3.Analysis on The risk factors of diabetic nephropathy in elderly type 2 diabetes mellitus

Objective To investigate the correlation risk factors of diabedc nephropathy(DN)in elderly type 2 diabetes mellitus(T2DM).Methods Eighty-six cases of elderly T2DM were divided into two groups,DN group(43 cases)and NDN group(43 cases).Their age,fasting plasma glucose(FPG),2 hour postprandial glucose(2hPG),triglycerides(TG),total cholesterol(TC),high-density lipoprotein cholesterol (HDL-C),low-density lipoprotein cholesterol(LDL-C),hemoglobin A1c(HbA1c),systolic blood pressure (SBP),diastolic blood pressure(DBP)were observed.Results Compared with that in NDN group,2hPG,TG,SBP,HbA1c wag obviously increased in DN group[(14.13±4.46)mmol/L vs(11.19 ±4.22)mmol/L,(1.51±0.79)mmol/L vs(1.20 ±0.53)mmol/L,(141.16±19.08)mm Hg(1 mm Hg=0.133 kPa)vs(132.79 ±17.40)mn Hg,(7.55±2.09)%vs(6.65±2.02)%](P<0.01 or<0.05).Multiple regression analysis showed that the risk factors of DN included 2hPG and TG.Conclusion DN in T2DM is related with 2hPG and TG.
5.Investigation of Skeletal Age of Adolescent in Yantai Area

ObjectiveTo assesses the skeletal development of adolescent in Yantai area, and investigates the characteristics of the tendency of their skeletal development. MethodsTotally 1180 healthy adolescents ( 574 males and 606 females ) at 10 to 20 years of age living in Yantai area were randomly selected, and their left wrist were radiated at the posterior-anterior position. The bones at wrist were scored according to the Chinese Wrist Skeletal Development Standard CHN Method. The correlation among skeletal age, chronological age and the sex were analyzed. ResultsSkeletal age positively correlated with chronological age (male: r=0.943,females: r=0.942,totally: r=0.942). ConclusionThe skeletal development of adolescent in Yantai area has the characteristic of acceleration. So it is necessary to establish a new skeletal age standard for the skeletal development of adolescent.
7.Clinical efficacy of intermittent androgen suppression treatment of advanced prostate cancer in elderly patients

Objective To explore clinical effect and safety of intermittent androgen suppression treatment of advanced prostate cancer in elderly patients.Methods 78 patients with advanced prostate cancer were enrolled,and randomly divided into the observation group and the control group (n=39 each).Patients in observation group were treated with intermittent androgen suppression treatment.Patients in control group were treated with persistent androgen suppression treatment.Results The time of therapy was much longer in observation group than in control group (P< 0.001).Quality of life was better in observation group than in control group (P<0.05).The incidences of adverse reactions including anemia,fever and abnormal liver function were lower in observation group than in control group (all P<0.05).Conclusions Intermittent androgen suppression treatment prolongs time to androgen-independent progression,improve the quality of life,reduce drug dosage and the incidence of adverse reactions in the treatment of advanced prostate cancer in the elderly.
8.Evaluation the Effect of Dexamethasone Addition in Anesthesia on Postoperative Quality of Life after Extraction of Impacted Wisdom Teeth

Objective To investigate the effect of dexamethasone addition in block anesthesia before surgery on postoperative quality of life after extraction of impacted mandibular wisdom teeth. Methods Using a random number table, 120 cases of patients were divided into experimental and control groups, each group contains 60 cases whose mandibular wis?dom tooth will be extracted. Before the surgery, the experimental group patients perform local block anesthetic injection with 3 mL out of 5 mL mixture of 2%lidocaine 4 mL+dexamethasone 1 mL (5 mg) to block nerve. The control group was given block anesthesia of inferior alveolar nerve with 3 mL of 2%lidocaine block anesthesia alone. Improved postoperative symp?toms severity scale (PoSSe) was used to evaluate patients’quality of life after tooth extraction comprehensively and quantita?tively by analysis and comparison of the two sets of scores. Results The experimental group’s PoSSe scores were 18.059 1 ± 13.210 9, which was lower compared with control group (27.819 3 ± 15.716 9). In experimental group, diet, language, swelling, pain and daily points were lower than the control group (P<0.05). Conclusion Topical application of dexamethasone in mandibular wisdom tooth extraction reduces postoperative adverse reactions in a certain degree and im?prove postoperative quality of life.
9.Transplantation of marrow mesenchymal stem cells through renal artery in repair of acute tubular necrosis in nude mice

Objective:To investigate whether transplantation of mesenchymal stem cells(MSCs)through renal arteries can protect kidney from acute tubular necrosis(ATN),so as to lay a foundation for MSC transplantation in treatment of ATN.Methods:Five-week-old nude mice were randomly divided into three groups:normal control group(n=10),acute tubular necrosis(ATN)model group without(n=10)and with MSCs treatment group(n=11).ATN nude mice were induced with 50% glycerin.MSCs labeled with enhanced green fluorescent proteins(EGFP)were injected into kidney through renal arteries.Serum creatinine was determined in all groups and pathological changes of renal tissues were detected using H-E staining.The amount and distribution of the EGFP-marked MSCs in renal tissues were determined with fluorescence microscope.Results:Degeneration and exfoliation of renal tubular epithelial cells,and even renal tubular tamponade with cast-off cells were observed in the ATN group;these pathological changes were mainly located at renal cortex and juncture of renal cortex and medulla.The damages were greatly alleviated in the ATN+MSCs transplantation group,with no swelling of epithelial cells,nuclear condensation or edema.Fourteen days after MSCs transplantation,EGFP positive cells were increased in renal tubules of recipient mice.Conclusion:The MSCs transplantation via renal artery can locate in renal tubular epithelium,and promote the repair of injured renal tubular epithelial cells.
10.Construction of a siRNA expression plasmid targeting CXCR4 and its effect on the invasion capability of the breast cancer cells in vitro

Objective To construct and identify the siRNA eukaryotic expression vector targeting gene CXC chemokine receptor-4 and explore its role in invasion process of breast cancer cells in vitro.Methods Two siRNAs were designed and synthesized according to the coding sequence of CXCR4 gene and cloned into eukaryotic expression plasmid pGE-1-U6/kna.The constructed CXCR4-siRNA expression vector was transfected into MDA-MB-231 cells by liposome.Western blot was used to evaluate the suppression of CXCR4 expression in different groups.The invasion and migration of MDA-MB-231 cells were evaluated by cell invasion assay in vitro.Results Enzyme digestion and DNA sequencing confirmed that the CXCR4-siRNA expression vector was successfully constructed.After transfection,the CXCR4-siRNA obviously suppressed the expression of CXCR4 compared with control groups and the ability of cell migration was decreased markedly.Conclusion CXCR4-siRNA expression vector can effectively suppress CXCR4 expression in the breast cancer cells and decrease potential of cell invasion,which may provide a novel strategy for gene therapy of breast cancer metastasis.
