1.Effects of Lycii Cortex on express of PI3K/PKB in PCOS rats.

The effect of Lycii Cortex on the PCOS rat model and the mechanism of action were investigated in the present study. The PCOS rat model was induced with Poretsky methods. Then the rats were randomly divided into four groups: the model group, melbine group (0.45 g x kg(-1)), low (2.5 g x kg(-1) and high (10 g x kg(-1)) dosage group of Lycii Cortex. The animals were orally administrated with the drugs for 14 days. In addition, another control group was added in this study. The rats were weighted before and after drug treatment. After 14 days treatment, oestrous cycle of rats were detected; blood serum was separated to determine T and FINS and rat's uteri were isolated. The mRNA and protein (total and phosphorylated) expressions of PI3K and PKB in uteri were measured with Real-time RT-PCR and Western blot, respectively. Compared with the control rats, the body weight gain and serum level of T and FINS were significantly increased. While, the mRNA and protein (phosphorylated) levels of PI3K and PKB were markedly decreased in PCOS group. Lycii Cortex treatment significantly decreased the body weight gain and serum level of T and FINS in a dose-dependant manner. It also markedly increased the mRNA and protein (phosphorylated) expressions of PI3K and PKB. Meanwhile, the melbine treatment also showed the curative effect. Lycii Cortex can relieve the symptoms of PCOS and the mechanism might be related to PI3K/PKB pathway.

2.The electromyographic activity of masseter and temporal muscles in the subjects with oral-facial pain caused by experimental occlusal interference

Objective:To investigate the oral-maxillofacial pain and t he surface electromyographic (EMG) activity of masseter muscles and anterior and posterior portions of temporalis muscles after experimental interference. Methods:A cast onlay of 0.5 mm in height was placed on the right low er first molar and remained for three days in 10 adult volunteers with normal oc clusion and without any symptom of TMD. The oral-maxillofacial pain was continu ously scored on a visual analogue scale(VAS)by the subjects before and after t he placement of the onlay. 3.Laparoscopic rectopexy with Douglas pouch elevation associated with the procedure for prolapse and hemorrhoids (PPH) for complete rectal prolapse in adults

Objective To analyze postoperative effect of a new rectopexy technique with Douglas pouch elevation for complete rectal prolapse (CRP) in adults.Methods From January 2010 to May 2011, 52 CRP cases were treated by rectopexy with Douglas pouch elevation.In terms of different surgical techniques, patients were divided into two groups : Group A (n =28) received laparoscopic rectopexy with Douglas pouch elevation;and Group B (n =24) received laparoscopic rectopexy with Douglas pouch elevation combined with procedure for prolapse and hemorrhoids (PPH).Rectal prolapse, constipation, and fecal incontinence in the two groups were assessed respectively before surgery, and on the 6th, 12th, and 24th month after surgery, postoperative complications were evaluated with severe grading of surgical complications.Results Symptoms of rectal prolapse disappeared at half a year after surgery in all patients, and relapsed to different extent afterwards.However, two years after operation, the status of rectal prolapse in Group B tended to be stable.The trend of constipation after surgery was consistent with that of rectal prolapse.In addition, fecal continence improved gradually on half a year after surgery in the two groups and recovered to the optimal status in the first postoperative year.However, in the second year, the results of fecal incontinence reduced slightly in Groups A, and Group B became stable.Two years after surgery, four cases in Groups A relapsed while there was no recurrence in Group B.Difference of Grade I to Grade Ⅲ complications among the two groups was statistically insignificant (x2 =0.05, P > 0.05).Conclusions The clinical effect of laparoscopic rectopexy with Douglas pouch elevation associated with the procedure for prolapse and hemorrhoids (PPH) is better than that without PPH for female and male CRP patients with severe symptoms.

5.Reactive oxygenmetabolites measurement, establishment of a new method and its performance evaluation

Objective To establish a simple, cost-effective method to measure reactive oxygen metabolites ( ROMs) using automatic biochemical analyzer.Methods Serum samples were collected from 50 healthy individuals and 50 hospitalized patientsin Beijing Hospital from September 2013 to November 2013.By setting up and optimizing parameters on automatic biochemical analyzer, a new method of measuring ROMS was established with TBHP as calibrator.130 healthy non-smokers were recruited in Beijing Hospital from November 2013 to December 2013 to establish reference interval, including 73 males and 57 females.The average age was 68.8 ±12.5.According to CLSI documents, there was the establishment of the new method including precision, limit of blank ( LoB ) , limit of detection ( LoD ) , limit of quantitation (LoQ), linearity,interference testing, and reference interval.Results The intra assay imprecision was 1. 5%-4.1% and the total imprecision was 4.4%-9.9%.LoB was 0.85 mmol/L TBHP;LoD was 2. 7mmol/L TBHP;LoQ was 5.9mmol/L TBHP.The linearity was 5.9-600 mmol/L (R2 =0.995).When triglyceride≤28.58 mmol/L, hemoglobin≤173 g/L, vitamin C≤60 mg/dl and bilirubin≤73.8 μmol/L, the deviation was -7.6%, 4.6%, -98.9%, 19.1%respectively.The normal reference interval was 61.9 -88.1 mmol/L TBHP.Conclusions The newly-established method has good performances of precision,LOD and linearity, which can meet the clinical needs.The interference of triglyceride and hemoglobin is small, while vitamin C and bilirubin have stronger interference on measurement.( Chin J Lab Med,2015,38:163-167)
6.Construction of a recombinant BCG secreting BP26 and the effects of BP26 on CD4+ and CD8+ T cells in mice

Objective To develop a BP26 recombinant BCG (rBCG-BP26) vaccine,and to observe the effects of rBCG-BP26 on CD4+,CD8+ T cells in immunized mice.Methods The recombinant shuttle vector pMV261-Ag85B-BP26 was constructed by using traditional molecular biological technology.The recombinant strains were obtained by kanamycin resistance screening and PCR identification after electroporation.Western blotting was used to detect the expression of recombinant BP26 vaccine in immunized mice.Safety experiment was carried out in three different groups:the target experiment(rBCG-BP26) group,the positive control(BCG) group and the negative control(PBS) group,15 BALB/c mice in each group.Intradermal inoculations of 100 μl rBCG-BP26 [containing 106 colony forming units(CFU)],BCG,and PBS were carried out,respectively.Signs of mice in each group were observed.After immunization for 10,20,30,and 40 days,body weight was weighed,and tail blood was collected to observe the change of peripheral blood CD4+ and CD8+ T cells by flow cytometry.Results The rBCG-BP26 was successfully constructed.The expression of BP26 protein was detected in the liquid medium and the bacteria cells.The results of safety test analysis showed that there were no significant differences in signs and body weights(F=2.468,0.331,1.520,0.739,all P> 0.05),between PBS group[ (19.24 ± 0.54),(21.37 ± 0.66),(22.83 ± 0.62),(25.06 ± 0.37)g],BCG group[ (19.90 ± 0.02),(21.53 ± 1.57),(21.95 ± 0.55),(24.70 ± 0.39)g]and rBCG-BP26 group[ (19.16 ± 0.55 ),(20.89 ± 0.20),(22.15 ± 0.76),(24.60 ± 0.64)g].The results of flow cytometry showed that the percentages of CD4+ T cell level were lower in BCG group(26.70%,33.07%) and rBCG-BP26 group( 13.40%,26.70%) than that of the PBS group(33.85%,29.33%) and the values of CD4+/CD8+ T cells increased in rBCG-BP26 group (0.69%,1.27%,1.57%,1.70% ) 10,20 and 30 days after immunization.Conclusions Recombinant BCG-BP26 vaccine strain can express brucella BP26 protein efficiently.Furthermore,its virulence is mild,and it can activate CD4+,CD8+ T cells in the body.It can be used as one of candidate vaccine strain against brucellosis.
7.Clinical trial of lanthanum carbonate chewable tablets combination with calcium acetate tablets in the maintenace treatment of hemodialysis patients with hyperphosphatemia

Objective To evaluate the clinical efficacy of lanthanum carbonate combined with calcium acetate in the treatment of hemodialysis patients with hyperphosphatemia.Methods A total of 70 cases with hemodialysis were randomly divided into treatment group (n =35) and control group(n =35).Patients in control group were orally given calcium acetate 667 mg,tid.Patients in treatment group were orally given lantha num carbonate 250 mg,tid on the basis of control group.All patientswere treated for 12 weeks.The clinical effect and levels of phosphorus,cal-cium and intact parathyroid hormone (iPTH) in two groups were observed.Also,the adverse drug reactions in two groups were observed.Results After treatment,the total efficacy rates in treatment group and control group were 91.43% (32/35),71.43% (25/35),with significant difference (P < 0.05).Mter treatment,the levels of phosphorus,calcium and iPTH in treatment group were (1.73 ±0.16)mmol · L-1,(2.25 ±0.15) mmol · L-1,(189.60 ±68.49) pg · mL-1,had significant difference with those in control group,which were (1.88 ± 0.29)mmol · L-1,(2.45 ±0.19)mmol · L-1,(217.83 ±64.O0)pg · mL-1 (P < 0.05).There were 2 cases of nausea and vomiting in treatment group,with the incidence of 5.71% (2/35 cases).There was 1 case of abdominal pain in control group,with the incidence of 2.86% (1/35 cases,P > 0.05).Conclusion The clinical effect of lanthanum carbonate combined with calcium acetate is great in the treatment of hemodialysis patients with hyperphosphatemia.
9.Value of transthoracic echocardiography on guiding occlusion of soft-rim atrial septal defect

Objective To evaluate the feasibility and effect of transthoracic echocardiography(TTE)on guiding the occlusion of the soft-rim atrial septal defect(ASD).Methods Sixty two patients with the soft-rim ASD were enrolled.The size of ASD was measured and rim of ASD was observed by TTE on various views by using color Doppler system with tissue harmonic function before occlusion,and filmy rim of ASD with flapping which could not sustain occluder was eliminated.The size of occluder was selected by integratively judging the size of ASD and"sustainable diameter of ASD"The waist size of occluder was measured after releasing occluder and compared with the longest diameter of ASD and"sustainable diameter of ASD"measured by TTE.Results The longest diameter of ASD measured by TTE before occlusion was 11-35 mm[average(21.6±5.2)mm],the "sustainable diameter of ASD"was 15-37 mm[average(25.6±5.(J)mm],the size of selected occluder was 18-44mm[average(30.7±5.5)mini and the waist size of released occluder was 13-35 mm[average(24.2±5.6)mm].Fine correlation was existed between the longest diameter of ASD measured by TTE and the waist size of released occluder(r=0.86,P<0.000I).Morever,improved correlation was found between the"sustainable diameter of ASD"measured by TTE and the waist size of released occluder(r=0.89,P<0.0001).Occluder was firmly fixed without falling in all patients.Conclusions TTE with tissue harmonic function can be used to measure the size of soft-rim ASD and the"sustainable diameter of ASD".It is a feasible,and effective method on guiding occlusion of soft-rim ASD.
10.Virological impact of stalk region of neuraminidase in influenza A/Anhui/1/05 (H5N1) and A/Ohio/07/2009 (H1N1) viruses.

This study aims to investigate the virological impact of the stalk region and cysteine (C) in neuraminidase (NA) of influenza A/Anhui/1/05 (H5N1) and A/Ohio/07/2009 (H1N1) viruses. The NA of A/ Anhui/1/05 (H5N1), defined as AH N1, lacked 20 amino acids (including C, defined as s20) as compared with NA of A/Ohio/07/2009 (H1N1) (defined as 09N1). We deleted s20 of 09N1 to construct 09N1-s20, and inserted s20 into AH N1 to construct AH N1+s20. To investigate the impact of C on the biological function of NA, we deleted C in 09N1 to construct 09N1-C and inserted C into AH N1 to construct AH N1-C. The pseudo-type viral particle (pp) system was used to evaluate the impact of these mutants on virology. The combination of 09N1-C and 09H1 (defined as 09H1::09N1-C) showed an infectivity 8 times that of the wild type 09H1::09N1, while the infectivity of the combination of AH N1+C and AH H5 (defined as AH H5::AH N1+C) was much lower than that of the wild type AH H5::AH N1. The infectivity of the combination of 09N1-s20 and 09H1 (defined as 09H1::09N1-s20) was 4 times that of the wild type 09H1::09N1; the infectivity of the combination of AH N1+s20 and AH H5 (defined as AH H5:: AH N1+s20) was 1/7 that of the wild type AH H5::AH N1. The co-existence of 09N1-C and AH H5 displayed 6 times the infectivity of AH H5::09N1, while the infectivity of 09H1::AH N1+C was very low. Multimer analysis showed that in the wild type 09N1, the forms of NA were dimer > tetramer > monomer; the major component of NA in 09N1-C was monomer; in 09N1-s20, the forms of NA were monomer > dimer. AH N1 was mainly composed of monomer; in AH N1+s20, the forms of NA were dimer > monomer > tetramer; in AH N1+C, the forms of NA were dimer > tetramer. Deletion of C or s20 from 09N1 did not change the expression of NA. The study suggested that deletion of C from the stalk region of NA in A/Ohio/07/2009 (H1N1) increases infectivity. Insertion of C into NA's stalk region of A/ Anhui/1/05 (H5N1) significantly decreases infectivity. Cysteine deletion in the stalk region is important for the infectivity of A/Anhui/1/05 (H5N1) and A/Ohio/07/2009 (H1N1). It may interfere with the infectivity via changes in NA polymerization.
