1.Casticin inhibits proliferation and invasion of breast cancer MCF-7 cells

Objective To investigate the effects of casticin on proliferation and invasion on human breast cancer MCF-7 cells.Methods MTT assay and invasion assay were performed in MCF-7 cells treated with casticin,respectively.The expression of Bax,Caspase-3,MMP-2 and MMP-9 was analyzed by RT-PCR and Western blot.Results Casticin inhibited MCF-7 cells proliferation and invasion in a dose-dependent manner (P < 0.05).Additionally,Bax and Caspase-3 expression was increased in MCF-7 cells treated with casticin,whereas MMP-2 and MMP-9 were down-regulated.Conclusion Casticin may play a critical role in inhibiting breast tumor cell proliferation and invasion,reinforcing its potential in clinical application.
2.Clinical evaluation of ¹⁸F-FDG-SPECT/CT imaging in diagnosis and staging of patients with suspected lung cancer.

BACKGROUNDCorrect diagnosis is very important to patients with suspected lung cancer. According to the higher ¹⁸F-FDG intake of tumor tissues much more than normal tissues, this study evaluates the clinical value of ¹⁸F-FDG dual-head coincidence SPECT/CT in diagnosis of lung cancer.
METHODSSixty-one patients with lung mass underwent ¹⁸F-FDG dual-head coincidence SPECT/CT imaging and thoracic CT imaging before operation. These imageological results were compared with histological examination of lung mass.
RESULTSThe sensitivity, specificity and accuracy of ¹⁸F-FDG-SPECT/CT in differentiating malignant and benign lesions were 82%, 87%, 84%, respectively. Compared with thoracic CT imaging, diagnostic rate of mediastinal lymphatic metastasis was significantly higher by ¹⁸F-FDG-SPECT/CT (52% vs 81%, P < 0.05) .
CONCLUSIONSCombined functional-anatomical image of ¹⁸F-FDG-SPECT/CT has potential to improve staging and localizing procedures of lung cancer. It is an excellent modality in diagnosis and differential diagnosis of lung cancer.
3.Explore the expression of FHIT,WWOX and MDR1 gene in nasopharyngeal carcinoma

Objective To investigate the expression of FHIT,WWOX and MDR1 gene in nasopharyngeal carcinoma and FHIT and WWOX mechanism of inactivation.Methods Real-time PCR was used to test FHIT,WWOX and MDR1 gene′s mRNA expression in 89 nasopharyngeal carcinoma patients (experimental group) and 61 inflammatory patients (control group).Q-MSP was used to test the FHIT and WWOX promoter methylation status.Denatured polyacrylamide gel electrophoresis was used to test the LOH of FHIT and WWOX gene.Results (1)The three genes′ mRNA expression were different between experimental group and control group (P<0.05).After grouped according to the histological type and clinical stages,the expression of FHIT and WWOX mRNA between the patients with serious illness or poorly differentiated squamous cell carcinoma and mild cases or highly differentiated squamous cell carcinoma were significantly different in the experimental group,the difference is statistically significant (P<0.05)Meanwhile,the FHIT and WWOX mRNA expression had statistical association with the clinical stage and histological type(r=-0.731,P=0.000;r=-0.816,P=0.000;r=-0.626,P=0.000;r=-0.536,P=0.001).The MDR1 mRNA expression was different between poorly and highly differentiated squamous cell carcinoma (P=0.021),which was statistical associated with the histological type (r=-0.697,P<0.001).(2)The degrees of FHIT and WWOX promoter methylation in the experimental group was higher than those in the control group,the difference was statistically significant (P<0.05);Also,the expression of FHIT and WWOX mRNA were closely related to the degree of promoter methylation(r=-0.689,P=0.000;r=-0.594,P=0.000).(3) In the experimental group,there were 39 cases (43.8%) of LOH in the FHIT gene,and 42 cases (47.2%)of the WWOX genes were significantly higher than those in the control group (4.9% and 3.3%),the difference was statistically significant (P<0.05).The FHIT and WWOX gene mRNA were negatively correlated with the loss of heterozygosity(r=-0.239,P=0.049;r=-0.364,P=0.013).Conclusion Promoter methylation is the main reason for the down-regulation of FHIT gene and WWOX gene expression in nasopharyngeal carcinoma patients,which may be the main reason for the occurrence and development of nasopharyngeal carcinoma.The higher expression of MDR 1 mRNA is statistical association with the histological type.
4.Vitreous surgery for macular holes

Objective To review the anatomic and visual outcomes of a series of patients with macular holes who underwent vitreous surgery without adjunctive therapy.Methods We performed a retrospective review of 16 patients who underwent vitrectomy, fluid-gas exchange and perflu oropropane(C3 F8) gas temponade for 14 eyes with idiopathic and 2 eyes with traumatic macular holes.Results Anatomical closure of the macular holes was achieved in 15(93%) of 16 eyes with an average follow-up of 7 months. Only one eye needed performing second surgery. Three(19%) eyes achieved postoperative visual acuity of 0.4, four (25%)eyes 0.2~0.3,and nine(56%)eyes 0.1 or worse. Conclusions Vitreous surgery without adjunctive therapy with intraocular gas tamponade and postoperative posturing is a well tolerated and effective intervention for idiopathic or some traumatic macular holes. Anatomical closure of the macular hole is associated with a significant improvement in visual acuity in most eyes.
5.Problems and countermeasures of clinical practice teaching management

This paper aims to analyze the current situation and problems as to the teaching management during clinical training of full-time clinical major students, and put forward the corresponding countermeasures . The clinical practice teaching management and the clinical intern training quality can be significantly improved by establishing normative practice teaching management sys-tem, innovating teaching management mechanism, and correct treatment of the contradiction between clin-ical practice, employment and taking part in the entrance exams for postgraduate schools.

Objective To explore the effect of dimethylformamide(DMF)on the histological structure and enzymes activity in the testis of mice.Methods The KM male mice were treated with DMF by gavage at the doses of 0,0.5,1 and 2 g/kg respectively, once a day,for 30 consecutive days.On day 31,the mice blood samples were collected through eyes and then the mice were killed. Objective To investigate spinemagnetic resonance imaging(MRI) findings in patients with axial spondyloarthritis (SpA) and to analyze the correlation between imaging and clinical manifestation.Methods The clinical data of patients with axial SpA were recorded.All patients underwent whole spine MRI scanning.The MRI findings of spinal involvement were explored.Moreover,the correlation between lesions in spinal MRI and Bath Ankylosing Spondylitis Disease Activity Index(BASDAI),Bath Ankylosing Spondylitis Functional Index (BASFI),Bath Ankylosing Spondylitis Metrology Index (BASMI),nocturnal pain visual analogue scale (VAS) score,back pain VAS score,global disease activity VAS score,Ankylosing Spondylitis Disease Activity score (ASDAS),erythrocyte sedimentation rate,C-reactive protein was analyzed.Results Thirty-three patients with axial SpA were included in this study.The image abnormalities of the spine were confirmed in 29 axial SpA patients by MRI,including Romanus lesion,Andersson lesion,the inflammation of facet joints and syndesmophyte.In correlation study,BASMI was positively correlated to the numbers of acute Romanus lesions,chronic Romanus lesions,chronic Andersson lesions and whole spinal lesions(r =0.440,P<0.05; r =0.483,P<0.05; r=0.421,P<0.05; r=0.589,P < 0.05 respectively).There was a statistically significant correlation between chronic Andersson lesions and BASFI(r =0.392,P <0.05).But no significant associations were found between MRI lesions and other clinical findings.Conclusions MRI lesions in axial SpA were associated with findings reflecting the spinal function,which can better guide the clinical treatment.
8.Comparison of planning parameter selection for volumetric modulated arc therapy of nasopharyngeal carcinoma in two different treatment planning systems

Objective To investigate the impact of planning parameter settings on plan quality and delivery efficiency of VMAT for nasopharyngeal carcinoma with two treatment planning systems (TPS),as references for clinic plan optimization. Methods 25 patients with nasopharyngeal carcinoma were selected and planned for SIB?VMAT treatment. The same planning aims were used in the two kinds of TPS ( TPS?1 and TPS?2). Multiple planning parameters were set for plan optimization. Dose distribution to the target volumes and organs at risk,monitor unit ( MU) and delivery time were compared. Paired t?test or one?way ANOVA was used for the data which was in accordance to normal distribution;otherwise, nonparametric Wilcoxon signed rank test or nonparametric Friedman test was used. Results More segments lead to better plan quality and less MU but longer delivery time ( Minor impact was observed when segment number was larger than 120) in TPS?1,while it had little impact on both plan quality and delivery efficiency in TPS?2. Comparing to single?arc plans,dual?arc VMAT achieved no significant benefit in plan quality but had more MU and longer delivery time in TPS?1 ( P= 0?000 ) . However, dual?arc VMAT plans had better dose distribution in TPS?2, decreased the maximum and mean dose for spinal cord in 3?9% and 13?7%respectively (P=0?000,0?000).Changing the settings of maximum or minimum dose rate did not affect the plan quality in both of the tested TPSs. Increasing the maximum or minimum dose rate reduced the delivery time but the latter increased the number of MU ( P=0?000,0?000) . Conclusions VMAT plan quality and delivery efficiency is affected significantly and differently by planning parameter settings for two TPSs. Trial test should be conducted for different TPS to determine the optimal parameter settings.

AIM: To study the effect and mechanism of chlorophyllin (CHL) inhibiting HT29 cells. METHODS: IC 50 value and growth curve of HT29 cells were detected with MTT method. Apoptosis was detected with Wright-Giemsa staining, FCM and DNA electrophoresis. Telomerase was detected by PCR-ELISA, and protein and mRNA expression of COX-2 gene were detected through RT-PCR and Western blot. RESULTS: CHL inhibited the growth of HT29 in a dose-dependent manner. CHL blocked HT29 cells in G 1 phase but did not induce apoptosis. Different concentration of CHL inhibits the expression of telomerase and COX-2 in HT29 cells. CONCLUSION: CHL inhibits the growth of HT29 cells by inhibiting the expression of telomerase and COX-2 and blocking cells in G 1 phase. [
