In this study, nattokinase gene was amplified by PCR using bacillus subtilis chromosomal DNA as template and cloned into expressed vector pBV220. After transforming recombinant plasmid into E.coli HB101, the recombinant strain was yielded. It was proved that expression products was secretive and expression protein was 12% of total cell protein by SDS-PAGE. Optimum culture time and inducing time was determined as 6h and 5h respectively. The plasmid stability studies showed that recombinant plasmid has excellent segregational stability but the structural stability was not good in the host cell.
3.Clinical efficacy evaluation of needle-knife for lumber disc herniation based on surface electromyography signals.

OBJECTIVETo apply needle-knife to treat lumber disc herniation (LDH) and surface electromyography were used to analyze biomechanical characteristic of patient's lumber muscle to make a comprehensive evaluation on its efficacy.
METHODSThirty patients who met the inclusive criteria were selected and treated with needle-knife, once a week for 2 weeks. Visual analogue scale (VAS), ASLR and JOA score before and after treatment were observed. Surface electromyography was applied to test the surface electromyography signals. AEMG, MFs and MPF were calculated before and after the treatment.
RESULTSAfter treatment, VAS was significantly reduced, ASLR, JOA, AEMG and MPF were obviously increased, and the absolute value of MFs was lowed (all P < 0.01).
CONCLUSIONThe needle-knife could significantly relieve lumbar muscle strength, muscle tone and muscle fatigue, improve in the imbalance of lumbar extensor muscle group, leading to the recovery of biomechanical characteristic, and the clinical efficacy is superior.

5.Effects of estradioi on radiation-induced apoptosis of bone marrow hematopoietic cells in mice

Objective:To investigate the effects of estradiol on ~(60)Co?-ray induced apoptosis of bone marrow hematopoietic cells of mice,and to discuss the related anti-irradiation mechanism.Methods:KM mice were randomly divided into 3 groups(15 mice/each group):control group(without radiation),pure radiation group and estradiol+radiation group(ER group).The pure radiation group was irradiated by 4.0 Gy?-ray at a dose rate of 1.15Gy/min;the ER group was administered with 0.1 mg estradiol(IM)at 10 days before 4.0 Gy?-ray radiation;and the control group received no special treatment.The apoptotic DNA segments of bone marrow hematopoietic cells were analyzed by DNA agarose gel electrophoresis;flow cytometry was used to examine the apoptosis rate of cells and expression of Fas and Bcl-2 at 4 h,8 h,and 12 h after irradiation.Results:Eight hours after radiation,the apoptotic DNA segments were obviously increased and apoptotic DNA ladder appeared,which was not seen in the other 2 groups.The apoptosis rate of bone marrow hematopoietic cells in ER group was significantly lower than that in the pure radiation group at 4,8,and 12 h after irradiation(P
6.Current status of health literacy among residents in Yixing City in 2017

Objective To ascertain the health literacy level and its influencing factors of Yixing residents, to find out regional characteristics and weak links in this regard, and to exercise the data analysis and processing capabilities of grassroots health education staff. Methods Multi-stage stratified cluster sampling methods were used to investigate 640 residents of 15-69 years old with the national residents' health literacy questionnaire.The results were analyzed by general description and chi-square test. Results In 2017, the health literacy level of residents in Yixing City was 20.36%.Of the three dimensions, the level of healthy lifestyle and behavior was the highest, while the level of basic knowledge and philosophy the lowest; the highest level of the 6 health problems was safety and first aid, and the lowest level was basic medical care, followed by chronic disease prevention.The factors affecting residents' health literacy were region, age and education level. Conclusion There is still space for improvement in the health literacy level of residents in Yixing City.It is necessary to intervene in groups with different characteristics in this regard.The whole society should work together to create a healthy cultural environment, continuously improving the health literacy of residents.
7.Effects of carbaryl on serum steroid hormone and the function of antioxidant system in female rats.

OBJECTIVETo investigate the effects of carbaryl on serum steroid hormone and function of antioxidant system in female Sprague Dawley rats.
METHODSCarbaryl was administrated to the adult female rats at doses of 0, 1.028, 5.140 and 25.704 mg x kg(-1) x d(-1) for 30 d. Vaginal smears of rats were taken to determine estrous cycle. Serum 17beta-estradiol (E(2)) and progesterone (P(4)) concentrations were measured by radioimmunoassay. The activities of superoxide dismutase (SOD) and glutathione-s-transferase (GST), and the levels of malondialdehyde (MDA) and glutathione (GSH) were measured by spectrophotometry.
RESULTSThe number of estrous cycle in exposed groups were obviously lower than in control group (P < 0.05, P < 0.01). Body weight gain in high dose group (25.704 mg x kg(-1) x d(-1)) was significantly lower than that in control. Meanwhile, the organ coefficient of ovary and uterus declined in a dose-dependent manner. Serum E(2) level [(19.93 +/- 2.21) nmol/L] in 25.704 mg group was lower than in control group [(28.76 +/- 6.12) nmol/L, P < 0.05], and P(4) level (1.21 +/- 0.40) nmol/L in 1.028 mg group was higher than that in control group [(0.63 +/- 0.39) nmol/L, P < 0.05]. The activity of SOD first reduced then rose in ovary, and first rose then reduced in serum. The contents of MDA increased in ovary, while decreased in the serum. GSH contents and GST activity increased in ovary, while in serum GSH contents decreased and GST activity first increased then decreased.
CONCLUSIONCarbaryl could disrupt estrous cycle and affect serum steroid hormone, and the function of antioxidant system in female SD rats.

9.Effect of flying burthen on blood lipid in civil aviation aircrew.

OBJECTIVETo investigate the effect of flying burthen on blood lipid level in civil aviation flight personnel.
METHODSThe level of total cholesterol (TC), triglyceride (TG) and high density lipoprotein cholesterol (HDL-C) were determined by kits. The relationship between the blood lipid levels and the flight personnel's flying hours, body weight index, smoking, drinking and sport were investigated.
RESULTSThe average annual flying hours of hyperlipidemia group [(561.14 +/- 234.90) h] was significantly higher than that of non-hyperlipidemia group [(500.62 +/- 243.65) h] (P < 0.05). The level of TG of the average annual flying hours more 800 h [(1.61 +/- 0.10) mmol/L] were significantly higher than that of less than 400 h group [(1.31 +/- 0.65) mmol/L]. The morbidity rate of hyperlipidemia among the age 30 to 39 years old flight personnel was the highest.
CONCLUSIONThe average annual flying hours is one of primary factors affecting blood lipid in flight personnel.

10.Effects of methyl cantharidimide tablets on urinary protein and enzymes in Beagle dogs.

OBJECTIVETo investigate the nephrotoxic effects of methyl cantharidimide tablets on urinary protein and enzymes in Beagle dogs.
METHODBeagle dogs were randomly divided into negative control group(blank tablet), methyl cantharidimide tablets group (6.11,12.21, 24.42 mg x kg(-1)), continuously 30 days of oral adminiStration, once a day. The drug and control group were collected and determined fresh urine in 1, 2, 3 and 4 weeks of the administration; Serum urea nitrogen (BUN), creatinine (Crea), total protein (TP) and albumin (ALB) as well as sodium, potassium, chloride electrolyte were determined on 15 and 30 days of the administration; Urine albumin (mAlb), kidney injury molecule-1 (KIM-1), neutrophil gelatinase-associated lipocalin( NGAL), N-acetyl-beta-D-glucosaminidase (NAG), clusterin, beta2-microglobulin (beta2-MG), alpha1-microglobulin (alpha1-MG), alanine aminopeptidase( AAP) and im- munoglobulins IgG were tested on 15 and 30 days of the administration.
RESULTCompared with the control group, urine protein and white blood cells was significantly increased in each dose group. On 15 days of the administration, mAlb were higher in each dose group, KIM-1, NGAL, clusterin, NAG and AAP were significantly higher in high-dose group, while the middle and low dose group had no significant difference, as well as blood SCr and BUN no obvious abnormalities. On 30 days, mAlb, KIM-1, clusterin, NAG, AAP were increased in each dose group, appearing dose-effect relationship, beta2-MG and NGAL levels were significantly increased in high-dose group. Contents above indicators were increased with significant dose and time relationship, and serum BUN, Scr were correlated, suggesting that urine mAlb, KIM-1, clusterin, NAG and AAP indicators that can sensitively respond the changes of proteins and enzymes in urine.
CONCLUSIONMethyl cantharidimide tablets has a renal toxicity, urine mAlb, KIM-1, clusterin, NAG and AAP can be used as the early nephrotoxic biomarkers of methyl cantharidimide tablets.
