1.Immune Responses of Th1/Th2 in Children with Mycoplasma Pneumoniae Pneumonia

Objective To study the immune responses of Th1 and Th2 subsets of T cells in 26 children with mycoplasma pneumo-niae pneumonia(MPP).Methods ELISA was used to detect the levels of interferon- ?( IFN- ?) and interleukin-4(IL-4) in the serum in 12 healthy children as normal controls and 26 patients of acute stage as acute stage MPP group, 9 of whom in recovery stage were as recovery stage MPP group. Results IFN - ? level in acute stage MPP group was significant higher than that in normal controls (P 0.05); and IFN - 7/IL - 4 ratio was significant higher than that in normal controls( P0.05).Conclusion Thl responses increase and Th2 responses decrease in children with MPP,and this kind of response persists during the recovery stage.

OBJECTIVETo investigate the mechanism of action of Chinese drug Langchuang-3 Granule (LC-3) in treating systemic lupus erythematosus (SLE) through observing its effects on body weight (BW), antinuclear antibody (ANA), spleen index and thymus index in BXSB SLE model mice, as well as the deposition of immune complex in mice' renal glomeruli.
METHODSSLE model mice were randomized into 4 groups: the control group (A), the LC-3 treated group (B), the Western drug treated group (C), and the LC-3 combined Western drug treated group (D). BW of mice was dynamically observed; spleen index (SI) and thymus index (TI) were measured by weighting method; serum anti-ANA level was detected by ELISA; IgG and complement C3 in renal glomeruli were analysed by direct immunofluorescence.
RESULTSBW of mice remarkably increased after treatment in Group B and D, showing a significant difference to that in Group A (P < 0.05). Lower TI level and higher SI level were found in the model mice. As compared with Group A, TI was higher in Group B and D, SI was lower in Group D, and both indices were lower in Group C (P < 0.05 or P < 0.01). Level of ANA decreased (converting from positive to negative) remarkably in all the three treated groups (P < 0.01). The fluorescence intensity of IgG and C3 in the renal tissue was weaker in Group B, C (both P < 0.05) and D (P < 0.01) than that in Group A.
CONCLUSIONBoth LC-3 and Western medicine showed remarkable effects in treating SLE model mice, and the combination of the two could display an effect better than that of using either alone.

4.Controllable Growth of Nanoporous Metal Oxide Composites on Nickel-Titanium Alloy Fibers for Selective Solid-Phase Microextraction of Polycyclic Aromatic Hydrocarbons

Hydrothermal oxidation was used to prepare a nickel-titanium alloy ( NiTi ) solid-phase microextraction fiber. The experimental results demonstrated that a nanoporous NiTi oxide composite coating was in situ grown on the surface of NiTi substrate by direct oxidation in aqueous H2 O2 solution at 80℃. The resulting composite oxide coating included more nickel and less titanium. The prepared NiTi fiber with Ni-rich oxide coating was used to extract typical aromatic compounds coupled with HPLC-UV and exhibited good extraction selectivity for polycyclic aromatic hydrocarbons ( PAHs). The key factors affecting extraction efficiency of PAHs were examined. Under the optimized conditions, the calibration curves were linear in the range from 0. 05 to 500 ng / mL with correlation coefficients ≥0. 999, and the limits of detection were 0. 026-0. 056 ng / mL. Furthermore, the relative standard deviations ( RSDs) for intra-day and inter-day repeatability of the single fiber varied from 4. 8% to 6. 2% and from 5. 4% to 6. 5% for five replicates of PAHs at the spiking level of 50 ng / mL, respectively. The RSDs for the fiber-to-fiber reproducibility of five fibers prepared in different batches ranged from 6. 4% to 8. 4% . This method was suitable for selective enrichment and detection of target PAHs in environmental water samples with relative recoveries of 87. 4% -108. 2% and RSDs <8. 1% . Moreover, this novel NiTi fiber was mechanically strong and chemically stable, and its preparation was precisely controllable.
5.The Prevalence of NIDDM and IGT and Related Factors Among Residents in Some Areas of Hubei Province, China

The epidemiological survey of prevalence of NIDDM (non-insulin dependent diabetes mellitus) and IGT(impaired glucose tolerance)was conducted among 9450 residents aged 25~70 in some areas of Hubei Province, China. The results show that NIDDM and IGT prevalences are 2.62% and 4.48%, respectively. There is no significant difference between male and female (P>0.05). The NIDDM prevalence in cities is slightly higher than that in countryside, but the difference is not significant (P>0.05). However, the IGT prevalence in city is significantly higher than that in countryside (P<0.01). The prevalence of both NIDDM and IGT is increasing along with the age of the population. It is also significantly related to the family history of NIDDM, hypertension, and high body mass index (BMI). By using stepwise logistic regression to analyse the risk factors of NIDDM, age (OR=1.86),BMI(OR=2.69), family history (OR=2.84) and hypertension (OR=2.23) entered the model (significance level is α=0.05).

7.Early total care pattern for intertrochanteric fracture of femur in the elderly.

OBJECTIVETo evaluate clinical results of early total care (ETC) treatment for elderly patients with intertrochanteric femur fractures.
METHODSClinical data of 106 elderly patients with intertrochanteric fracture treated from January 2012 and February 2015 were retrospectively studied. According to whether receiving the early total care mode, the patients were divided into 2 groups, 34 cases were diagnosed and treated with early total care pattern (ETC group), including 14 males and 20 females with an average age of (74.88 ± 4.38) years old ranging from 70 to 86. According to Evans types, 4 cases were type I, 5 cases were type II, 13 cases were type III, 11 cases were type IV, 1 case was type V. Seventy-two patients were treated with conventional trauma method (conventional group), including 35 males and 37 females with an average age of (74.46 ± 3.63) years old ranging from 70 to 85. According to Evans type, 8 cases were type I ,13 cases were type II, 25 cases were type III, 25 cases were type IV, and 1 case was type V. All fractures were treated with proximal femoral nails anti-rotation (PFNA). Operative time, hospital stays, leaving bed time, complications, cases of death at 1 year after operation, postoperative Harris score at 12 months were observed and compared.
RESULTSAll patients were followed up, the time of ETC group ranged from 9 to 18 months with an average of 13.29 ± 1.51, and the time in conventional group ranged from 12 to 16 months with an average 12.93 ± 1.15, while there was no significant difference between two groups in time of following-up (t = 1.368, P = 0.174). There was no significant meaning in cases of death between ETC group (2 cases) and conventional group (8 cases). Three cases occurred complications in ETC group, and 20 cases in conventional group,there was obvious meaning between two groups (χ² = 0.739, P = 0.318). Operative time,hospital stays,leaving bed time in ETC group respectively was (2.03 ± 0.67) d, (15.41 ± 2.87) d and (3.62 ± 0.74) d; while in conventional group respectively was (4.17 ± 1.59) d, (20.11 ± 4.24) d and (5.35 ± 1.22) d; there were significant differences between two groups in operative time, hospital stays, leaving bed time. Postoperative Harris scores at 12 months in ETC group was (82.32 ± 4.56), and (79.24 ± 5.52) in conventional group, there was obvious meaning between two groups (t = 2.833, P = 0.006).
CONCLUSIONETC pattern is a novel method for diagnosis and treatment of intertrochanteric femur fractures in elderly, it could shorten operative time, hospital stays, leaving bed time, decrease complications and promote recovery of function.

8.Peripheral radial chop technique for phacoemulsification of hard cataracts.

BACKGROUNDPhacoemulsification yields successful outcomes in eyes with standard cataract. Though techniques have been improved, it is still challenging to perform phacoemulsification in cases of hard cataracts for difficulty in nuclear management and much more complications. This study aimed at describing and evaluating the efficacy and safety of a peripheral radial chop technique to remove hard cataracts.
METHODSIn this prospective study conducted between January 2003 and January 2004, 107 consecutive eyes with hard cataract underwent modified phacoemulsification surgery with peripheral radial chop technique by the Bausch & Lomb Millennium phacoemulsifier with preset parameters of power less than 30%; vaccum, 150 mmHg; and bottle height, 85 cm when a DP8145 phaco tip was used, and vaccum, 380 mmHg; bottle height, 95 cm when a DP8245 phaco tip was used.
RESULTSThe mean ultrasonic power was 14.7% (range 9% to 19%), ultrasonic time was 1.98 minutes (range 1.55 to 3.18 minutes). At 1, 7 and 30 days postoperatively, the eyes with uncorrected visual acuity 0.5 or better accounted for 76.42%, 87.16% and 90.67% respectively. At 1 month, the endothelial cell loss rate was 9.74% (range 8% to 17%). There were 6 cases of posterior capsule rupture in an early period of study. No serious intraoperative or postoperative complications were noted.
CONCLUSIONSThe peripheral radial chop technique was effective without serious complications in hands of an experienced surgeon.

9.Expression of PTEN protein and its correlation with p27kip1 and cyclin D1 expression in primary breast cancer.

OBJECTIVETo study the expression of phosphatase and tensin homology deleted on chromosometen ten (PTEN) protein, a tumor suppressor gene in breast cancer and its correlation with p27(kip1) and cyclin D1 expression.
METHODSPTEN protein expression, p27(kip1) and cyclin D1 protein expression were detected by immunohistochemical method in paraffin sections from 61 women with primary breast cancer. PTEN protein expression was compared with clinico-pathologic parameters as related to p27(kip1) and cyclin D1.
RESULTSPTEN, being shown in the cytoplasm, was negative in 6.6% (4/61), reduced in 41.0% (25/61) and positive in 52.5% (32/61) samples. PTEN expression level was correlated with axillary lymph node status, loss of estrogen receptor stain, recurrence and metastasis. On univariate analysis, the disease-free survival rate of patients with higher PTEN expression (> 50% cells stained) was better than those with lower expression (P = 0.0101). However, there was no correlation between p27(kip1), cyclin D1 expression or PTEN expression.
CONCLUSIONPTEN, its lower expression being correlated with poor outcome of breast cancer patients, plays a prominent role in breast cancer. p27(kip1) or cyclin D1 may not be the primary downstream genes of PTEN in breast cancer.

10.Effects of shuangshen ningxin capsule on mini-porcine myocardial ischemia model induced by thrombosis via cardiac catheterization intervention.

OBJECTIVETo investigate the establishment of mini-porcine myocardial ischemia (MI) model induced by thrombosis via cardiac catheterization intervention, and observe the anti-MI action of Shuangshen Ningxin Capsule (SNC).
METHODSMI model of Chinese mini-porcine was established by injection of self-thrombus in left anterior descending coronary artery (LAD) with guiding catheter through carotid artery. The effect of SNC on MI was evaluated comprehensively by analyzing the parameters as coronary embolism, 30 dots surface mapping electrocardiogram and quantitative histology, etc.
RESULTSLAD in the model animals was basically embolized 6 days after modeling, showing increase of amplitude and dot of ST segment elevation. After being administered with SNC for 6 days, the self-thrombus blocked LAD was recanalized and the degree and extent of MI reduced.
CONCLUSIONIt is the first time to successfully establish MI model of Chinese mini-procine by thrombosis via cardiac catheterization intervention. SNC has anti-MI effect.
