2.Biological features and ultrastructure of rabbit mesenchymal stem cells in vitro

Objective To isolate and culture the rabbit mesenchymal stem cells,and explore the biological features in vitro.Methods Two milliliter of bone marrow was extracted from the tibia of juvenile rabbit under anesthesia and sterile conditions,and mesenchymal stem cells were obtained by density gradient centrifugation and inoculated to 100ml culture flask.The morphological changes,adherence rate and ultrastructure of the stem cells were observed.The surface markers and growth cycle of the mesenchymal stem cells were detected by flow cytometry,and biological features were observed by inducing the cells differentiate into osteoblasts.Results The adherence of mesenchymal stem cells started 2 hours after culturing,basically finished 20 hours later,and more than 90% of the cells were fused at the 15th day.Observed with transmission electron microscopy,the cultured cells showed typical ultrastructure of stem cells: numbers of microvillus mainly distributed on one side of the cells,irregular nuclei with indentation,and lots of cell organs.Analysis for the cell surface markers indicated positive expression of CD44 and CD90 and negative expression of CD45 in mesenchymal stem cells.Cell growth cycle assay showed that most mesenchymal stem cells(about 97%) stayed in stage G1 and G2.Three weeks after osteoblastic induction,the mesenchymal stem cells gradually produced small calcium salt crystals,the cells were positive in alkaline phosphatase staining,and Von Kossa staining showed many calcium salts in the cell clone as black reaction deposits.Conclusions The rabbit bone marrow mesenchymal stem cells isolated by density gradient centrifugation are of high purity and stable growth rate,and the cells' characters can be still maintained after serial subcultivation.Since easy to operate on isolation and proliferation,the mesenchymal stem cells could be used as the tissue engineering seed cells in repairing of intervertebral disc degeneration.

Objective To discuss the therapy modalities and their efficacy for Cobb's syndrome Methods Data from 14 cases of Cobb's syndrome were retrospectively reviewed of which diagnosis, management and follow-up were discussend. Results Follow-up period of the 14 cases was 4 months to 3 years, of them, no complete cure occurred, 12 cases improved, 2 cases deteriorated. Conclusions Early diagnosis and early treatment using in terventional and surgical method are the key point for this kind of in curable diseases.
4.Efficiency of resin infiltration versus fluride varnish for treatment of post-orthodontic white spot lesions

BACKGROUND:White spot lesions caused by enamel demineralization are a common side effect of orthodontic treatment. Resin infiltration treatment for white spot lesions has obtained favorable effects, but there is no report on the difference between resin infiltration and fluoride varnish treatment for post-orthodontic white spot lesions. OBJECTIVE:To compare the efficiency of resin infiltration and fluride varnish for treatment of post-orthodontic white spot lesions. METHODS:Twenty-nine patients with post-orthodontic white spot lesions were randomly divided into two groups:one group accepted the resin infiltration treatment and the other group accepted fluoride varnish treatment. Frontal intraoral photos before treatment and 6 months after treatment were col ected for study. The difference in efficiency of resin infiltration and fluride varnish for treatment of post-orthodontic white spot lesion was analyzed and evaluated by both dentists and College students who were not engaged in oral medicine. RESULTS AND CONCLUSION:Except one patient treated with fluride varnish, al the patients completed the referral on schedule. There was significant improvement in the two groups after treatment (P<0.05). Resin infiltration treatment was better than fluoride varnish treatment in post-orthodontic white spot lesions. Thus, resin infiltration that requires fewer referrals and has better effects is an ideal therapy for post-orthodontic white spot lesions.
5.Clinical Effcicacy of Antiplatelet Combined with Anticoagulation Therapy for Restenosis Prophylaxis after Stent Placement in Diabetic Patients with Leg Atherosclerosis Obliterans

Objective:To investigate the clinical efficacy and safety of antiplatelet combined with anticoagulation therapy for restenosis prophylaxis after stent placement in diabetic patients with leg atherosclerosis obliterans(LASO). Methods:83 diabetic LASO patients with stent placement were collected and randomly divided into the treatment group(42 cases)and the control group (41 cases)with 49 sick limbs in each. The control group was treated with clopidogrel(75mg,for 1 year)plus bayaspirin(100 mg),while the treatment group was treated with low molecular heparin(withdrawal when INR = 2. 0- 3. 0)plus warfarin(for 6 months)additionally. All cases were followed up for 12 months and the efficacy and safety were compared and evaluated. Results:After therapy,there were no differences in FPG,GHbA1C,TC and LDL-C at each time point between the two groups(P >0. 05). Compared with that in the control group,MLD in the treatment group was improved obviously(P < 0. 05)in the 12th month after the surgery,while restenosis rate and level of D-D in plasma were declined significantly(P < 0. 05). And there was no difference in bleeding rate between the two groups(P > 0. 05). Conclusion:Compared with antiplatelet therapy,antiplatelet combined with anticoagulation therapy for restenosis prophylaxis after stent placement in diabetic LASO patients is more effective and safer,which is worthy of clinical promotion.
6.Management of point-of-care testing glucose monitoring program in hospital in the JCI accreditation

Point-of-care testing (POCT) of blood glucose has been widely used for continuous monitoring of glucose in clinic due to high sensitivity, convenience, accessibility and shorter TAT.It has been developed rapidly in recent years. But glucose POCT is often out of control and management from central laboratory and usually it was performed by non-laboratorians.It becomes the growing concerns that how to avoid the discrepancy between POCT results and central laboratory results, how to ensure POCT quality of each batch of blood glucose results, how to solve issues in the application and ensure that patients' results are accurate and reliable. The hospital should put the POCT management under the whole management system and improve the accuracy of POCT to achieve the complete quality assurance. This article introduced the management program experiences of glucose POCT in Huashan hospital according to the JCI accreditation and CAP-LAP.
8.Experimental Study on Antitussive, Expectorant and Antiasthmatic Effects of Fortunella Hindsii Leaves

Objective: To study the antitussive, expectorant and antiasthetic effects of Fortunella hindsii leaves. Methods: The antitussive effects were observed by the method of ammonia-induced cough in mice, the expectorant effects were observed by the method of phenolsulfonphthalein excretion in mice, and the antiasthmatic effects were observed by the method of histamine phosphate spray in guinea pigs. Results:The leaves of Fortunella hindsii at low, medium and high doses could decrease the cough times and prolong the cough latent period (P<0. 05 or P<0. 01), high dose could promote the phenol red excretion (P<0. 05), and high and medium do-ses could prolong the incubation period of guinea pigs for asthma caused by histamine phosphate (P<0. 05 or P<0. 01). Conclu-sion:The leaves of Fortunella hindsii have promising antitussive, expectorant and antiasthmatic effects.
9.The protective effect of resveratrol on arterial remodeling in spontaneous hypertensive rats

Objective To observe the effect of resveratrol on arterial remodeling in spontaneous hypertensive rats.Methods Spontaneous hypertensive rats (SHRs, n=20) and Wistar-Kyoto rats (WKYs, n =10) were randomly assigned to three groups: control group, model group and resveratrol group.Systolic blood pressure (SBP) and pulse wave velocity (PWV) of the three groups was observed.Morphological observations were obtained by hematoxylin and eosin or victoria blue and picrosirius red.The content of nitric oxide (NO) and endothelin-1 (ET-1) were determined to assess the endothelial function.Oxidative stress was assessed by malondialdehyde (MDA), hydrogen peroxide (H2O2) , and activities of antioxidant enzymes superoxide dismutase (SOD) and catalase (CAT).Results Compared with control group, SBP, PWV, wall thickness, wall-lumen ratio,vascular cross-sectional area (VCSA), luminal cross-sectional area (LCSA) and collagen-elastic ratio were increased, and the levels of NO, SOD, CAT in rat aortic tissues were decreased, while ET-1,MDA, H2O2 levels in rat aortic tissues were enhanced in model group at the end of 22 weeks.After the treatment of resveratrol, SBP had no significantly difference between model and resveratrol groups, while PWV, aortic wall thickness, wall-lumen ratio, VCSA, LCSA and collagen-elastic ratio were decreased in resveratrol group as compared with model group [(6681.1 ± 2154.9) cm/s vs.(4283.1±946.1) cm/s, (234.7±51.8) μm vs.(123.4±21.5)μm, (10.3±2.3) % vs.(5.8±1.4) %, (6.4±0.4) mm2 vs.(4.8±0.6) mm2, (4.1±0.1) mm2 vs.(3.2±0.4) mm2, (1.1±0.3) vs.(0.4±0.2), P<0.05 or 0.01].Compared with model group, the expressions of NO, SOD, CAT in rat aortic tissues were increased and levels of ET-1, MDA, H2 O2 in rat aortic tissues were decreased in resveratrol group after treatment of resveratrol (P<0.01 or P<0.05).Conclusions Resveratrol has an antioxidant effect, and it could attenuate arterial remodeling by improving oxidative stress.
10.Application of levels of serum CA199,C3,C4 and lipid metabolism in clinical diagnosis of pancreatic cancer

Objective:To evaluate the diagnostic values of single and combined detection of serum CA199, complement 3 (C3),complement 4 (C4),total cholesterol (TC),triglyceride (TG),and lipid metabolism levels in the patients with pancreatic cancer, and to explore their correlations with TNM stage and pathological stage of pancreatic cancer.Methods:Total 185 subjects were enrolled into the study by three groups:pancreatic cancer patients group (Pc group,n=77),non-digestive system cancer patients group (Ndc group,n=58)and healthy control group (Hc group,n=50).The levels of serum CA199,C3,C4,and high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL-C), apolipoprotein A (ApoA), apolipoprotein B (ApoB), apolipoprotein E (ApoE),and lipoprotein a (Lpa)levels were detected.Results:① The serum level of CA199, C3,C4,and ApoE of the patients in Pc group were higher than those in Ndc and Hc groups (P<0.01).No statistical difference was observed in the serum CA199 between Ndc and Hc groups (P>0.05).The levels of C3, C4,and ApoE in Pc group and Ndc group was higher than those in Hc group (P<0.01),and the levels of the biomarkers in Pc group were also higher than those in Ndc group (P<0.01).The levels of HDL-C,ApoA and Lp (a)of the patients in Pc group were significantly lower than those in Ndc and Hc groups (P<0.05).② The area under ROC curve (AUC)of serum CA199,C3,C4,ApoE,HDL-C,and ApoA were 0.916,0.841, 0.788,0.785,0.834,and 0.810,respectively.Furthermore,multiple factor analysis showed that the combined detection of CA199,C3,and HDL-C (AUC=0.968)improved the diagnosis compared with detecting CA199 alone (P<0.05).③ The CA199 level of the patients inⅢ-Ⅳ stage of TNM stage was higher than that in the patients inⅠ-Ⅱ stage (P<0.01).For the pathological stage,the ApoA level in low differentiation group was higher than that in moderate and high differentiation group (P<0.05).There was no statistical difference in other biomarkers between the different TNM stages and pathological stages.Conclusion:The levels of CA199,C3,C4 and ApoE of pancreatic cancer patients are significantly increased, while the levels of HDL-C, ApoA, and Lp (a ) are significantly reduced.Combined detection of CA199,C3,and HDL-C can improve the early diagnosis of pancreatic cancer compared with the single assessment of each biomarker.
