1.Anti-UACA autoantibody in thyroid-associated ophthalmopathy

Serum anti-uveal autoantigen with coiled coil domains and ankyrin repeats (UACA) antibody titer was measured using ELISA in 32 patients with thyroid-associated ophthalmopathy ( TAO ) and 20 patients with Graves' disease (GD) without ophthalmopathy.The level of anti-UACA antibody in TAO group was higher than that in GD group [ (225.91 ± 144.21 vs 187.42 ± 46.77 ) pg/ml,P<0.05 ],and especially higher in patients with ocular myopathy [ (254.35 ± 168.29 ) pg/ml ].It seems that anti-UACA antibody is the sensitive clinical marker of ocular myopathy in TAO patients.

The platelet membrance glycoprotein plays a key role in the adhesion,aggregation and release of platelet.It is the specific molecular marker for platelet activation.Many studies have confirmed that platelet membrane glycoprotein CD62P may be more sensitive and specific to reflect the occurrence of coronary artery development,CD62P level is positively correlated with the severity.A variety of traditional Chinese medicine can lower the CD62P level,inhibit platelet activation and the treatment play an important role in the treatment.
3.The study of perioperation parenteral nutrition in patients with malignant gastrointestinal tumor and diabetes

Objectives: To study safety method of control of blood glucose in perioperation patients with malignant gastrointestinal tumor and diabetes. Methods: The twenty eight cases with malignant gastrointestinal tumor and diabetes were analyzed retrospectively. Results: The patients with diabetes were safely controled in blood and urine glucose during perioperetion.There were no significant changes in serum lipids.The excretions of urea nitrogen in urine increased 24 hours after operation.The body weights declined and plasma proteins decreased after operation. Conclusions: The nutritional support is necessary and of feasibility in patients with malignant tumor and diabetes during perioperation.It is a key step to control blood glucose by using insuline.The nutritional support can protect function and structure of oragans.

8. Establishment of a methotrexate-resistant cell line of human osteosarcoma and its biological characteristics

Objective: To establish a methotrexate (MTX)-resistant human osteosarcoma cell line (Saos-2/MTX1 and Saos-2/MTX2) Lived from the Saos-2 cell line and observe its biological characters. Methods: Saos-2 cells were pulse-exposed to MTX with gradually increased dosage. Both MTX-resistant human osteosarcoma cell lines (Saos-2/MTX1 and Saos-2/MTX2) were established after 178 days. The sensitivity of the Saos-2, Saos-2/MTX1, and Saos-2/MTX2 cell lines to MTX, cisplatin (DDP), doxorubicin (ADM), ifosfamide (IFO), epirubicine (EPI), pirarubicin (PTH), and paclitaxel (PTX) was measured by MTT assay. The morphological and ultra-structural changes of the cells were observed by optical microscopy and transmission electronic microscopy. The cell proliferation ability was calculated from the growth curve. The cell cycle distribution was analyzed by flow cytometry. Results: The MTX-resistant human osteosarcoma cell lines, Saos-2/MTX1 and Saos-2/MTX2, were established after 178 d induction. The resistance indexes to MTX were 4.87 and 12.73 times than that of Saos-2, respectively. The two cell lines had cross resistance to ADM, EPI, PTH and TAXOL and were still sensitive to DDP (P > 0.05). Optical microscopy showed the volume of Saos-2/MTX1 and Saos-2/MTX2 cells were enlarged and the number of poly-nucleus cells was increased. Transmission electronic microscopy revealed the cell process was decreased and the nucleoli were increased. Cell proliferation curve and flow cytometry demonstrated that the proliferation abilities of Saos-2/MTX 1 and Saos-2/MTX 2 cells were decreased significantly and the proportion of cells in S phase was reduced (P < 0.05). Conclusion: We establish two MTX-resistant human osteosarcoma cell lines (Saos-2/MTX1 and Saos-2/MTX2) and observe their biological characteristics. It provides a new experimental model for further studying the mechanism of MTX-resistance.
9.Evaluation of the Self-rated Health Measurement Scale-the Revised Version 1.0

Objective: To evaluate a self-rated health measurement scale-the revised version 1.0 (SRHMS V1.0).Method:2000 subjects, including university students, patients in the department of traditional medicine, patients with dermatosis and military retired cadre, received the assessment with SRHMS V1.0. Result: The test-retest reliability of the above four groups was 0.80-0.98. The Cronbach alpha was 0.93, equal-length Spearman-Brown was 0.74, Guttaman Split-half was 0.73. The correlation between each item and its dimension was high, that between item and other dimension was low; furthermore, the correlation between each dimension and the subscale it belong to was high, that between the dimension and other subscales was low. Factor analysis produced ten main factors that were consistent with the original theoretical structure about SRHMS V1.0. The correlation between total score of SRHMS and that of SF-36 was significant (r=0.53). People of different age group, of different marriage status, with different occupations or education background, of different economic class had different results in SRHMS V1.0, while there was no difference significantly between male and female, between with different races or residencies. Conclusion: SRHMS V1.0 has good psychometric quality and can be used in Chinese population.
10.SCREENING AND CLONING OF THE GENES OF PROTEIN INTERACTING WITH THE NONSTRUCTURAL PROTEIN 5B OF HEPATITIS C VIRUS

Protein-protein binding is the basis of virus and host cell interactions. With the application of technology of studying protein interactions, more knowledge of replication and pathogenesis of hepatitis C virus (HCV) was acquired. Non-structure protein 5B(NS5B) of HCV is a kind of viral protein, which plays an important role in replication of HCV. However, the effect of NS5B is not clear. To investigate the biological function of NS5B, we performed yeast two hybrid to look for proteins in hepatocytes interacting with NS5B. We constructed NS5B bait plasmid by cloning the gene of NS5B into pGBKT7, then transformed it into yeast AH109(a type). The transformed yeast was mated with yeast Y187(? type)containing liver cDNA library plasmid in 2?YPDA medium. Diploid yeast was plated on synthetic dropout nutrient medium (SD/-Trp-Leu-His-Ade) containing x-?-gal for screening. Thirty-three colonies were selected and sequenced. Among them, two colonies were new genes with unknown function. The preliminary successful cloning of gene of protein interacting with NS5B paved the way for the study of the physiological function of NS5B and its associated protein.
