1.Maternal Toxicity and Offspring Neurobehavioral Effects in Wistar Rats Induced by Exposure to Methylmercury During Pregnancy

Objective To explore the maternal toxicity and offspring's neurobehavioral effects in Wistar rats induced by exposure to methylmercury during pregnancy. Methods Methylmercury were continuously orally perfued with different doses of 0.00, 0.01, 0.05 and 2.00 mg/(kg?d) to 52 pregnant Wistar rats during the 6th~9th day after conception. The toxicity to maternal rats was observed and general teratology test were carried out. The parameters of physical and neurobehavioral develpment at early stage of 205 offspring were measured and the operant behavior test of 32 offspring was carried out at the age of 10 weeks. The brain's morphology and the levels of monamine neurotransmitter were tested in 24 maternal rats after 5 weeks of the delivery and 24 offspring in 10 weeks old. Double blind randomized design was used in whole experiment. Results No maternal toxicity and offspring teratogenicity were observed. Objective To investigate the influential factors of systemic lupus erythematosus(SLE) activity during pregnancy and their relationship with pregnancy outcome.Methods A retrospective analysis of the clinical history of 66 pregnant women with SLE from 1991 to 2005 was carried out.Results(1) Those patients with unstable status progestation,patients being newly diagnosed with SLE during pregnancy or patients irregularly using prednisone became active during pregnancy.The disease was active in 32 cases(the active group)and inactive in 34 cases(the inactive group).(2)Obstetric complications in the active group included:9 cases of preeclampsia,13 cases of fetal growth restriction(FGR),7 cases of therapeutic abortion and 15 cases of premature labor;and the corresponding numbers in the inactive group were 1,5,1 and 4, respectively.All the numbers were significantly different between the two groups(P
6.Biodegradation of a novel bionic scaffold with nanostructure in vivo

BACKGROUND: Bone implant materials have been previously reported to be not coincident between inducing velocity of new bone formation and degradation velocity itself; therefore, the materials could not be completely degraded but formed into foreign substances. A novel artificial bone implant material, characterizing by well biocompatibUity, biodegradation, and biomechanics, is focused in biomaterials field recently.OBJECTIVE: To study the biodegredation of a novel bionic scaffold with nanostructure, i.e., poly (3-hydroxybutyrate-co-3-hydroxyvalerata)/sol gel bioactive glass (PHBV/SGBG), in vivo. DESIGN, TIME AND SETTING: A controlled animal experiment was performed at Animal Experimental Center of the Third Hospital affiliated to Sun Yat-sen University from May 2005 to October 2006. MATERIALS: PHBV/SGBG was provided by Materials Institute of South China University of Technology, and ethylene oxide was sterilized for preparation.METHODS: Eight hybrid dogs were used to make models of Ubia diaphyseal defect, having two defects on both left and right sides. The tibia diaphyseal defects at proximal part were considered as the control group, and those were not performed with any treatment; while, the tibia diaphyseal defects at distal part were considered as the experimental group, and PHBV/SGBG was fully implanted into the defect regions. Every two dogs were sacrificed at different time points of 2, 4, 8, and 12 weeks, respectively. MAIN OUTCOME MEASURES: In vivo biodogradation and osteogenesis were monitored under optic microscopy and electron microscope.RESULTS: The PHBV/SGBG scaffold had well biodegradation and rapid degradation velocity, and it began to degrade at two weeks after operation. The PHBV/SGBG scaffold was almost replaced by new bone tissues at 8 weeks after operation and completely degraded at 12 weeks after operation. In addition, the PHBV/SGBG scaffold had a good ability to induce new bone formation from edge to center. Whereas, surface depression in the defect region was still visible in the control group, cortical bone was not formed in embedded region of soft tissue; furthermore, electron microscopy demonstrated that calcium salt deposition was increased in the bone defect region, and the structure was tight; however, the defect was not completely repaired, and some voids were still visualized.CONCLUSION: The novel bionic scaffold, PHBV/SGBG, degrades fast in vivo to generate new bone tissues. The new bone regenerate accompanied by a fitting degradation of the novel bionic scaffold that achieve complete repair.
7.Evaluation of Boned Metabolism Quota in the Prediction of Osteoporosis of Climacteric Female in Xi' an Area

Objective To investigate the relationship among age,menopausal status,serum 25 (OH)VD and skeletal metabolism index as PTH (Parathyroid Hormone),CA(Calcium),P (Phosphorus) and boned mineral density (BMD) of climacteric female in Xi'an.Methods 352 cases of healthy women at the age of 40~60 were collected who did routine examination in outpatient building of the First Affiliated Hospital of Xi'an Jiaotong University from January 2016 to April 2016.According to the results of skeletal density test,all subjects were divided into 3 groups as the normal group (T>-1),the reduction group (-2.5<T<-1),the osteoporosis group (T<-2.5).Electrochemiluminescent immunoassay (ECLIA) was used to detect levels of serum 25 (OH)VD,PTH,CA and P.Kruskal-Wallis test and chi-square test were used to analyze differences of age,serum 25(OH)VD,PTH,CA and P in different levels of bone mineral density groups.Results Compared with bone density normal group 2.24(2.18~2.29)mmol/L,serum CA levels of bone mineral density reduction group 2.27 (2.20 ~ 2.32)mmol/L and osteoporosis group 2.27(2.23~2.33)mmol/L were significantly higher (Z=9.669,P<0.01).Serum P levels of bone mineral density reduction group 1.15 (1.09~ 1.23)mmol/L and osteoporosis group 1.18 (1.09~ 1.25)mmol/L were obviously higher than bone density normal group,1.11 (0.99 ~ 1.23) mmol/L (Z =13.64,P< 0.01).Ages of osteoporosis group 54(50~57) years and bone mineral density reduction group 51 (47~ 54) years were significantly older than Bone density normal group 47(43~50) years (Z=73.08,P<0.01).The rate of menopause in osteoporosis group and bone mineral density reduction group were significantly higher than in bone density normal group (x2 =13.81,P< 0.01).There was no statistical difference in serum 25(OH)VD and PTH between groups.Conclusion This study found boned density of climacteric women is more likely correlated with age,menopause status and levels of serum CA and P.There was not enough evidence to prove that 25 (OH)VD and PTH have effect on skeletal density.Therefore,to monitor and detect the menopause status and levels of serum CA and P of climacteric female is critical for the prevention and treatment on osteoporosis of women.
8.Effect of circadian rhythm disorder on cognitive decline and neurologic impairment after traumatic brain injury in rats

Objective To explore the effect of circadian rhythm disorder on cognitive decline and neurologic impairment after traumatic brain injury (TBI) in rats.Methods Rats were subjected to a weight-drop model of TBI,then the rat models were divided into 2 groups ac-cording to the environmental length of day and night ,namely,the whole day group (12 h light/12 h light) and the control group (12 h light/12 h dark).After 14 days,the Morris water maze test and step-down test were carried out to evaluate the memory of the rats in each group . HE staining and immunohistochemistry ( BrdU, Ki-67) were carried out to evaluate the degree of the neurologic impairment of the rats in each group.And the mRNA expressions of Caspase-3,Caspase-9,Bcl-2 and Bax were evaluated with realtime PCR .Results The memory function of the rats in whole day group was significant lower than control group ;the damage degree of the cells in the hippocampus and the cortical lesion volume in whole day group were significant higher than control group ;the cell proliferation rate and newborn cell survival rate in the whole day group decreased significantly compared with the control group ;the mRNA expression of Bcl-2 in whole day group was signifi-ant lower than control group ,and the mRNA expression of Caspase-3,Caspase-9 and Bax in whole day group was signifiant higher than control group.Conclusion Circadian rhythm disorder can worsen TBI-induced cognitive decline and neurologic impairment .
9.Effects of celecoxib combined with oncolytic adenovirus armed with IL-24 gene on the human urothelial carcinoma T24 cells

Objective To evaluate the ability to kill human urothelial carcinoma T24 cells selectively in vitro by celecoxib combined with ZD55-IL-24 and explore the effectiveness for this combination use.Methods The EGFP expression of cells was observed by fluorescence microscope.The human umbilical vein endothelial cells (HUVEC) were used as crotrols.The expression of IL-24 mRNA was detected by RT-PCRwhen the cells were transfected by ZD55-EGFP or ZD55-IL-24.After transfected by ZD55-IL-24 and treated by celecoxib,the inhibition effect on cells was measured by MTT assay,and the apoptosis rate was examined by flow cytometry.Results The fluorescence in T24 and HUVEC can be observed 24h after ZD55-EGFP transfection and the fluorescence intensity was increased corresponding with the times.Fluorescence intensity in T24 cells showed higher than that in HUVEC group at the same times.The result of RT-PCR showed that the T24 cells expressed higher level of IL-24 mRNA than HUVEC group at the same time when the cells were transfect by ZD55-IL-24 (P < 0.01).The inhibition rate of ZD55-IL-24 combined with celecoxib group was significantly higher than other groups (P < 0.001).The inhibition rate of T24 cells in each group was significantly higher than HUVEC group (P < 0.01).The flow cytometry results indicated that celecoxib combined with ZD55-IL-24 had the highest apoptosis rate on T24 cells than other single use group.Apoptosis rate of T24 cells showed a higher than HUVEC cells (P < 0.01).Conclusion Celecoxib combined with ZD55-IL-24 can inhibit T24 cells proliferation at a greatest degree and this effect may be contributed to apoptosis.
