1.The clinical value of HPV E6/E7 mRNA and HPV DNA in early screening of cervical cancer
Tianjin Medical Journal 2016;44(4):466-469
Objective To analyze the differences of positive detection rate and copy number of human papillomavirus (HPV) DNA and E6/E7 mRNA between different grades of cervical lesions, and evaluate their clinical values in early screen?ing of cervical cancer. Methods The cervical exfoliated cell samples from 154 women undergoing biopsy examination and 32 objects undergoing hysterectomy (control group) were collected in Tianjin Central Hospital of Gynecology Obstetrics in 2014. According to the pathological results of cervical biopsy, 154 samples were divided into low-grade squamous intraepi?thelial lesion group (LSIL, n=51), high-grade squamous intraepithelial lesion group (HSIL, n=71), and squamous cell carci?noma group (SCC, n=32). HPV DNA was tested with hybrid capture technology, and E6/E7 mRNA was detected with fluores?cence quantitative hybridization. Immunohistochemistry was performed by detecting E6/E7 protein in all patients after sur?gery or cervical biopsy. Results Combined results of HPV DNA and E6/E7 mRNA demonstrated that the positive detection rate was significantly lower in control group than that of all levels of lesion groups (P<0.05). The copy number of high risk HPV E6/E7 mRNA was significantly increased with the aggravation of lesions (P<0.05), whereas no difference was found in that of HPV DNA. Compared with the normal control and low-grade squamous intraepithelial lesion group, cervical cancer patients with mRNA copies > 10 000 E6/E7 were significantly increased in high-grade squamous intraepithelial lesion group. Immunohistochemical results showed that the positive detection rate of E6/E7 was significantly lower in control group than that of all levels of lesion groups (P<0.05). The positive rate of E6/E7 was significantly higher in the high-grade squa?mous intraepithelial lesion group than that of low-grade group (P<0.05). Conclusion HPV infection is closely related to cervical abnormalities, which is one of effective measures for early screening of cervical cancer. The negative result of HPV DNA is very helpful to exclude the cervical abnormality, whereas the positive detection of mRNA has great value in predict?ing the disease. Combined results of positive detection and copy number make a comprehensive evaluation for the risk of cer?vical lesions.

BACKGROUND:There is little attention focused on the effect of isokinetic testing about the variation of angular velocity on pain and muscle strength in the evaluation of curative effect of McConnel taping. OBJECTIVE: To evaluate the changes of quadriceps muscle strength and perceived pain levels in patients with patelofemoral pain syndrome before and after corrective McConnel taping in isokinetic testing. METHODS:A total of 34 patients with patelofemoral pain syndrome were evaluated in isokinetic testing for concentric contraction muscular strength, including the maximum torque, the total work, and the average work in 60 (°)/s and 180 (°)/s angular velocity, before and after the interventions of McConnel taping. The visual analogue scale scores were recorded. RESULTS AND CONCLUSION: The visual analogue scale scores were slightly decreased in angular velocity of 60 (°)/s after obturation when compared to before obturation (P > 0.05), while the scores were significantly decreased in angular velocity of 180 (°)/s (P < 0.05). The maximum torque, the total work, and the average work were slightly increased after obturation, but showed no significant difference in angular velocity of 60 (°)/s (P > 0.05), while significantly increased in 180 (°)/s angular velocity (P < 0.05). McConnel taping significantly improves the curative effect in patients with patelofemoral pain syndrome under low load joint movement.

Objective: To evaluate the effects of stent implantation for treatment of Budd-Chiari syndrome. Methods:A total of 16 patients with pasthepatic inferior vena cave (IVC) obstruction syndrome were treated by balloon catheters ( Polythene and Inoue ) in percutaneous transluminal angioplasty (PTA) , then stent were inserted into the obstruction. Results: The residual pressure gradient after PTA and stent implantation was decreased from (4.6?0. 3 ) kPa to (2. 1?0. 1) kPa. The diameter of IVC after PTA and stent implantation was increased from (3.8?1. 7)mm to (14. 9?2. 0)mm. Conclusion : PTA and stent implantation are effective treatment for pasthepatic inferior vena cave obstruction.

Ramie and kenaf were traditional fiber crops in China, but their stalk after decorticating has not been used effectively. The stalk contains a lot of cellulose, and can therefore be used for the production of bioethanol. We studied the effects of different chemical pretreatment on enzymatic digestibility of ramie stalk and kenaf stalk. Ramie and kenaf stalks pretreated with alkali were chosen to produce ethanol using quasi-simultaneous saccharification and fermentation (Q-SSF) process. The results show that for the stalks pretreated with 4% NaOH and 0.02% anthraquinone-2-sulfonic acid sodium salt (AQSS) as catalyzer at 170 degrees C for 1 h, the ethanol concentration could reach 51 g/L after fermentation for 168 h at 18% of solid substrate concentration. By fed-batch to 20% of solid substrate concentration, the ethanol concentration could reach 63 g/L, 77% and 79% of the cellulose conversion could get for ramie stalk and kenaf stalk, respectively. For kenaf stalk pretreated with 5.2% NaHSO3 and 0.2% H2SO4 at 170 degrees C for 1 h, the ethanol concentration and cellulose conversion could reach to 65 g/L and 72%, respectively.

Chinese Journal of Biotechnology 2014;30(5):765-773
Recent concerns about the gradual depletion of conventional fossil resources and the pressure from global climate change have accentuated the need for new alternative feedstock. As one of the main components in biomass, lignin is the second most abundant natural polymer after cellulose, and has the potential to serve as a sustainable source of energy and organic carbon to replace petroleum-based chemicals. Efficient conversion of lignin into high value-added chemicals is crucial to improve the economic feasibility of biomass refinery. In the present study, several pretreatment technologies on industrial lignin were carried out to enhance phenol production. A microwave irradiation assisted biphasic reaction system was used to convert pretreated industrial lignin into phenolic compounds. Lignin conversion, reaction temperature, time and pretreatment method, were optimized. The highest phenol yield was 8.14% obtained from lignin pretreated by 1-ethyl-3-methylimidazolium acetate at 400 W for 60 min in a biophasic system catalyze by 1-aminoethyl-3-methylimidazolium tetrafluoroborate.

Microbiology 1992;0(02):-
CBS(Case Based Study)is a new teaching method, which is based on the case given. It requires students to answer questions according to the case. In the teaching, the students who are divided into groups, seek for resolvent by themselves. They should combine the information provided in the case, find the key answer and the relationship. The students gain the knowledge by reviewing and studying in the textbook, then analyse and solve questions with them. The application of CBS could make knowledge more relatively and systematic, moreover,it also make study more actively and initiatively.
