1.SELECTION OF HIGH-YIELD CAROTENOID PRODUCING STRAIN BY LASER MUTAGENESIS OF PROTOPLAST OF PENICILLIUM SP. PT95

A mutant strain L05 was screened from its parent strain Penicillium sp. PT95 by laser irradiation of protoplast. When LOS strain was incubated in Czapek' s agar plates for 20 d, both the sclerotia biomass and carotenoid content accumulated in sclerotia increased significantly compared with that of PT95 strain, and the increase rate reached respectively 98.6% and 28.3% . The carotenoid yield of L05 strain reached 381ug/plate, which was 2.54 times higher than that of PT95. The character of both sclerotia and carotenoid high productivity remained stable after three times of subculture. No sectored colony appeared during subculture.
2.Modeling Traumatic Brain Injury Rats with Controlled Cortical Impact: Coma and Cognitive Dysfunction

Objective To establish the model of traumatic brain injury in rats with controlled cortical impact (CCI), and investigate the features of coma and cognition. Methods 40 male Sprague-Dawley rats were randomly divided into control group (n=10), sham group (n= 10) and CCI group (n=20). CCI group was impacted at bilateral frontal lobe with the velocity of 3.5 m/s, depth of 7 mm and dwell time of 250 ms. The time of consciousness loss was recorded, and all the rats were investigated with Morris Water Maze 10 and 20 days after operation. Results The time of consciousness loss was longer in the CCI group than in the control group and the sham group (P<0.001). The escape latency was longer in the CCI group than in the control group and the sham group (P<0.001), while the percent of time spend in goal quarter during probe trial was less (P<0.001), both 10 days and 20 days after operation. Conclusion CCI at bilateral frontal lobe of rats can establish the model of severe traumatic brain injury with coma and long-term cognitive dysfunction.

ObjectiveTo investigate the effects of sleep deprivation on expressions of Mir-132,mir-134 in the different regions of rat brain.MethodsAll the male SD rats were divided into control group ( normal sleep group),sleep deprivation (SD).The modified multiple platform method (MMPM) was used to establish sleep deprivation model.Mir-132,mir-134 level was detected by real time PCR.ResultsMir-132 were significantly increased in SD groups in hippocampus compared with the control groups ( 51.87 ± 8.13 vs 67.25 ± 7.59 ) (P <0.01 ).Mir-134 were significantly decreased in SD groups compared with the control groups( 1.82 ±0.15 vs 1.45± 0.12 )(P < 0.01 ).There were no statistically significant differences in cortex and thalamus (P > 0.05 ).Cortex mir-132 level in SD group and control group was 1.57 ±0.10,1.48 ±0.11 respectively,and it was 1.37 ±0.09,1.36 ±0.11 in thalamus;Cortex mir-134 level in SD group and control group was 98.26 ± 5.17,100.80 ±4.15respectively,and it was 97.56 ± 6.28,91.01 ± 4.07 in thalamus.ConclusionThe upregulation of mir-132 and downregulation of mir-134 implies that two miRNAs did opposite actions in the processes of sleep deprivation.This findings indicate that hippocampus mir-132,mir-134 levels in the SD rat may reflect associated depressive patho-physiological processes.
6.Dosimetric comparison between RapidArc and fixed gantry dynamic IMRT for central-type lung cancer radiotherapy

Objective To compare the dosimetric difference between RapidArc and fixed gantry angle dynamic IMRT (dIMRT) for central-type lung cancer radiotherapy. Methods Therapy for 10 patients previously treated with dIMRT was replanned with RapidArc. Dose prescription was 66 Gy/33 fraction. Comparative endpoints were planning target volume (PTV) dose, doses to surrounding structures,number of monitor units, and treatment delivery time. Results There was no significant dosimetric difference between RapidArc and dIMRT. Compared with dIMRT, RapidArc slightly elevated target volume dose, lung V5, V10. The average values of lung V20, V30 and heart V30 were larger in dIMRT than those in RapidArc. The number of monitor units was reduced by 32% and the treatment time by 66% in RapidArc.Conclusions Both RapidArc and dIMRT plans could meet the clinical therapy needs. RapidArc could achieve similar target coverage and sparing of organs at risk, with fewer monitor units and shorter delivery time than dIMRT.
7.STUDIES ON HARMFUL MICROBES IN RECIRCULATING WATER SYSTEM OF HEATING PIPELINE IN TAIYUAN

The microbial counts, type, as well as relationship between microbial counts and the temperature of water in reticulating water system of heating pipeline in Taiyuan were studied, which the main biofouling harmful microbes included slimeforming heterotrophic bacteria, sulfate reducing bacteria, iron bacteria and fungi, respectively. The results showed that the harmful microbes in water system were lower than that of control guideline during heating period, whereas the microbes were higher than that of control guideline, which would result in biofouling of water tube during non- heating period.
8.CT feature analisis of renal cell carcinoma:report of 300 cases

Objective To analyze the CT features of renal cell carcinoma,so as to improve the diagnostic accuracy of renal cell carcinoma.Methods Three hundred cases of renal cell carcinoma proved by pathology were examined by means of CT.There were 214 male and 86 female in this group.Their age ranged from 9 to 81 years,with a mean of 53.7 years.Their CT features were retrospectively reviewed. Results The masses were 1.5—16.0 cm(mean,4.8 cm)in greatest dimension,125 masses on left kidney and 175 masses on right kidney.According to WHO histological classification of tumours of the kidney in 2004,there were 238 cases of clear cell renal cell carcinoma,6 cases of multilocular clear cell renal cell carcinomas,23 cases of papillary renal cell carcinoma,14 cases of chromophobe renal cell carcinoma and 19 cases of renal cell carcinoma,unclassified.The above subtype of renal cell carcinoma demonstrated characteristic features.Clear cell renal cell carcinoma exhibited inhomogenous(due to hemorrhage,necrosis or cystic degeneration)and hypervaseular.Multilocular clear cell renal cell carcinoma presented as a multilocular cystic mass lacking an,expansile nodule,and with regular thin cyst wall and septa.Papillary renal cell carcinoma exhibited inhomogenous and hypovascular.Chromophobe renal cell carcinoma was relatively homogenous and hypovascular.Renal cell carcinoma,unclassified showed inhomogenous and hypervascular,and was more invading growth compared to clear cell renal cell carcinoma. Conclusion Common subtype of renal cell carcinoma demonstrated characteristic features in CT and it is helpful for differentiation.
9.Effect of static pressure on the expression of glial fibrillary acidic protein and heat shock protein 70 by the cultured retinal M?ller cells

Objective To observe the effects of static pressure on the number of cultured retinal M?ller glial cells(RMGC)and expression of glial fibrillary acidic protein(GFAP)and heat shock protein(HSP)70 by these cells.Design Experimental study. Participants Cultured rat RMGC.Methods Rat RMGCs were cultured and identified according to previous method described by Reichenbach.These cells were treated with different static pressures and divided into 4 groups:A(1.33kPa),B(2.67kPa),C(5.33kPa)and D(10.67 kPa)while the cells without treatment was as control group(NC).The morphologies of RMGC in these groups were observed under inverted phased contrast microscope,the number of RMGC counted with conservative method and the viability were studied with trypan blue staining.The expressions of GFAP and HSP70 in RMGCs were detected with the method of western blot.Main Outcome Measures The morphologies of RMGC,cell number,cell viability.Results There were pressure-dependent changes of RMGC number. The cell number of group C and D was less than that of group NC,A and B(P<0.01).High static pressure resulted directly in the decreased ratio of unstained RMGCs(P<0.01).The ratio of unstained RMGCs in group C and D was less than that in group NC,A and B(P<0.01).Many cells in group C and D were injured and the higher the pressure elevated,the more the degree of injury became.The expressions of GFAP and HSP70 in group NC were less than other pressure treated groups and the expression of GFAP in group C and D was higher than that in group A and B.There was no obvious difference between these pressure treated groups.Conclusions High static pressure could cause the injuries of RMGCs.The increased expression of GFAP and HSP70 in RMGC might be regarded as a sign of retinal injury response to high intraocular pressure.
10.Association study between polymorphism of tumour necrosis factor-α-863 and gout

Objective To investigate the association between the tumour necrosis factor-α-863(TNF-α-863) polymorphism and gout in Han population from the city of Tangshan.Methods We recruited 80 gout patients and 80 healthy individuals into this study.The polymorphisms of TNF-α-863 site were analyzed by polymerase chain reaction-ligase detection reaction(PCR-LDR).The frequencies of different TNF-α-863 genotypes/alleles were analyzed in the gout group and the control subjects.Results No significant differences were observed in the genotype frequencies(x2=2.8807,P=0.0897) and allelic frequencies(x2=4.2646,P=0.1187) of TNF-α-863 site in the comparison between gout and control groups.Conclusion The result of our study suggests that the polymorphism of TNF-α-863 site may not related to gout in Han population in Tangshan.
