Objective To explore the effect of HgCl2 exposure on energy metabolism of renal mitochondria in rats and the preventive effect of N-acetylcysteine(NAC).Methods Twenty-four Wistar rats were randomly divided into control group,HgCl2 group and NAC+HgCl2 group by body weight,8 in each.Control group and HgCl2 group were treated with physiological saline through intraperitoneal injection,NAC+HgCl2 group rats were intraperitoneally pretreated with NAC(0.4 mmol/kg),two hours after treatment,HgCl2 group and NAC+HgCl2 group were injected subcutaneously with HgCl2(1.84 ?mol/kg),but control group was injected with physiological saline,the volume of was 5 ml/kg,once a day for 14 consecutive days.Twenty-four hours after the last injection,all rats were sacrificed and the cortices of rat kidneys were collected.The mitochondria were prepared by using the differential centrifugation.The levels of malondialdehyde(MDA),the activities of phospholipase A2(PLA2) and succinate dehydrogenase(SDH),the mitochondrial membrane potential were measured.Results Compared with control group,the level of MDA and the activity of PLA2 significantly increased in HgCl2 group(P

ObjectiveTo explore the long-term fluctuation of fasting plasma glucose (FPG) and its effect on prognosis in patients surviving for more than 1 year after renal transplantation.MethodsFour hundred and forty-six patients underwent kidney transplantation from January,1993 to December,2008.According to preoperative FPG levels,patients were divided into diabetic,impaired fasting glucose (IFG),and normal fasting glucose (NFG)groups. The changing trend of FPG level was observed and analyzed. For 428 non-diabetic patients before transplantation,the prevalence and different outcomes of post-transplantation diabetes( PTDM ) according to FPG after transplantation were analyzed.The characteristics of the patients with persistent PTDM ( P-PTDM ) and transient PTDM (T-PTDM) were compared.The incidence of complications and patient survival between the PTDM group and non-PTDM groups were also compared.ResultsFPG level was increased early and then decreased in patients after renal transplantation.Of the 428 patients,87 developed into PTDM ( 20.3％ ) including 15 T-PTDM patients ( 17.2％ of total PTDM ),who eventually recovered to NFG or IFG.Compared with P-PTDM group,the incidence of acute rejection episodes was higher for T-PTDM ( P =0.043 ).The incidence of infections,hypertension,and dyslipidemia within the first year,was higher in PTDM group compared with non-PTDM group but patient survival was not different within a mean follow-up of ( 5.65 ± 3.68 ) years.ConclusionPTDM will not be permanent and may recover to NFG or IFG in the course of the disease.Acute rejections are associated with the onset of T-PTDM.The overall patient survival is not affected by PTDM,although complications,such as infections,hypertention,and hyperlipidemia are more frequently encountered in PTDM patients.

BACKGROUND: A large number of preclinical experimental data have shown that stem cells can regulate the immune function, and serve the function of cell replacement. OBJECTIVE: To summarize the anti-inflammatory effects of stem cells in the treatment of ischemic stroke, based on which, we further discuss the specific mechanisms. METHODS: We conducted a systematic and comprehensive search in PubMed, Elsevier, Springer, Wiley, Ovid, EBSCO databases. The range of retrieval time was from 2012 to 2017. The keywords were stem cells, stroke, inflammation, immune. Totally 110 articles were retrieved initially, and 47 articles were included in result analysis. RESULTS AND CONCLUSION: By reading extensive literature, we analyzed and summarized the research status quo of the anti-inflammatory effects of neural stem cells, mesenchymal stem cells and endothelial cells in the treatment of ischemic stroke. The mechanisms mainly include reducing focal inflammation, immune regulation, promoting the secretion of various neurotrophic factors, reducing secondary cell death, protecting neurons and promoting cell function recovery and further promoting the recovery of nerve function. The mechanisms underlying local immune regulation and anti-inflammatory effects of stem cells are mainly described as the shift from M1 to M2 macrophages under the intervention of stem cell factors, to intervene secreted immune cytokine profiles and exert effects on inhibition and polarization of glial cells. Further investigation is required on the anti-inflammatory effects and immune regulation of stem cell therapy for stroke.

Objective To discuss the relationship between the fracture of the clavicle which was fixed by tor-sional-shaped plate and nonunion after operation.Methods Retrospective analysis was conducted in 426 patients with midshaft clavicle fractures.Among the patients,the A group used torsional-shaped plate to meet the anatomical structure of the clavicle:the medial plate matched under the front face of the clavicle,and the lateral plate matched the up face of the clavicle.And the traditional method was used in the B group.Patients were followed up for at least 12 months,the patients were judged by X-ray criteria for nonunion.Results A total of 28 patients with nonunion, including 4 cases in A group,24 cases in B group.After statistics processing,the difference between A group and B group was significant (χ2 =6.679,P=0.010).Conclusion We find that the treatment of the fracture of the middle part of the clavicle with torsional-shaped plate can effectively reduce the incidence of nonunion.

Objective To investigate the expression and significances of DPP-4 and IL-6 in febrile seizures(FS).Methods FS were induced in Sprague-Dawley(SD) rats at P14 in a hot water bath by using classical model of hyperthermia-induced seizures.A genome-wide microarray experiment was generated in the rats.The relationship between the differentially expressed genes were analyzed by the method of bioinformatics, and the gene and protein levels of DPP-4 and IL-6 were detected by QPCR,WB and ELISA.Selected 50 children with FS(FS group) and 25 healthy children(control group), and to compare the gene and protein levels of DPP-4 and IL-6 between the two groups.Results Interaction network diagram of differential gene expression showed that there may be interactions between DPP-4 and IL-6.Animal and clinical experiments showed that the DPP-4 and IL-6 gene and protein levels were significantly higher in FS group compared with control group(P<0.05).Conclusion There were high gene and protein expressions of DPP-4 and IL-6 in the FS group compared with the control group.These results indicated that immune and inflammations may play an important role in the FS, and it has provided an attractive pharmacological target for the treatment of FS in clinic.

Objective: To research antithrombotic effect and mechanism of total saponins of Astragalus (TSA).Methods: Effects of TSA on artery thrombosis anud platelet aggregation were studied, at dosage of 50,100,200mg/kg. And level of TXA 2,PGI 2 in plasma, content of NO in serum of rats were determined.Results: TSA could restrain artery thrombosis and platelet aggregation of rats, and increase PGI 2 and NO.Conclusion: There was apparently antithrombotic effect of TSA.

Objective To assess the distribution of blood lead levels and the sources of lead exposure in children with lead poisoning,and thus to offer recommendations for clinical diagnosis and treatment of childhood lead poisoning.Methods The clinical data of 129 patients with lead poisoning was collected and analyzed at the Out-patient Department of Lead Poisoning Xinhua Hospital Affiliated to Shanghai Jiaotong University School of Medicine during Sep.2012 and Aug.2013 were collected and analyzed.All children were required to fill out theoutpatient questionnaire on heavy metal (including children's demographic data,growth assessment,frequency of hand-mouth behavior,and the behavior of washing hands before eating,dietary,sources of lead exposure,and the family environment,etc.).Other data of medical history,physical examinations (height,weight,growth and development) were also collected.Blood samples were collected to measure blood lead level by graphite furnace atomic absorption spectrometry.Results (1) The above 129 patients were from 14 provinces (municipalities and autonomous regions),including 64 cases in Zhejiang (49.6％),30 cases in Shanghai (23.0％),13 cases in Jiangsu (10.1％),6 cases in Jiangxi (4.7％),5 cases in Hebei (3.9％),2 cases in Anhui and Guangdong (1.6％) ;and 1 case in Fujian,Henan,Hunan,Jilin,Inner Mongolia,Heilongjiang and Shandong (0.8％),respectively.(2) In the patients,the blood lead level was 17.0-892.0 μg/L[(251.5 ±155.8) μg/L] and the median was 235.0 μg/L.(3)The mean age of the children was 4.3 years.Fifteen cases were less than 1 year old,and the mean blood lead level was (367.8 ± 137.7) μg/L.Thirty-seven cases were 1-3 years old children,and the mean blood lead level was (250.5 ± 116.3) μg/L.Fifty cases were 3-6 years old children,and the mean blood lead level was (237.7 ± 179.7) μg/L.Twenty-seven cases were over 6 years old,and the mean blood lead level was (213.9 ± 141.8) μg/L.(4) One hundred and eleven cases of the children could find the definite sources of lead exposure,mainly from industrial pollution (35.7％) and domestic pollution (64.3％).The blood lead levels in 18 cases were less than 100 μg/L,and their definite lead pollution source was not found.(5) Most of the children had the symptoms of inattention,hyperactivity,aggressive behavior,constipation and abdominal pain,and so on.(6)Logistic regression analysis of children with blood lead levels ≥ 235.0 μg/L showed that lead pollutants and age were the main risk factors for lead poisoning.Conclusions Industrial pollution are associated with higher blood lead levels among children aged 0-6 years old (occupying one-third of the pollutants).The younger children tend to have higher blood lead levels,and the data also suggest that greater attention should be paid to children who used red powder.

After several thousand years of development,TCM has formed its own sets of research methods.such as observation,comparison,classification,trial and error,and deduction,etc.Based on the unique TCM theory,experiences of clinical diagnosis and treatment of TCM,the author put forward that the research of TCM should combine macrophenomenon and microphenomenon,quality and quantity,and"Zheng"and"Xie",emphasize locality feature for the purpose of promoting the science research level.

Cinobufacini is an aqueous extract of Bufo bufo gargarizans Cantor dried skin, which has been widely used for cancer therapy in China. So far, its active components are still not very clear. In previous reports, bufadienolides with low-concentration were usually studied because of their anticancer effects. However, the high polarity constituents in cinobufacini are less investigated. The present study found that more than 50% contents of cinobufacini were water-soluble peptides. Then, in vitro anticancer experiments were carried out, including human stomach cancer cell lines BGC823 and MCG803, human colon cancer cell lines DLD-1 and HT-29, and human pancreatic cancer cell line MIAPACA-2. The IC50 for these cell lines model were ranged from 25-123 microgmL(-1). The results indicated that these peptides showed similar activity with cinobufacini injection. As a conclusion, this study provides a new and further understanding of anticancer components in cinobufacini injection.

Objective To investigate the protective effects of midazolam on noise-induced hearing loss in guinea pigs by testing reactive oxygen species (ROS) level in the cochlea and plasma SOD and MDA. Methods Totally forty male pigmented guinea pigs were randomly divided into four groups: control (C) , midazolam (M), normal saline (S) and noise-induced deafened (D) groups, with ten guinea pigs in each. Groups M, S and D were exposed to a continuous noise (4kHz , octave band, 100dB SPL) 3h every day for 3 consecutive days. Group M was treated with midazolam, which was administered intramuscularly (0.1mg/kg) 24h before noise exposure, and immediately upon and during noise exposure. Group S was exposed to noise and treated with the same volume of normal saline intramuscularly, the time of injection was the same as that of Group M. Group C was not exposed to noise, but was treated with midazolam intramuscularly, the time of injection and the dosage were the same as those of Group M. Group S was exposed to noise and treated with normal saline intramuscularly ,the time of injection was same with that of Group M.Group D was exposed to noise only. All animals received auditory brainstem response (ABR) threshold recording before and immediately after noise exposure. Blood was collected when the guinea pigs were killed after the last ABR threshold recording, and serum SOD activity and MDA content were detected. Both the cochleae were removed and prepared for ROS assay. Results After noise exposure, ABR threshold shift (1.6±1.5) and ROS content [(291.10±2.30)u/mL] in Group M were significantly lower than those in Groups S and D [41.7±3.3, 44.3±3.9; (348.52±3.60)u/mL, (315.56±6.70)u/mL, P<0.05]. Serum SOD activity and MDA content were significantly increased in Group M, but the amplitude was less than that in Groups S and D.Conclusion Midazolam can prevent noise-induced hearing loss by reducing the increased ROS level in the cochlea after noise exposure.
