Objective To study the structure and the use of the information sharing platform for long-distance medical information network in integrating long -distance medical information network, HIS, PACS system and other resources, thereby enhancing the quality of long -distance medical consultation. Methods The study was conducted by integrating hospital internal data resources, structuring information platform and standardizing consultation process. Results The information sharing platform was achieved based on long-distance medical information network, thus achieving the greatest degree of information sharing and raising the level of long-distance medical consultation.

Objective: To explicit whether miR-200b can suppress the proliferation and invasion by targeting CD133, and to explore a molecular mechanism that miR-200b plays a role in tumor suppression in glioma. Methods: The CD133 3'-untranslated region (3'-UTR) mRNA-luciferase reporter vector was constructed and the dual-luciferase reporter gene assay was employed to examine the effect of miR-200b on activity of luciferase. U251 cells were transfected with miR-200b mimics and CD133-small interfering RNA (siRNA) (as a positive control) by LipofectAMINE 2000, and the expressions level of CD133 protein was detected by Western blotting. The inhibition effects of CD133 on cell proliferation and invasion were observed after CD133 siRNA were transfected into U251 cells. U251-SC cell mammosphere assay was performed after cotransfection with miR-200b mimics and CD133 siRNA. Results: miR-200b could bind to the 3'-UTR of CD133 and inhibit the activity of luciferase. CD133 protein expression was significantly down-regulated when miR-200b was overexpressed in U251 cells. Overexpression of miR-200b inhibited the invasion of U251 cells. Overexpression of miR-200b antagonized a role of proliferation and invasion in U251 cells. Overexpression of miR-200b reduced mammosphere number and the size of glioma stem cells. Conclusion: miR-200b can suppress cell proliferation and invasion by targeting CD133 in glioma. Copyright © 2014 by TUMOR.

Objective:To understand the influence of neoadjuvant chemotherapy combined with psychological and ethical intervention on mood and life quality of patients with cancer. Methods: The 120 patients with locally advanced breast cancer were randomly divided into control group ( neoadjuvant chemotherapy group) and interven-tion group ( neoadjuvant chemotherapy combined with psychological ethics intervention group) , Chemotherapy and after chemotherapy respectively adopt Zung Self -Rating Depression Scale ( SDS ) , Zung Self -Rating Anxiety Scale (SAS) and Quality of Life Core Scale (QLQ-C30) as an investigative tool, mood and quality of life of the patients were assessed. The patients' mood and life quality were investigated. Results: Emotional state intervention group patients more significantly than control group patients improved (P<0. 01) and improve life quality. Con-clusions:Neoadjuvant chemotherapy for localized breast cancer patients with psychological ethics intervention can effectively reduce the patient's anxiety and depression, and improve the life quality of patients.

Purpose To Prepare anti- HIV-1 gp120 monoclonal antibodies and to identify the specificity of antibodies in order to provide technique for preparing HIV remedial antibodies. Methods The gene fragment of HIV-1 gp120 was connected to PEGX-4T-2 prokaryotic expressing vector. The vector was cut by enzyme. GST-HIV protein was expressed by E. coli XL1-blue. The BALB/c mice were immunized with purified GST-HIV antigen, and then the fusion of mice spleen cell and myeloma cell SP2/0 was executed as the routine cell-fusion technique. Positive cells were screened by Indirect ELISA. Immuno-blotting assay and Western blot identified the specificity of antibodies. Results External gene section from the recombinant plasmid by sequencing showed the same size of HIV-1 gp120 gene sequences. An external expressed protein band of 32 KD was obtained after purified protein SDS-PAGE electrophoresis. It indicated that six strains of hybridoma cells secreting special monoclone antibodies had been obtained. ELISA results showed that six strains monoclonal antibodies only reacted to HIV-1 gp120 antigen. Western blot results showed that a band with molecular weight 32 kDa was obtained, which could interact with HIV-1 gp120 monoclonal antibody. Conclusion Six strains of hybridoma cells secreting special monoclone antibodies had been obtained. The prepared monoclonal antibodies have established a basis for HIV remedial antibody.

OBJECTIVE To investigate the disinfectant and antibiotic resistance genotypes in 60 strains of Escherichia coli isolated from urine.METHODS Sixty strains of E.coli isolated from inpatients′ urine were collected from Jan 2006 to Oct 2008.Antibiotic susceptibility tests for fifteen antibiotics were performed by Kirby-Bauer method.And three kinds of disinfectant and antibiotic resistance genes(qacE△1,tehA,merA)were analyzed by polymerase chain reaction(PCR) and DNA sequencing.RESULTS More than 70.0% of the sixty strains of E.coli were susceptible to imipenem,piperacillin/tazobactam,cefoxitin,amikacin and gentamicin,and less than 50.0% were susceptible to the other ten antibiotics.There were 42 strains with qacE△1 gene(70.0%),10 strains with merA gene(16.7%) and all strains with tehA gene.The sequence of the first strain was different from those reported in GenBank,so it was a new subtype.CONCLUSIONS There are 70% of E.coli strains isolated from urine samples with qacE△1 gene.And disinfectant resistance may be one of the main factors for hospital infection in the future.

Methods Twenty patients (14 males and 6 females) aged (46.7 ?4.6) years, undergoing orthotopic liver transplantation without venovenous byp asss, were included in this study. A gastric tube technique was used to draw gas tric juice 3～5 mL to determine pHi and PiCO 2 before OLT, at 20 min preanhep atic phase, 30 min anhepatic phase, 30 min new hepatic phase and at 2 h and 2 4 h after operation. The arterial blood samples collected taken for the measure ments of PaCO 2, PO 2, pH, BE and HCO 3- at the same time point s as above. pHi was calculated through Henderson-Hasselbach equation. Objective To evaluate the effects of endogenous NO on the chemosensitivity of human breast cancer cell. Methods MCF-7 cells were cultured as monolayer, incubated with cytokine IL-1β. The pro-duction of NO was detected by NO assay. The expression of iNOS protein was measured by Western blotting. Establishing control group and experimental group, the chemosensitivity of MCF-7 cells incubated by L-NMMA and L-Arg to ADM and 5-Fu was studied by MTT assay. Results There was a positive correlation of dose-dependence between NO production and IL-1β concentration. MCF-7 cells expressed plenty of iNOS by induetion of IL-1β. There was no significant difference on iNOS whether L-NMMA and L-Arg existed or not. Incubating MCF-7 cells with 0. 5 μmol/L or 1 μmol/L ADM, the survival rate of experiment group was remarkablely decreased(P < 0.05) ; L-NMMA significantly increased survival rate of experiment group(P < O. 05) ; L-Arg decreased survival rate of experiment group(P < 0.05). Conclusion The induction of IL-113 in MCF-7 cells can increase the production of endogenous NO, which increases MCF-7 cells' sensitivity to chemotherapy.

Objective To observe the therapeutic efficacy of continuous arterial infusion chemotherapy combined with radiofrequency hyperthermia in the treatment of patients with advanced colorectal cancer. Methods A total of 21 advanced colorectal cancer patients with various distant metastasis were treated by the combined continuous intra arterial infusion chemotherapy and radiofrequency hyperthermia. For the chemotherapy, a dose of 200mg/m 2 surface body area of calcium folinate (CF) was given for 1 to 3 days. 80mg/m 2 of cisplation was infused intravenously for the first day and replaced by etoposide Vp 16, 60mg/m 2 for 1～3days in patients with renal dysfunction. For intra arterial infusion, a dose of 500mg/m 2 5 FU 2 was given for 72 hours. For patients with liver metastases, chemoembolization(ADM 30mg/m 2+MMC 6mg/m 2 mixed with ultra liquefied lipiodol) was carried out. Radiofrequency hyperthermia with a frequency of 41MHz was performed on the second day after chemotherapy. Results Response rates were assessed by CT scan and ultrasonography. The overall response rate of the cases was 66.67%. No serious side effect or complication was found in the course of chemotherapy. Local pain and lipid nodule were occasionally observed in some patients after hyperthermia. Conclusion Continuous intra arterial infusion chemotherapy in combination with radiofrequency hyperthermia is an useful and safe method for the treatment of patients with advanced colorectal cancer.

Objective To investigate the mechanism of extremely low frequency electromagnetic field (ELF-EMF) and X-ray irradiation on liver carcinoma cell lines BEL-7402. Methods Liver carcinoma cell lines BEL-7402 had been incubated with ELF-EMF (100 Hz, 0.7 mT, 30 min, 3 days) after X-ray irradiation at different doses (0, 2, 4, 6, 8,10 Gy). The cells were observed on morphologic changes with scanning electron microscope. Flow cytometry and gene microarray were used to investigate the mechanism of cell apeptosis. Results ELF-EMF plus X-ray induced apoptosis on BEL-7402 was observed under scanning electron microscope.When X-irradiation was 2, 4, 6, 8 and 10 Gy, the apoptosis ratios of combined group and only X-irradiation group were 10.0%, 14.5%, 4.3%, 5.1%, 7.1% and 0.1%, 8.1%, 0.1%, 0.4%, 2.2% (P < 0.05) on flow cytometry. The result of microarray indicated that 1465 genes were up-regulated and 1108 down*regulated in the ELF-EMF plus X-ray group in comparison with the control group. The change rates of 110 apoptosis related genes were above 2 times, which including 71 up-regulated and 39 down-regulated. Gene microarray showed that ELF-EMF and X-ray irradiation had a mainly effect on different gene of apoptosis paths (CDC25 and CHKI, ATM, p38, PTEN, p53, G1/S, Fas, G2/M, Cell Cycle, Apoptosis and Caspase). The same genes of ELF-EMF plus X-ray group were showed 13 in ELF-EMF group and 42 in X-ray group. Conclusions Apoptosis paths were significantly different between ELF-EMF and X-irradiation. ELF-EMF cooperates with X-irradiation on inducing BEL-7402 cell apoptosis.
