1.IGF-Ⅱ inhibits adriamycin-induced apoptosis of HepG2

Objective To investigate the effects of IGF-Ⅱ on adriamycin-induced apoptosis of hepatocellular carcinoma cell line(HepG2).Methods Cultured HepG2 cells were divided into:①group A:control group;②group B:ADM group(200 ng/ml);③group C:2 ng/ml IGF-Ⅱ+200 ns/ml ADM group;④group D:20 ng/ml IGF-Ⅱ+200 ng/ml ADM group;⑤group E:200 ng/ml IGF-Ⅱ+200 ng/ml ADM group. After HepG2 were treated for 48 h,MTT colorimetry was performed to determine the cell viability,Flow cytometry was used to detect the cell apoptosis rate,and Weastern-blot was performed to evaluate the expression of survivin. Results The cell viability of group A、B、C、D、E was respectively 0.568±0.025、0.201±0.020、0.232±0.027、0.268±0.013 and 0.304±0.019;The cell apoptosis rate of group A、B、C、D;E was respectively 6.9%±1.3%、35.4%±2.1%、31.2%±2.2%、26.4%±1.7%and 21.7%±1.9%;Survivinβ-actin of group A、B、C、D、E was respectively 0.527±0.039、0.147±0.081、0.311±0.069、0.421±0.033 and 0.469±0.031.The cell viability in IGF-Ⅱ+ADM group was better than ADM group(P<0.01),the cell apoptosis rate in IGF-Ⅱ+ADM group was significantly lower than ADM group(P<0.01),the expression level of survivin in IGF-Ⅱ +ADM group was significantly higher than ADM group(P<0.01). Conclusions IGF-Ⅱ inhibits ADM-induced apoptosis of HepG2 cells,probably by up-regulating cell's survivin expression.
2.The interventional effect of anti-basic fibroblast growth factor on the model of bleomycin-induced pulmonary fibrosis in rats.

OBJECTIVETo study the effect of anti-basic jibroblast grouth factor (bFGF) on bleomycin-induced pulmonary fibrosis in rats and its possible mechanism.
METHODSPulmonary fibrosis was induced in Wistar rats by intratracheal instillation of bleomycin (model group, group M). Then the rats received anti-bFGF on 1, 2, 3, 8, 12, 19, 25 days intracavitary injection (group K), normal saline(group C) orally. Normal controls received normal saline both intratracheally and orally. Five rats in each group were sacrificed on 1.4 week after intratracheal instillation. Histological changes of the lungs were evaluated by HE stain and Massons trichrome stain. Lung expression of bFGF proteins was assessed by immunohistochemistry and the level of bFGF protein in serum and BALF was further measured by ELISA.
RESULTSPulmonary fibrosis of group M was higher than that of group C. bFGF in group M was higher than that in group C in lung, serum and BALF on 1.4 week. Pulmonary fibrosis of group K was lower than that of group M. bFGF in group K was lower than that in group M in lung, serum and BALF on 1.4 week.
CONCLUSIONAnti-bFGF alleviates bleomycin-induced pulmonary fibrosis in rats. Inhibiting the expressions of bFGF in lung tissues may be one of the mechanisms.

3.Endobronchial Ultrasound-guided Transbronchial Needle Aspiration Increases the Yield of Transbronchial Lung Biopsy for the Evaluation of Peribronchial Lesions

Background:Due to absence of visible endobronchial target,the diagnostic yield of flexible bronchoscopy for peribronchial lesions has been unsatisfactory.Convex probe endobronchial ultrasound (CP-EBUS) has allowed for performing real-time transbronchial needle aspiration (TBNA) of enlarged hilar and mediastinal lymph nodes and therefore could also be used as a means of diagnosing proximal peribronchial lesions.Methods:We retrospectively analyzed the results related to 72 patients who underwent CP-EBUS for peribronchial lesions without endobronchial involvement and adjacent to three-grade bronchi based on chest computed tomography (CT) scan.We recorded the images during EBUS as well as the diagnostic results of TBNA and conventional-transbronchial lung biopsy/brush (C-TBLB/b),and final diagnoses were based on pathologic analysis and follow-up.Results:In all cases,the mass was able to be identified using EBUS in 97.2% patients (70/72) who were performed with EBUS-TBNA + C-TBLB/b.Sixty-six patients had a final diagnosis,80.0% patients (56/70) had malignancies,and 14.3% patients (10/70) had benign disease.In malignancies,the diagnostic yield of C-TBLB/b was 57.1% (32/56) and in EBUS-TBNA was 85.7% (48/56),whereas pathologic diagnosis reached 94.6% when EBUS-TBNA was combined with C-TBLB/b.C-TBLB/b + EBUS-TBNA also exhibited stronger potency ofhistolytic diagnosis for malignancies than either EBUS-TBNA or C-TBLB/b alone.Furthermore,there are data supporting the value of EBUS-TBNA for the diagnosis of benign lung disease.Conclusion:The combined endoscopic approach with EBUS-TBNA and C-TBLB/b is an accurate and effective method for the evaluation of peribronchial lesions,with better results than using each technique alone.
4.Inhalation of glucocorticosteroid in the rat model of bleomycin-induced pulmonary fibrosis and its impact on the expression of basic fibroblast growth factor.

OBJECTIVETo study the effect of inhalation of glucocorticoid on bleomycin-induced pulmonary fibrosis in rats and its possible mechanism.
METHODSPulmonary fibrosis was induced in Wistar rats by intratracheal instillation of bleomycin (group M, group B, group D). Group B inhaled glucocorticoid daily from the next day of received bleomycin. Group D intraperitoneal injection glucocorticoid daily from the next day of received bleomycin. Normal controls received normal saline both intratracheally. Five rats in each group were killed at 1, 4 week after intratracheal instillation. Histological changes of the lungs were evaluated by HE, Masson trichrome stain. Lung expression of bFGF proteins was assessed by immunohistochemistry and the level of bFGF protein in serum and BALF was further measured by ELISA.
RESULTSPulmonary fibrosis of group M was higher than that of group C, pulmonary fibrosis of group B, D was lower than that of group M at 1, 4 week. bFGF in group M was higher than that in group C, bFGF in group B, D was lower than that in group M in lung, serum and BALF on 1, 4 week.
CONCLUSIONInhalation of glucocorticosteroid alleviates bleomycin-induced pulmonary fibrosis in rats. The mechanism may be related to the changes that bFGF is degrade or prevent it step up.

5.Professor YANG Zhao-min's contribution to the education of modern acupuncture and moxibustion.

It was completely collected and summarized the teaching history and the relevant historical materials of Professor YANG Zhao-min via consulting the literatures, investigation, interview, etc. in this paper. The summaries indicated that from the beginning of running a school, Professor YANG Zhao-min adhered to the CHENG's moral, worked hard and successfully explored and practiced the training pattern of teaching tour for the personnel training of acupuncture and moxibustion. He established the specialty of acupuncture and moxibustion, formatted courses and inherited the feature of practice teaching. He was in charge of the compilation of textbook and engaged in scientific innovation. He inherited the past and forged ahead into the future and had made the decision to cultivate the personnel of acupuncture and moxibustion. Professor YANG Zhao-min had practiced and developed the modern educational thought from the CHENG Dan-an and had made the important contribution to the training of acupuncture and moxibustion professionals.

6.SOX40L: an important inflammatory mediator in adult bronchial asthma.

INTRODUCTIONThe role of soluble OX40 ligand (sOX40L) in adult bronchial asthma is unclear. This study aims to determine the serum concentrations of sOX40L in adult patients with bronchial asthma, and discussed its relationship with pulmonary function.
MATERIALS AND METHODSWe measured the pulmonary function using the spirometer and detected the serum concentrations of sOX40L by enzyme linked immunosorbent assay (ELISA) in 19 healthy persons in the control group, 58 acute asthmatic adult patients who were grouped according to their disease severity: 18 mild grade, 24 moderate grade, 16 severe grade, and 24 persons in a stable asthmatic group.
RESULTSThe serum concentrations of sOX40L in asthmatic adult patients (6.80 ± 4.95 ng/L) were distinctly higher than those in the control group (3.98 ± 2.83 ng/L, P <0.05), and they were negatively correlated with pulmonary function indexes (FEV1%, FVC%, FEV1/FVC) (r = -0.754, P <0.01, r = -0.557, P <0.01, r = -0.457, P <0.01, respectively). Moreover, the serum concentrations of sOX40L showed obvious differences among control, mild, moderate, and severe groups (3.98 ± 2.83, 4.87 ± 1.89, 6.97 ± 5.91, 8.71 ± 5.18 ng/L, respectively; P <0.01). The concentrations of sOX40L decreased to the same extent as the control group after therapeutic treatments were provided to the asthmatic adult patients.
CONCLUSIONThe concentrations of sOX40L were found to be high in adult asthmatic patients and were associated with the severity of the disease. Therefore, sOX40L could be a potential inflammatory mediator in the pathogenesis of asthma.

7.Efficacy of S-1 combined with cisplatin for advanced gastric carcinoma

Objective To evaluate the short-term efficacy and toxicity of the combination of gimeracil and oteracil porassium ( S-1 ) and cispla-tin in the treatment of advanced gastric carcinoma.Methods Fifty-six patients with advanced gastric carcinoma were randomly divided into two groups with 28 cases in each group.Experimental group was treated with S-1 (60 mg bid d1 -d21) combined with cisplatin (60 mg· m-2 iv d1-d3) with 4 weeks as a cycle.While control group was treated with oxaliplatin (130 mg· m-2 iv d1), tegafur (600 mg· m-2 iv d1-d5), calcium folinate ( 120 mg · m-2 iv d1 -d5 ) with 3 weeks as a cycle.Short-term efficacy and adverse reactions were evaluated after three cy-cles.Results The tumor response rate of experimental group and control group was 75.0%and 42.9%(P<0.05).Besides, compared with the con-trol group , the occurrence of gradeⅢ-Ⅳadverse reactions in experimental group were apparently lower.Conclusion The regimen of S -1 combined with cisplatin has shown a better efficacy in the treatment of patients with ad-vanced gastric carcinoma , and the toxicity is relatively mild and tolerable .
8.Clinical observation on acupotomy for treatment of simple obesity.

OBJECTIVETo compare the efficacy differences among acupotomy, electroacupuncture and acupuncture for treatment of simple obesity.
METHODSOne hundred and five cases were randomly divided into an acupotomy group, an electroacupuncture group and an acupuncture group, 35 cases in each group, and Zhongwan (CV 12), Tianshu (ST 25), Shangjuxu (ST 37), Sanyinjiao (SP 6), etc, were selected in three groups and also with selection of acupoints according to symptoms. The acupotomy group was treated with acupotomy 40 mm in length and 0.6 mm in diameter, the electroacupuncture group with the Han's LH402A electroacupuncture stimulator and the acupuncture group with simple acupuncture treatment. The clinical therapeutic effects of three groups were compared. The obesity signs such as body weight, body mass index (BMI), obesity degree, etc., and blood lipid and fasting blood sugar (FBS) were observed.
RESULTSThe markedly effective rate of 91.4% (32/35) in the acupotomy group was higher than that of 71.5% (25/35) in the electroacupuncture group and that of 42.9% (15/35) in the hand acupuncture group (both P<0.05). There were significant differences in the obesity signs and blood fat and FBS of the three groups before and after treatment (P<0.05, P<0.01). The differences rates of body weight, BMI, obesity degree, chest circumference, waistline, thighline, waist-hip ratio, total cholesterol before and after treatment in the acupotomy group were all better than those in the electroacupuncture group and in the acupuncture group (all P<0.05), and the differences rates of hipline, FBS in the acupotomy group were better than those in the acupuncture group (both P<0.001).
CONCLUSIONAcupotomy treatment can obviously reduce fat, FBS and blood lipid and has obvious therapeutic effects on simple obesity.

9.Application of low tension Roux-y sigmoid neobladder in urine diversion.

OBJECTIVETo assess the therapeutic effects of low tension, anti-reflux Roux-y sigmoid neobladder.
METHODSA total of 21 patients (7 male and 14 female) were included, aged 43-87 years. All cases received radical cystectomy and low tension Roux-y sigmoid neobladder procedure for invasive bladder cancer were included in this study. The period of follow-up was from 8 to 79 months (the average was 36 months). Evaluations included urinary flow rate, post voiding residual and filling cystometry.
RESULTSThe mean maximum urinary flow rate, the voiding time and the post voiding residual were 28.1 ml/s (21.4-38.4 ml/s), 17 s(9-28 s) and 0 ml respectively. The cystometric capacity was 480 m1 (350-560 ml). The volume of desire to void was 330 ml (120-410 ml). The bladder pressure was from 14.2 to 18.6 cm H2O (the average bladder pressure was 16.4 cm H2O) at high filling volumes. The maximum voiding pressure was 45.0 cm H2O (23.6-63.4 cm H2O).
CONCLUSIONSThe Roux-y sigmoid neobladder has an adequate capacity at low pressure with a satisfactory continence, and it is an effective method for continent urinary diversion.

10.Analysis of transmission dynamics and effectiveness of control of local epidemics caused by the Omicron BA.2 and BA.5.2 COVID-19 strains in Fujian Province

This study evaluated the scientific nature and effectiveness of iterative optimization of prevention and control measures for local outbreaks caused by the BA.2 and BA.5.2 COVID-19 strains in Fujian Province in 2022,to provide a scientif-ic basis for responding to future new or recurrent respiratory infectious diseases.According to the theory of infectious disease dynamics,relevant information regarding the local epidemic situation caused by the BA.2 sub-type Omicron virus strain in March 2022 and BA.5.2 sub-type Omicron virus strain in October 2022 in Fujian Province was collected.The susceptible exposed infectious removed(SEIAR)model of COVID-19 infection with a latent period and asymptomatic infected persons was used to analyze the transmission dynam-ics of two local epidemic situations,and evaluate the preven-tion and control effects.The incubation period of the BA.2 epidemic was 3 days(1~9 days),the intergenerational inter-val was 3 days(1~5 days),and the initial Rt was 3.0(95%CI:2.7~3.3).The incubation period of the BA.5.2 epidemic was 2 days(1~6 days),the intergenerational interval was 1 day(0~2 days),and the initial R,was 1.9(95%CI:1.7~2.1).The fittingresults for the BA.2 and BA.5.2 epidemics were good,and no statistical difference was observed between the predic-ted and actual numbers of cases(x2BA.2=31.53,x2BA.5.2=27.88,P>0.05).If an emergency response had not been initiated,the BA.2 epidemic would have continued to spread andpeak on April 7th,with an estimated 638 035 cases.The BA.5.2 epidemic would have rapidly spread,reaching a peak on November 14th,with an estimated 685 940 cases.If one incubation period were detected early,the scale of the BA.2 epidemic would have decreased by 25.73%;if two incubation periods were detected early,the scale would have decreased by 79.56%,and if one incubation period had been delayed,the scale would have expanded by 13.72%.If one incubation period had been detected early in the BA.5.2 epidemic,the scale would have decreased by 35.04%;if two incubation periods had been detected early,the scale would have decreased by 92.47%;and if one incubation period had been delayed,the scale would have increased by 19.75%.The guiding ideology,and the prevention and control measures for handling two local epidemics were optimized and iterated.Our study indicated that implementing the"four early"measures ef-fectively decreased the scale of the epidemic,and earlier detection was associated with more significant control effects.This study provides valuable information for the prevention and control of new or recurrent respiratory infectious diseases.
