5.Study on Molecular Characterization of Microbial Communities in Tibetan kefir Grain

The diversity in Tibetan kefir grains and dynamics of the microbial community during the fermentation of Tibetan kefir by PCR-DGGE fingerprinting technique were studied.The results showed that bacterial community of Tibetan kefir grains was more complex than that of yeast.The bacteria communities between different Tibetan kefir grains showed 78%~84% similarity, and yeast 80%~92%.Bands B, E and N of DGGE profiles of the microbial community during fermentation of Tibetan kefir were present throughout the fermentation.Analysis of sequence date showed the majority of the DGGE bands of bacteria community corresponded to LAB, and the most intense band (band E) was completely homology to Lactococcus lactis.
6.Expression and Purification of Receptor Tyrosine Kinase PDGFR?

A fusion expression vector pPIC3.5K-PDGFR? was constructed to express recombinant receptor tyrosine kinase PDGFR? and the right Pichia pastoris transformants were screened on his-deficient plates and YPD-G418 plates by turns after electroporation of strain GS115, a high yield strain named M3 was screened. The strain M3 was cultured in a 5 L fermentor and His-GFP-PDGFR? fusion protein was purified by Ni2+ chelating affinity chromatography. One distinct peak was obtained after elution with 250 mmol/L imidazole. Fusion protein was proved to be 90.08 kD by western blotting, and have tyrosine kinase activity by ELISA. Results showed that the receptor tyrosine kinase PDGFR? was successfully expressed in P. pastoris and could be used as a target for small molecule selective inhibitors screening.
7.Clinical effects of arthroscopic-assisted allograft reconstruction of anterior cruciate ligament injuries

Objective To determine 2-4 year intermediate-term clinical effects of deep-freezing bone-ACL-bone allograft plus endobutton plate fixation in reconstruction anterior cruciate ligament(ACL) injuries after ACL injury and estimate the feasibility and validity of the technique.Methods We car- ried out a retrospective study on 15 cases with ACL injury of lateral knee joint reconstructed with deep- freezing bone-ACL-bone allograft plus endobutton plate fixation method from September 1999 to October 2002.The average follow up was 36.9 months.The myodynamic recovery,muscular activity,Rom Lach- man test,Pivot shift test,Lysholm score and X-ray of tunnels enlargement at intermediate term were com- pared with normal ones.Results The circumference of thigh was(0.976?0.119)cm shorter than that of the normal side.Extension limitation of less than 3?was found in 12 cases and knee flexion limita- tion for below 5?in 13.Lachman test showed negative results.Lyshohn score was increased from preoper- ative 66.2?4.6 to postoperative 89.4?3.2.X-ray showed no tunnel enlargement.Conclusions Re- construction of ACL with deep-freezing bone-ACL-bone allograft plus endobutton plate fixation under ar- throscopy can attain equal length and anatomic reconstruction of ACL.Deep-freezing bone-ACL-bone is beneficial to functional recovery of ACL.
8.Comparison of Clinical Training Outcomes between the Synthetic and Traditional Teaching Modes

Objective To evaluate the clinical training outcomes from two clinical practice modes for undergraduate students. Methods Total two grades 2009 and 2010,150 undergraduate students were randomly divided into two groups: 69 students in group A and 81 students in group B. The synthetic teaching mode was applied in group A,and group B received the traditional teaching mode. The Objective Structure Clinical Examination (OSCE)and a written comprehensive examination were used to evaluate the training outcomes by T test analyzed by SPSS 17.0(α= 0.05). Results The theoretical level of 2009 grade students was better than that of 2010 grade (P < 0.05). The clinical practice skills of the two groups had no significant difference(P > 0.05). There were no significant differences between two groups in theoretical and clinical training(P > 0.05). Comparing with the 2010 grade,the theoretical level of the 2009 was better,but the clinical skill level was weaker. Conclusion Synthetic teaching mode and traditional teaching mode both achieved good outcomes. In order to obtain better comprehensive progresses,the two teaching modes need to be combined. The orientation by following the requirements of oral practicing qualification examination may enhance the clinical skills of the students.
9.Outcomes of decompressive craniectomy for severe traumatic brain injury in children

ObjectiveTo evaluate the mortality and morbility of children with severe traumatic brain injury (sTBI) following treatment with decompressive craniectomy and further analyze its long-term outcomes.Methods Seventeen children with sTBI undergone decompressive craniectomy between 2004 and 2010 were retrospectively studied.Quality of life of the patients who survived the operation was assessed by using the King' s outcome scale for childhood head injury (KOSCHI).ResultsOf 17 children with sTBI,the mean preoperative Glasgow Coma Scale (GCS) score was 5.27.Five children (29%) died postoperatively,of whom three children were died of cerebral infarction.Twelve children who survived the operation were followed up for average 4.6 years,which showed the mean KOSCHI score of 4.75.Among the 12 survivors,five patients (42%) experienced posttraumatic shunt-dependent hydrocephalus and four (33%) suffered ipsilateral and/or contralateral hygroma.ConclusionsAlthough a high mortality rate is observed in the children with sTBI after decompressive craniectomy,the survived patients have satisfactory outcomes. Posttraumatic hydrocephalus and hygroma are two common complications after decompressive craniectomy for children with sTBI.
