4.Relationship Between Melatonin Hypo-secretion and Kidney-yin Deficiency Syndrome During Prepubescence

[Objective] To explore the relationship between neuroendocrine changes caused by melatonin (MT) hypo-secretion and kidney-in deficiency syvndrome during prepubescence. [Methods] Thirty SD rats after ablactation were randomly divided into normalgroup (group. A), model group (group B) and herbal medicine group (group C, treated with kidney-yin-nourishing ande(?)ficiency-fire-clearing herbs). Rats in groups B and C were exposed to light continuously for 30 days and simultaneously groupC was given kidney-yin-nourishing and deficiency-fire-clearing herbal medicine (13.3 g?kg-1?d-1, qd) for 30 days . After treatment, blood samples were collected at 0:00-2:00 o'clock to observe the changes of blood MT, testosterone (T), epinephrine (E) and norepinephrine (NE) levels. Objective To study the molecular mechanism of berberine induced apoptosis in chemoresistant EU-4 acute lymphocytic leukemia cells.Methods EU-4 cells were treated with 0,10 and 100 μmol/L berberine for 72 h.The apoptosis induced by berberine was detected by flow cytometry.The expression of Caspase-3,PARP and X-linked inhibitor of apoptosis protein (XIAP) were determined by Western bolt assay.Caspase-3 activity was measured using microplate reader.After transfected with XIAP siRNA,the apoptosis was detected by flow cytometry.Results After treated with 0,10 and 100 μmol/L berberine for 72 h,the apoptosis rates of EU-4 cells were (9.08±1.20) %,(22.36±2.16) % and (59.81±4.17) %,respectively.Berberine induced potent apoptosis in a dose-dependent manner.The apoptosis involved activation of Caspase-3.The Caspase-3 activities were 1.70±0.25,1.92±0.10 and 2.89±0.25,respectively.Berberine inhibited XIAP expression in a dose-and time-dependent manner (P < 0.05).Down-regulation of XIAP by siRNA increased apoptosis of EU-4 cells.The apoptosis rates were (9.23±1.66) % and (22.15±0.63) %.Conclusion Berberine could induce apoptosis of EU-4 cells,and inhibition of XIAP leading to Caspase-3 activation is responsible for the apoptotic effect on EU-4 cells.
6.Changes and significance of induced sputum and interleukin - 6, interleukin 8 in children with acute asthma

Objective To investigate the characteristic and clinical significance of airway inflammation in children with acute asth-ma. Methods Underwent sputum induction and sputum induction in children (n=34) with acute asthma was repented in recovered children ( n = 24).Induced sputum were also taken from 15 healthy children as controls.Total and differential cell counts were per-formed. Interleukin(IL)-8、IL-6 were measured,The relationship between inflammatory cells and IL-8、 IL-6、peak expiratory (PEF) were analyzed.Results The inflammatory cell infiltrate was mixed including eosmophilic granulocytes, neutrophils, and macrophages. They decreased significantly, but eosinophilic granulocyte remained a higher percentage compared with healthy subjects. There was low-er percentage of lymphocytes at acute exacerhation.Eosinophilic granulocytes were correlated with the degree of airflow obstruction. Levels of IL-8、IL-6 were elevated during the acute exacerbation and decreased at resolution.IL-8 was correlated significantly with neutrophils at acute exacerhation and resolution.IL-6 was correlated signifficantly with eosinophilic granulocytes at acute exacerbation. Conclusions Airway inflammation is chataeterized by infiltration of eosinophils, neutrophils and macrophages.IL-8、IL-6 possibly is the important cytokines of airway inflammation in children with acute asthma. 7.Papillary cystadenoma of the epididymis: a report of 2 cases and review of the literature.

OBJECTIVETo study the clinicopathological characteristics of papillary cystadenoma of the epididymis.
METHODSUsing routine pathology and immunohistochemistry, we observed the surgically obtained samples from 2 cases of papillary cystadenoma of the epididymis, analyzed their pathological features and clinical presentations, and reviewed the related literature.
RESULTSThe 2 patients were both adult males. The tumors typically manifested as painless swelling in the epididymis, with occasionally dull pain and tenesmus in 1 of the cases. Pathologically, the lesions exhibited three morphological features, i. e., dilated ducts and small cysts surrounded by fibrous connective tissue, adenoid papillary hyperplasia into the cysts embraced by fibrovascular stroma, and acidophil substance present in the cysts. Immunohistochemistry showed that the tumors were strongly positive for CK8/18, CK7, and EMA, but negative for CK20, CEA, MC, Calretenin, P53, P63, SMA, VHL, and CD10, with the positive rate of Ki-67 <1%. Follow-up visits revealed good prognosis in both cases.
CONCLUSIONPapillary cystadenoma of the epididymis is a rare benign tumor in the male urogenital system, which may be accompanied by the VHL syndrome. Surgery is the first choice for its treatment.

8.Experiment treatment of collagen-induced arthritis in rats with recombinant plasmid containing vasoactive intestinal peptide gene.

OBJECTIVETo investigate the therapeutical effect of recombinant plasmid containing vasoactive intestinal peptide gene (pcDNA3.1+/VIP) on collagen-induced arthritis (CIA) in rats.
METHODSThe experimental arthritis was induced by intradermal injection of bovine type II collagen emulsified in Freund's adjuvants in male SD rats. The rats then were given intra-articular injection with recombinant plasmid (pcDNA3.1+/VIP). The levels of serum TNF-alpha, IL-4 and IL-2 were detected by Avidin-Biotin Peroxdase Complex-enzyme-linked immunosorbent assay (ABC-ELISA) and the pathological changes in the joint of rats were observed.
RESULTHistological examination showed massive inflammatory infiltration in the joint with destruction of bone and cartilage, while the severity of pathological changes in synovia of VIP-treated rats was markedly reduced. Compared with normal group, the serum TNF-alpha, IL-2 levels of CIA rats were significantly increased (P <0.05) and IL-4 level was decreased (P<0.05). Compared with control and pcDNA3.1+ -treated CIA rats, serum TNF-alpha and IL-2 levels of pcDNA3.1+/VIP-treated rats were decreased and IL-4 level was increased (P<0.05).
CONCLUSIONRecombinant plasmid containing vasoactive intestinal peptide gene (pcDNA3.1+/VIP) can reduce the clinical and histological severity of established CIA and it might be a promising candidate for treatment of rheumatoid arthritis.

9.Construction of semantic type framework in top-level ontology of military medicine

Objective To explore the construction method of semantic types of military medical ontology and to establish a framework for the semantic types of the top-level ontology of military medicine in order to lay the foundation for the construction of military medical ontology and provide support for knowledge organization, knowledge disclosure and knowledge service of military medical information resources.Methods Using the method of document measurement, the core concept set of military medicine was constructed.On the basis of the framework of the top-level semantic types of Unified Medical Language System(UMLS), the semantic type framework was completed by the combination of bottom-up and top-down modes, using semantic analysis and expert consultation.Results By classifying the same kind of knowledge points of military medicine into one semantic type, the hierarchical structure of the semantic type framework of the top-level ontology of military medicine was obtained.Conclusion The proposed method is effective and practical for the construction of the semantic types of military medical ontology.The established semantic type framework of the top-level ontology of military medicine could reconstruct military medical knowledge and provide the basic framework for the establishment of the semantic relationships of the top-level ontology of military medicine.

I t can be utilizedto develop cell-catalyst, cell-adsorbent , live vaccine, biosensor and so on, and have a wide application perspective. But in our county, the microbe cell-surface engineering is studied just now. This review explain the development of the microbe cell surface engineering, overview the study and progress of microbe cell-surface engineering, and look this technology into the future.
