1.Coronary artery triple vessel lesion: clinical analysis of 68 cases

Objective To study the clinical characters of coronary artery triple vessel lesion, and to provide a theoretical basis for the therapy to the disease. Methods The clinical data of 68 cases of coronary artery triple vessel lesion (group A) and 71 cases of coronary artery of single or double vessel lesion (group B) were analyzed retrospectively. Results The related risk factors (hypertension, hyperlipidemia, diabetes and smoking) and the clustering of these factors were not significantly different between the two groups (P>0.05). While the positive rate of the exercise stress test existed remarkable difference between the two groups(P<0.01).The extreme differences between the two groups existed also in the cases of building collateral circulation (P<0.001), and in LVEF and LVEDP (P<0.0001) demonstrated by left ventricular angiography. The majority of the coronary artery lesion in group A was on level Ⅳ, and much more lesions of level Ⅲ and level Ⅳ occurred in group A than in group B. Most patients in group A accepted the coronary intervention, while after operation the angina pectoris emerged in a high rate, then the rebuilding of coronary circulation had to be done. Conclusions The best treatment for the patients with coronary artery triple vessel lesion should be the operation of coronary artery bypass graft (CABG), if it's possible.
2.Structural feature and biological function of PPP2R5C gene.

PPP2R5C is one of the members of regulatory subunits of protein phosphatase 2A (PP2A), which plays a critical role in cell proliferation, differentiation and transformation, based on its induction of dephosphorylation of P53 at various residues. Recently, it was characterized that the alteration of expression pattern of PPP2R5C is associated with cell malignant transformation, thus PPP2R5C was thought as a marker for progressive disease in B-CLL. In this article the gene structure and biological function of PPP2R5C as well as relation of PPP2R5C with genesis and development of cancer were discussed.

3.Substrate specificities of bile salt hydrolase 1 and its mutants from Lactobacillus salivarius.

In order to analyze the correlation between critical residues in the catalytic centre of BSH and the enzyme substrate specificity, seven mutants of Lactobacillus salivarius bile salt hydrolase (BSH1) were constructed by using the Escherichia coli pET-20b(+) gene expression system, rational design and site-directed mutagenesis. These BSH1 mutants exhibited different hydrolytic activities against various conjugated bile salts through substrate specificities comparison. Among the residues being tested, Cys2 and Thr264 were deduced as key sites for BSH1 to catalyze taurocholic acid and glycocholic acid, respectively. Moreover, Cys2 and Thr264 were important for keeping the catalytic activity of BSH1. The high conservative Cys2 was not the only active site, other mutant amino acid sites were possibly involved in substrate binding. These mutant residues might influence the space and shape of the substrate-binding pockets or the channel size for substrate passing through and entering active site of BSH1, thus, the hydrolytic activity of BSH1 was changed to different conjugated bile salt.

4.Down-regulation of HIV-1 Infection by Inhibition of the MAPK Signaling Pathway

The human immunodeficiency virus type 1(HIV-1)can interact with and exploit the host cellular machinery to replicate and propagate itself.Numerous studies have shown that the Mitogen-activated protein kinase(MAPK)signal pathway can positively regulate the replication of HIV-1,but exactly how each MAPK pathway affects HIV-1 infection and replication is not understood.In this study,we used the Extracellular signal-regulated kinase(ERK)pathway inhibitor,PD98059,the Jun N-terminal kinase(JNK)pathway inhibitor,SP600125,and the p38 pathway inhibitor,SB203580,to investigate the roles of these pathways in HIV-1replication.We found that application of PD98059 results in a strong VSV-G pseudotyped HIV-1NL4-3 luciferase reporter virus and HIV-1NL4-3 virus inhibition activity.In addition,SB203580 and SP600125 also elicited marked VSV-G pseudotyped HIV-1NL4-3 luciferase reporter virus inhibition activity but no HIV-1NL4-3 virus inhibition activity.We also found that SB203580 and SP600125 can enhance the HIV-1 inhibition activity of PD98059when cells were treated with all three MAPK pathway inhibitors in combination.Finally,we show that HIV-1virus inhibition activity of the MAPK pathway inhibitors was the result of the negative regulation of HIV-1 LTR promoter activity.
5.A Genetic AnaIysis of 800 Non-syndromic Deafness Patients from Shanxi Province

Objective The patients with non-syndromic deafness in Shanxi Province were retrospectively an_alyzed for the common deafness gene mutations and frequency of mutations carrying rate ,to understand the molecu_lar pathogenesis of deafness in Shanxi area .Methods Genomic DNAs of 800 patients of non -syndromic deafness within Shanxi were obtained from peripheral blood .Genes GJB2 ,GJB3 ,SLC26A4 and mitochondrial 12SrRNA 1494 and 1555 loci were sequenced after polymerase chain reaction (PCR) amplification and compared with the NCBI website for the analysis of the formation of mutations .ResuIts Among 800 patients ,353 cases (44 .13% ) showed detected deafness related mutations and the genetic etiology was found for 294 patients (36 .75% ) .Among them , 153 cases (19 .13% ) carried double allele mutations in the GJB2 gene .The most frequent mutation of GJB2 gene was 235delC ,and the carrying rate was 13 .5% (216/1 600) .The double mutant allele of SLC26A4 gene was detec_ted in 123 cases (15 .38% ) ,and the most common mutation was IVS7-2A>G ,identified in 7 .44% (119/1 600) of patients .Homogenic mitochondrial 12S rRNA 1494C> T mutation in one patient and 1555A> G mutation in 15 patients were detected .GJB3 gene c .538C > T heterozygous mutation was found in two patients .Altogether , 36 .75% (294/800) of patients with deafness were caused by gene mutations .ConcIusionThe data containing GJB2 gene and SLC26A4 gene carrying rate are consistent with the published data of non-syndromic deafness in the Northwest region of China ,but the carrying rate of mitochondrial gene mutations is lower than the average level of China .Our data show that the gene mutations contribute to 36 .75% of etiology in patients with deafness .This study reflects the importance of deafness related genes screening in Shanxi area for early diagnosis and genetic con_sultation .
6.Up-regulation of tim-3 expression contributes to development of burn-induced T cell immune suppression in mice.

T cell immunoglobulin and mucin domain 3 (Tim-3) is well known to negatively regulate T cells responses, but its role in burn-induced T cells immune suppression remains unclear. In the present study, in order to identify the relationship between Tim-3 expression and post-burn T cells immune suppression, C57BL/6 mice were subjected to burn injury or sham injury, and the liver and spleen were harvested at the day 1 after operation. The expression level of Tim-3 on hepatic or splenic T cells and the functional properties of Tim-3(+) T cells were evaluated. It was found burn injury induced dramatically elevated Tim-3 expression on both hepatic and splenic CD4(+) and CD8(+) T cells in contrast with the post-burn depletion of T cells. Furthermore, Tim-3 expression was correlated with the suppressive phenotype of T cells following burn injury, including increased expression of anti-inflammatory cytokine IL-10, decreased expression of pro-inflammatory cytokines IFN-γ and TNF-α, reduced T cell proliferation and elevated co-expression of Tim-3 and PD-1. Moreover, Tim-3(+) T cells subsets were more prone to spontaneous apoptosis than Tim-3(-) T cells subsets. Our findings reinforce the idea that the up-regulated expression of Tim-3 on T cells after burn injury plays an important role in the development and maintenance of burn-induced T cell immune suppression.
7.Experimental study on the oral care effects of a finger toothbrush

Objective: To observe the oral care effects of finger toothbrush. Methods: The finger toothbrush worn on the index finger was made of silica gel .It was used to brush the in vitro healthy teeth pulled out on the same day and the standard hard plaster tooth model and to brush the in vivo healthy teeth in adults; the bacteria in the finger toothbrush, The dentofacial pluque and the abrasion on the tooth model were tested respectively. Results: After in vivo use the bacteria remained in the finger toothbrush were fewer than those in the commonly used toothbrush(1 683.24?1 355.59 and 2 353.76?1 582.06 respectively). The dentofacial plaque was decreased in the same extent by the two kinds of toothbrushes. The zero rate of abrasion on the tooth model was 93.3%(28/30) by finger toothbrush and 23.3%(14/30) by the commonly used toothbrush. Conclusion: The finger toothbrush worn on finger can remove dentofacial plaque effectively and produce a lower abrasion on tooth.
8.Observations on therapeutic effects of diltiazem on coronary myocardiai ischemia

The therapeutic effects of diltiazem in the dosage of 135~240 mg/d on 26 cas-s with coronary myocardial ischemia were observed.It was found that diltiazem could markedly slow down tho heart rate and decrease the blood pressure of the patient when he was at rest and 2,4 and 6 minutes after he underwent exercise test.It could also increase exercise tolerance,delay an exercise-induced anginal attack,improve the ischemic depression of ST segment,and PEP/LVET.Eventually it could pievent the patient from anginal attacks.The therapeutic effects of diltiazem would be more apparent in those cases of myocardial ischemia accompanied with hypertension.
9.Long-term effects of different drugs on contralateral testis with unilateral testicular torsion in prepubertal rats

[ABSTRACT]AIM:Toobservethelong-termeffectsoftaurine,Danxuetong(Salviamiltiorrhizacombinedwith Xueshuantong ) and breviscapine on contralateral testis with unilateral testicular torsion in prepubertal rats , and to compare their protective effect on testis with ischemia reperfusion injury .METHODS: Four-week-old healthy male SD rats ( n=64) were randomly divided into 8 groups:sham operation group, saline group, single-dose and successive taurine groups, single-dose and successive Danxuetong groups , and single-dose and successive breviscapine groups .The animal model of left testicular torsion reduction (720°for 2 h) was established.After 6 weeks of surgery, the right testes and epididymides from the rats were collected immediately to detect the total antioxidant capacity (T-AOC), the activity of superoxide dis-mutase (SOD), the activity of nitric oxide synthase (NOS) and the content of malondialdehyde (MDA).The sperm de-formity, sperm viability and sperm concentration were measured , and the pathological examination of the testicular tissues was performed.RESULTS:Compared with saline group, the activity of SOD, T-AOC, sperm viability and sperm concen-tration were increased , while MDA content and sperm deformity were decreased in 3 successive groups .SOD activity and sperm viability ( except single-dose breviscapine group ) were increased , but MDA content and sperm deformity were de-creased in 3 single-dose groups .T-AOC activity and sperm concentration were increased in single-dose taurine group .The saline group showed interstitial edema and degeneration of seminiferous tubules , and the rest groups showed a significant improvement in histological morphology .CONCLUSION:The contralateral testis in prepubertal rats was damaged after u-nilateral testicular torsion , and taurine , Danxuetong and breviscapine have long-term protective effect on the tissues under ischemia-reperfusion injury .The protective effect of Danxuetong is better than breviscapine , and taurine is the best of them.The effect of single-dose group is better than the successive application .
