China Biotechnology 2006;0(12):-
Alliinase is immobilized on pre-activated chitosan microspheres by glutaraldehyde. The optimum immobilization conditions are as follows: glutaraldehyde concentration is 4%,alliinase concentration 20.2U,immobilization time 2h.For immobilized alliinase, the highest activity was allowed at pH 7.0 and temperature 35℃,and the Michaelis constant(Km)was disclosed to be 7.9mmol/L by Lineweaver-Burk plot, and after 10 times reuses the immobilized alliinase lost no more than 10% activity.

Chinese Medical Equipment Journal 2004;0(08):-
Information has become the soul of military affairs, campaign, combat support, and other various elements in the battlefield. In the particular war of 5.12 Wenchuan earthquake,in the face of sudden major geological disasters, sensitive information is the premise to effectively run the emergency response mechanism. The information collection, processing and determination permeate throughout the course of earthquake resistance & disaster relief and serve as the core in the effective commanding and coordination.

OBJECTIVETo investigate the estrogenic activity of icariin and genistein with estrogen-dependent human breast cancer (MCF-7) cells.
METHODMCF-7 cells were incubated with media containing 5% charcoal dextran-treated FBS in phenol red-free media for 48 h. CCK-8 kit was used to study the impact of defferent concentration of icariin and genistein on MCF-7 proliferation in vitro. Optimal concentration icariin and genistein were added into medium and total RNA was isolated after 12, 24, 36, 48 h. The gene expression of ERalpha, ERbeta, PS2, and PR were investigated by Real-time RT-PCR Total protein was also isolated and secretion of ERalpha, ERbeta, PS2, and PR were examined by Western blot.
RESULT10 micromol x L(-1) icariin and genistein could promote the proliferation of MCF-7 evidently. However, the ability of genistein to promote the proliferation was better than icariin. With the concentration of 10 micromol x L(-1), genistein group had a stronger expression of ERa, PS2 and PR mRNA levels than icariin while ERbetaexpression had no significant difference in two group. The same effects were detected by western blotting.
CONCLUSIONBoth genistein and icariin have a strong estrogen-like effect, but the estrogenic activity of genistein is stronger than icariin. It showed that the activity of icariin is stron-ger than genistein to promote ROB maturation. So it must be that icariin promotes the maturation of osteoblasts in vitro by a estogen-independent mechanism.

Objective To analyse the clinical characteristics, gene mutation and enzymatic activity of αgalactosidase A(α-GalA)in a 15-year-old male patient with typical Fabry disease, whose mother was without any clinical manifestations. Methods Clinical features and laboratory data were collected from the patient and his mother.Genomie DNA was extracted from peripheral blood of the patient.his mother, and a healthy control subject.Seven exons of the GLA gene were amplified by PCR.PCR products were purified.cloned into T vector, and then sequenced. The enzymatic activity of α-GalA Was measured by fluorimetrie substrate assay. Results DNA sequencing results showed that a missense mutation of 10036-10038delAAG in exon 7 WaS identified in the patient, resulting in the replacement of 374 lysine and 375 glyeine by arginine, which Was not previously reported. The patient Was a hemizygote with gene mutation, his mother WaS a heterozygote carrying gene mutation, and the healthy control without mutation.α-GalA enzymatic activity assay showed that the enzymatic activity of the patient with GLA gene mutation was only 50%of the healthy control subject, while the enzymatic activity of the patient's mother Was about 70%of the heahhy control SObject.Conclusiolls Detecting GLA gene mutation and α-GalA enzymatic activity in patients with Fabry disease who have been clinically diagnosed seelns to be helpful in finding other patients in the family and in further understanding the molecular pathogenesis of that disease.

Objective To observe the therapeutic efficacy of continuous arterial infusion chemotherapy combined with radiofrequency hyperthermia in the treatment of patients with advanced colorectal cancer. Methods A total of 21 advanced colorectal cancer patients with various distant metastasis were treated by the combined continuous intra arterial infusion chemotherapy and radiofrequency hyperthermia. For the chemotherapy, a dose of 200mg/m 2 surface body area of calcium folinate (CF) was given for 1 to 3 days. 80mg/m 2 of cisplation was infused intravenously for the first day and replaced by etoposide Vp 16, 60mg/m 2 for 1~3days in patients with renal dysfunction. For intra arterial infusion, a dose of 500mg/m 2 5 FU 2 was given for 72 hours. For patients with liver metastases, chemoembolization(ADM 30mg/m 2+MMC 6mg/m 2 mixed with ultra liquefied lipiodol) was carried out. Radiofrequency hyperthermia with a frequency of 41MHz was performed on the second day after chemotherapy. Results Response rates were assessed by CT scan and ultrasonography. The overall response rate of the cases was 66.67%. No serious side effect or complication was found in the course of chemotherapy. Local pain and lipid nodule were occasionally observed in some patients after hyperthermia. Conclusion Continuous intra arterial infusion chemotherapy in combination with radiofrequency hyperthermia is an useful and safe method for the treatment of patients with advanced colorectal cancer.

AIM:To observe the clinical effect of femtosecond laser assisted penetrating keratoplasty.
METHODS: Twenty-four cases ( 24 eyes ) with corneal lesions were performed with femtosecond laser assisted penetrating keratoplasty. Preoperative and postoperative endothelial cell density and visual quality were compared.RESULTS: One week after operation, corneal grafts were clear in 21 eyes (87. 5%), mild cloudy in 3 eyes (12.5%);visual acuity ≥0. 5 in 18 eyes (75. 0%), 0. 2 ~0.4 in 6 eyes ( 25. 0%). After 3mo the mean corneal astigmatism was 2. 16±0. 21D ( range 2. 25 ~ 3. 09D). Compared to conventional penetrating keratoplasty which mean corneal astigmatism was average 3. 67±0. 38D after operation, there was significant difference between two groups ( P< 0. 05 ). There were significant differences between preoperative and postoperative visual acuity and astigmatism (both P<0. 05).
CONCLUSION: Femtosecond laser assisted penetrating corneal transplantation operation can improve patient's visual quality. And compared to traditional penetrating keratoplasty astigmatism decreased significantly, incision can be made in individual shape more precisely and neatly.
