3. Plasma level of salusin-α in patients with coronary heart disease and its clinical implications

Objective: To explore the relationship of plasma salusin-α level with the stability, severity, and other risks of coronary atherosclerosis. Methods: The patient group included 122 hospitalized patients with coronary artery disease (CAD), whose diagnoses were confirmed by coronary angiography (CAG). The CAD group was further divided into subgroups according to the clinical types, the number of diseased coronary branches, and Gensini's scores. Control group inlcuded 60 healthy subjects who underwent physical examination in our hospital. Salusin-α level and other general biochemical indicators were determined, and the general clinical data were obtained before CAG in all subjects. Results: The peripheral blood salusin-α level in CAD patients was significantly lower than that in the controls ([0.50±0.18] ng/ml vs [0.69±0.23 ng/ml], P<0.01), and that in the acute coronary syndrome patients was significantly lower than that in the stable angina pectoris patients ([0.46±0.17] ng/ml vs [0.56±0.19] ng/ml, P<0.01). Salusin-α levels were not significantly different between patients with different Gensini's scores or between patients with 1, 2 and 3 diseased branches. Conclusion: Peripheral blood salusin-α level might be associated with stability of the CAD, but not with the severity of coronary atherosclerosis.

9.Investigation on Levels of Polychlorinated Dibenzo-p-dioxins,Polychlorinated Dibenzo-p-furan and Polychlorinated Biphenyl in Milk in a City

Objective To know the levels of polychlorinated dibenzo-p-dioxins(PCDDs),polychlorinated dibenzo-p-furan (PCDFs)and polychlorinated biphenyl(PCBs)in milk and milk powder.Methods From Feb.2007 to Jun.2007,the milk and milk powder samples were collected and PCDDs,PCDFs and PCBs were extracted from the samples by Soxhlet extraction,cleaned up by FMS and quantified by HRGC-HRMS,using isotope dilution methodology.Results PCDDrFs and PCBs were detected in all samples.The mean levels of PCDD/Fs and dl-PCBs TEQ in the milk powder were 0.43 pg/g lipid(median:0.34 pg/g lipid),and in the packed milk were 3.83pg/g)lipid(median:2.04 pg/g lipid).The mean levels of PCDD/Fs and dl-PCBs TEQ in all samples were 2.13 pg/g lipid(median:0.815 pg/g lipid).The levels of PCDD/Fs and dioxin-like PCBs in the detected samples were far below the limitation of EU except of two milk samples,and the levels of PCDD/Fs and dioxin-like PCBs were higher in the packed milk than those in the milk powder.Conclusion Some of the milk and milk powder in the investigated city has been polluted by polychlorinated dibenzo-p-dioxins(PCDDs),polychlorinated dibenzo-p-furan(PCDFs)and polychlorinated biphenyl(PCBs).
10.RAPID DETERMINATION OF GLUTAMATE DECARBOXYLASE ACTIVITY FROM LACTIC ACID BACTERIA BY SPECTROMETRIC METHOD AND ITS APPLICATIONS

In this paper, a colorimetric method with high sensitivity, reproducibility and low cost for determining glutamate decarboxylase (GAD) activity was developed based on Berthelot reaction. The optimum substrate system is 0.2 mol/L MacIlvaine buffer pH4.7, containing 0.1 mmol/L pyridoxal phosphate (PLP) and 10 mmol/L L-monosodium glutamate (L-MSG). The reaction mixture consisted of 200 ?L substrate solution and 1~100 ?L enzyme preparation from lactic acid bacteria and was incubated at 30℃. The enzymatic reaction was terminated by immersion in ice-water and addition of 200 ?L 0.2 mol/L sodium borate buffer, pH9.0, then 1 mL 6% phenol solution and 400 ?L sodium hypochloride were added. Color development was carried out in boiling water for 10 min then immediately put in ice-water bath for 20 min. The optical density is read at 630 nm. ?-aminobutyric acid (GABA) produced was calculated using the standard curve. Applications of the method in GAD studies were discussed.
