losartan group (P < 0.01, P < 0.05); protein expression of AT2R also increased in middle and high dose CH groups (P < 0.05).
CONCLUSIONSAdventitia remodeling occurred in aged rats, manifested as thickened adventitia and accumulated collagens, disordered ratios of collagen I and III. Its mechanism might be possibly associated with aactivation of renin-angiotensin system (RAS). Extracts from Radix Ginseng, Radix Notoginseng, and Rhizoma Chuanxiong could improve adventitial remodeling possibly by interfering multi-targets, such as Ang II and AT1R, thereby delaying vascular aging.
