Chinese Journal of Tissue Engineering Research 2014;(39):6246-6251
BACKGROUND:Recent advances in polylactic acid-polyethylene glycol (PLA-PEG) block copolymer and hydroxyapatite surface-grafted with poly(l-lactide) (HA-g-PLLA), including their synthesis and preparation, have been approved for application in bone tissue engineering. But they have stil many disadvantages. Combination of HA-g-PLLA and PEG-PLA could show a great potential for development in bone tissue engineering. OBJECTIVE:To investigate the effect of PLA-PEG/HA-g-PLLA composite fibrous material with different content of HA-g-PLLA on adhesion, multiplication and cellviability of osteoblasts. METHODS:Four fibrous materials of 5%PLA-PEG, 5%PLA-PEG/5%HA-g-PLLA, 5%PLA-PEG/10%HA-g-PLLA, 5%PLA-PEG/15%HA-g-PLLA were prepared using electrospinning technique and divided into four experimental groups. Mouse osteoblasts were inoculated on the materials. A statistical analysis was done based on results from celldyeing and alkaline phosphatase detection at certain time points to evaluate the effect of new materials on osteoblast adhesion, proliferation and viability. RESULTS AND CONCLUSION:In the experiment of cellmultiplication and the test of alkaline phosphatase activity, both absorbance values and activity of alkaline phosphatase from high to low were 5%PLA-PEG/10%HA-g-PLLA>5%PLA-PEG/15%HA-g-PLLA>5%PLA-PEG/5%HA-g-PLLA>5%HA-g-PLLA (P<0.05). The absorbance value of PLA-PEG/HA-g-PLLA roughly presented a waveform with the increasing addition of HA-g-PLLA. The effect of PLA-PEG/HA-g-PLLA composite fibrous material on adhesion, multiplication and osteogensis of osteoblasts increases with addition of HA-g-PLLA in a certain range.

Objective To investigate the clinical results of bilateral transpedicular kyphoplasty for the osteoporotic vertebral compression fractures with single balloon. Methods From March 2002 to February 2004, percutaneous bilateral transpedicular kyphoplasty with single balloon was performed in 15 patients, who presented with painful vertebral compression fractures. In the group, there were 6 vertebrae of 6 males and 11 vertebrae of 9 females, and the average age of the patients was 70.5 years with a range of 62 to 82 years. 17 osteoporotic vertebral compression fractures, which involved 8 T12, 7 L1, 1 T8 and 1 T11, were treated. Osteoporosis was identified in all cases by the measurement of lumbar bone mineral density (BMD) and the evaluation of radiographs. The average BMD was 0.603 g/cm2. CT scan showed the posterior vertebral wall intact in all patients, and MRI displayed low signal on T1WI and high signal on T2WI of the vertebral fractures. Results All patients experienced dramatic pain relief after the procedures. The mean pain score (VAS) decreased significantly from 8.3 preoperatively to 2.1 after operation. The mean vertebral body height restoration was 54.2%?34.1% on the anterior border, 60.5%?35.4% in the middle portion, and 40.7%?32.3% on the posterior border respectively. And the sagittal alignment was improved significantly, as the average correction of the Cobb's angle reached 9.5?. No cement leakage was found on X-ray, except one with less cement at the anterior vertebral edge, but no clinical symptom occurred. All the patients were available at the final follow-up, the mean period was 10.5 months (range, 2-48 months). Conclusion Bilateral transpedicular kyphoplasty with single balloon not only can provide effective pain relief and vertebral height restoration, but also can improve the sagittal alignment significantly. Its clinical outcome was satisfactory.

OBJECTIVETo observe the effect of external application of Zhuangshenling Recipe (ZR) combined with Western medicine (WM) on the heart function of chronic heart failure (CHF) patients of Xin-Shen yang deficiency, interior retention of water-fluid syndrome (XSYDIRWFS).
METHODSTotally 140 CHF patients of XSYDIRWFS were randomly assigned to two groups, the treatment group and the control group, 70 in each group. All patients received WM therapy. Those in the treatment group were applied with ZR at Xinshu (BL15) and Shenshu (BL23), while those in the control group were applied with placebos at Xinshu (BL15) and Shenshu (BL23). The therapeutic course for all was 12 weeks. The integrals of TCM syndrome, grading of cardiac function, brain natriuretic polypeptide (BNP), and 6 min walking distance were observed before and after treatment.
RESULTSAfter twelve weeks of treatment, the effective rate of improved grading of cardiac function, the total effective rate of TCM syndrome efficacy, and the BNP level were obviously better in the treatment group (P < 0.05). There was no statistical difference in 6 min walking distance between the two groups (P > 0.05).
CONCLUSIONExternal application of ZR combined with WM could improve the heart function of CHF patients of XSYDIRWFS.
