2.Study on the detection of RhD(+) red blood cells mixed in D-negative blood by flow cytometry

Objective To establish a method for detection of RhD(+) red blood cells mixed in D-negative blood by flow cytometry(FCM).Method RhD(+) and RhD(-) RBCs were mixed according to predefined ratios.Cells were indirectly labeled,with IgG anti-D labeled as the first antibody,and FITC-anti-IgG F(ab')2 as the second antibody.The percentage of RhD(+) RBCs was determined by FCM,and the best dosage of IgG anti-D was also defined.The ratio of red cells in the two groups,measured by FCM,was compared with the actual ratio.The consistency of method was also evaluated.Results The effective dosage of IgG anti-D was 1∶4,and 50?l/1?106 cells.When the actual percentages of RhD(+) cell among RhD(-) cells were 2.5%-0.312%,the correlation coefficient between the percentages measured by FCM and the actual percentages was 0.987.The same tubes,containing 10% and 2.5% RhD(+) RBCs,were each tested for 10 times,and their coefficient of variation were 3.4%,and 4.9%,respectively.Conclusion The method of quantifying the RhD(+) RBCs in D-negative blood by FCM is feasible and repeatable,which deserves a further clinical application.

4.Effect of Early Rehabilitation on Motor and Cognitive Function of Hemiplegia after Stroke

ObjectiveTo study the effect of early rehabilitation on disability of motion and cognition.Methods107 patients were randomly divided into two groups,rehabilitation group (51 cases) and control group (56 cases).Patients in rehabilitation group were given regular early rehabilitation,while all patients in both groups were given usual clinical treatment. The Barthel Index (BI), Fugl-Meyer Assessment (FMA), Mini-mental Status Examination (MMSE) were used to evaluate these two groups before and 3 months after treatment. ResultsAfter treatment, the scores of BI, FMA and MMSE were improved significantly(P<0.05) in rehabilitation group compared with that before treatment, so did that of BI, FMA in control group(P<0.05).The scores of BI、FMMS、MMSE in rehabilitation group improved significantly compared with that in control group(P<0.05).ConclusionEarly rehabilitation may obviously improve the motive and cognitive function and increase the quality of living of hemiplegia after stroke.
5.Enhancing special knowledge training in newly engaging-system nurses in laminar air flow ward and improving nursing quality

Objective To strengthen special knowledge training in newly engaging-system nurses in laminar air flow cleaning ward so as to improve nursing quality.Methods Those measures such as establishing prospective target,standardizing training content,enhancing clinic technician and theoretic knowledge tutoring by experienced nurses,paying attention to looking ward and proposing nursing questions,strengthening rehearsing in clinic and periodical examination were put forward so as to ensure all-sided training in newly engaging-system nurses in laminar air flow ward.Results The newly engaging-system nurses have poor speciality knowledge on laminar air flow word.Mter three months' training,their speciality knowledge had improved a lot,and the score on speciality knowledge was significantly higher than that of pre-training.Conclusions Speciality knowledge training should be strengthened in newly engaging-system nurses so as to ensure improvement of nursing quality.
6.IDENTIFICATION OF A NEW TYPE OF AMYLASE AND MUTAGENESIS OF STRAIN ZX99 SECRETING THE ENZYME FOR PRODUCTION OF ISOMALTOOLIGOSACCHARIDE

This paper reported a new type of amylase (neoamylase) secreted by a Bacillus strain ZX99. The enzyme was a kind of ectoenzyme that could catalyze starch into isomalto-oligosaccharide effectively, but could not act on pullulan as substrate. The strain Bacillus ZX99 was mutated by ultraviolet ray and a mutant strain BS3.232 was screened. The activity of the neoamylase produced from BS3.232 increased by 60% over that from ZX99 under the same conditions. The results of thin-layer chromatography of products from starch and pullulan catalyzed by the enzyme demonstrated that the enzyme was different from neopullulanase and can be used to produce isomaltooligosaccharide from starch, including isomaltose, panose, isomaltotriose, isomaltotetose.
7.Clinical study on treatment of chronic aplastic anemia assisted by TCM for reinforcing shen and activating blood circulation to dredge channels.

OBJECTIVETo observe the clinical efficacy of TCM for reinforcing Shen and activating blood circulation to dredge Channels in auxiliary treating chronic aplastic anemia (CAA), and to explore its mechanism.
METHODSSeventy-nine patients with CAA were randomly divided into the treated group treated with TCM plus western medicine and the control group treated with western medicine alone. The clinical efficacy was observed and levels of plasma sFas and sFasL before and after treatment were determined by ELISA.
RESULTSThe basic cure rate and total effective rate in the treated group were 37.50% and 87.50%; while in the control group were 20. 51% and 61.54% respectively. Comparison between the two groups showed significant difference (P < 0.01). The sFas level was markedly lower in both groups before treatment, but after treatment, it got elevated in the treated group significantly (P < 0.05), as compared with that in the control group, the difference was significant (P < 0.01).
CONCLUSIONTCM is effective in treating CAA with less toxic and side-effect, the mechanism might be the inhibition on over apoptosis of hematopoietic cells in CAA patients through regulating immune function of organism.

9.Spatial clustering analysis of Budd-Chiari syndrome in Heze City of Shandong Province

Objective To investigate the spatial distribution and clustering areas of Budd-Chiari syndrome in Heze City,Shandong Province,and to provide epidemiological information for further exploring the etiology and related risk factors of the disease.Methods Detailed residential addresses of 342 cases of patients (residents of Heze City) with diaphragm type Budd-Chiari syndrome diagnosed between 1995 and 2004 in Heze Municipal Hospital,Heze Shan County Central Hospital,Affiliated Hospital of Xuzhou Medical College,Shandong Provincial Hospital and Beijing Xuanwu Hospital were collected.Geographic information system (GIS) was used as a platform for data management and display.The nearest neighbor index,Ripley's K(d) function,Ripley's L(d) function and the nearest neighbor clustering method were applied to detect the spatial characters of Budd-Chiari syndrome in Heze City,Shandong Province.Crimestat 3.0 was used for spatial analysis.Results The nearest neighbor distance analysis showed that the nearest neighbor index was 0.6767 (Z =-11.4387,P < 0.01).That was an aggregation at the first-order spatial scale.Within the study area,the first clustering radius of Budd-Chiari syndrome was 6.66 km,and the first clustering strength was 5.40; the average radius of the strongest clustering area was 126.61 km,and the clustering strength was 12.52,while the biggest clustering radius was larger than 222 km.After corrected by population,the gathering strength was slightly higher than that before the correction.Ten first-order hot spots were formed,and 95% confidence interval aggregation number was 7,which meant the results were statistically significant(P < 0.05),main clustering areas are in Mudan District,Shan County and Juancheng.One secondorder hot spot was gathered based on the first-order hot spot.Conclusions Spatial distribution of Budd-Chiari syndrome in Heze City,Shandong Province has showed spatial aggregation and heterogeneity.This study has a great epidemiological significance for further exploring the cause of Budd-Chiari syndrome.
