Objective To investigate the clinical efficacy of acupuncture plus Chinese herbal medicine in treating allergic rhinitis. Methods One hundred and thirty-five patients with allergic rhinitis were randomly allocated to combination, acupuncture and Chinese herbal medicine groups, 45 cases each. The combination group received treatment with acupuncture plus Chinese herbal medicine; the acupuncture group, treatment with acupuncture alone; the Chinese herbal medicine group, treatment with Chinese herbal medicine alone. IgE, IL-4, IL-5 and TNF-αlevels and clinical symptom and sign total score were observed in the three groups before and after treatment. The clinical therapeutic effects were compared between the three groups. Results There were statistically significant pre-/post-treatment differences in IL-4, IL-5, TNF-αand IgE levels in the three groups (P<0.05). There were statistically significant post-treatment differences in IL-4, IL-5 and IgE levels between the combination group and the Chinese herbal medicine or acupuncture group (P<0.05). There was a statistically significant pre-/post-treatment difference in clinical symptom and sign total score in the three groups (P<0.05). There was a statistically significant post-treatment difference in clinical symptom and sign total score between the combination group and the Chinese herbal medicine or acupuncture group (P<0.05). The total efficacy rate was 91.1%in the combination group, 71.1%in the Chinese herbal medicine group and 66.7%in the acupuncture group. There was a statistically significant difference in the total efficacy rate between the combination group and the Chinese herbal medicine or acupuncture group (P<0.05). Conclusions Acupuncture plus Chinese herbal medicine has a better therapeutic effect on allergic rhinitis. It can effectively relieve the clinical symptoms and signs and reduce IgE, IL-4 and IL-5 levels in the patients.

Objective To explore the clinical significance of combined detection of C‐reactive protein (CRP)and white blood cells (WBC)in children with anemopyretic tonsillitis .Methods A total of 284 patients with anemopyretic tonsillitis were divided into ex‐cessive pattern group (131 cases) ,wind‐heat invasion group (153 cases) ,other 120 healthy children were recruited into control group .The WBC ,percentage of neutrophils (NEU % ) ,percentage of lymphocytes (LYM % ) and CRP were analyzed among the three groups .Results WBC and NEU% in excessive pattern group and wind‐heat invasion groups were higher than those in the control group (P< 0 .05) ,CRP in wind‐heat invasion group was higher than that in excessive pattern group and the control group (P< 0 .05) .CRP in the excessive pattern group were higher than that in the control group (P< 0 .05) .When CRP cut‐off value in wind‐heat invasion group was 12 .65 mg/L ,the sensitivity was 90 .6% ,specificity was 86 .3% .Conclusion The combined detection of WBC and CRP have clinical value in differential diagnosis of children with anemopyretic tonsillitis .

Objective The patients with non-syndromic deafness in Shanxi Province were retrospectively an_alyzed for the common deafness gene mutations and frequency of mutations carrying rate ,to understand the molecu_lar pathogenesis of deafness in Shanxi area .Methods Genomic DNAs of 800 patients of non -syndromic deafness within Shanxi were obtained from peripheral blood .Genes GJB2 ,GJB3 ,SLC26A4 and mitochondrial 12SrRNA 1494 and 1555 loci were sequenced after polymerase chain reaction (PCR) amplification and compared with the NCBI website for the analysis of the formation of mutations .ResuIts Among 800 patients ,353 cases (44 .13% ) showed detected deafness related mutations and the genetic etiology was found for 294 patients (36 .75% ) .Among them , 153 cases (19 .13% ) carried double allele mutations in the GJB2 gene .The most frequent mutation of GJB2 gene was 235delC ,and the carrying rate was 13 .5% (216/1 600) .The double mutant allele of SLC26A4 gene was detec_ted in 123 cases (15 .38% ) ,and the most common mutation was IVS7-2A>G ,identified in 7 .44% (119/1 600) of patients .Homogenic mitochondrial 12S rRNA 1494C> T mutation in one patient and 1555A> G mutation in 15 patients were detected .GJB3 gene c .538C > T heterozygous mutation was found in two patients .Altogether , 36 .75% (294/800) of patients with deafness were caused by gene mutations .ConcIusionThe data containing GJB2 gene and SLC26A4 gene carrying rate are consistent with the published data of non-syndromic deafness in the Northwest region of China ,but the carrying rate of mitochondrial gene mutations is lower than the average level of China .Our data show that the gene mutations contribute to 36 .75% of etiology in patients with deafness .This study reflects the importance of deafness related genes screening in Shanxi area for early diagnosis and genetic con_sultation .

To investigate the role played by 8-hydroxy-deoxyguanosine (8-OHdG) in patients with Graves'ophthalmopathy(GO),and the association between 8-OHdG and the disease activity of GO.Serum levels of 8-OHdG were tested in 48 cases with GO and 30 cases with Graves' disease(GD) without ophthalmopathy.30 healthy subjects participated in this study as controls.8-OHdG was detected by enzyme-linked immunosorbent assay (ELISA).Compared with the controls,the mean level of 8-OHdG in GO patients and GD patients was increased significantly [(2.98±1.33),(2.07±1.30) vs(0.72±0.93)ng/ml,P＜0.05].Mean 8-OHdG level in GO patients was markedly higher when compared to GD patients(P＜0.05).Pearson regression analysis showed that elevated levels of 8-OHdG were significantly correlated with the Clinical Activity Score of GO (r =0.54,P＜0.01).The serum level of 8-OHdG in GO patients was raised obviously,it implies that oxidative stress plays an important role in the pathogenesis of GO,and the serum level of 8-OHdG might act as an activity index of GO.

Objective: To investigate the effect and mechanism of rapamycin inhibiting mammalian target of RAPA (mTOR) on heart valve cell calciifcation in experimental rats. Methods: The rat’s valvular interstitial cells were isolated and the cells were cultured in 4 groups:①Normal control group,②Calciifcation group,③Rapamycin group and ④Calciifcation + rapamycin group. The apoptosis rates of valvular interstitial cells were detected by flow cytometry, calcium deposition was observed by Alizarin S staining, the calcified nodules were counted and the protein expressions of bmp-2, osteocalcin, osteopontin, smad-1 and caspase-3 were examined by Western blot analysis. Results: The rat's valvular interstitial cells were suceessfully isolated; the cell apoptosis rates were similar among different groups,P>0.05. The calciifed nodule in Calciifcation group (0.471 ± 0.091) was more than Normal control group (0.104 ± 0.023), while the nodule in Calciifcation + rapamycin group (0.237 ± 0.039) was less than Calciifcation group, allP<0.05. Compared with Normal control group, the protein expression levels of bmp-2, osteopontin and smad-1 were obviously increased in Calciifcation group,P<0.05, and compared with Calciifcation group, the above indexes were obviously lower in Calciifcation + rapamycin group, P<0.05. The protein expression levels of caspase-3 were similar among 3 experimental groups,P>0.05. Conclusion: Rapamycin may down-regulate the targeting protein expressions of bmp-2, osteopontin and smad-1 via inhibiting mTOR, therefore, reducing the valvular interstitial cell calcification which might be related to mTOR pathway suppression in experimental rats.

Objective To investigate neuroprotective mechanisms of vinpocetine by observing the effects of vinpocetine injection on the expressions of peroxisome proliferators-activated receptor γ(PPARγ),nuclear factor (NF)-κB p65,cyclooxygenase-2 (COX-2) in the ischemic cortex,and infarct volume after focal cerebral ischemia-reperfusion in rats.Methods A focal cerebral ischemia-reperfusion injury model was induced by suture method.The rats were randomly divided into a normal control,a cerebral ischemiareperfusion and a vinpocetine groups.They were also divided into either a day 7 subgroup or a day 14 subgroup (n =6 in each subgroup) according to the reperfusion time.Western blot was used to detect the expression levels of PPARγand NF-κB P65 in the ischemic cortex.Triphenyl tetrazolium staining was used to detect the volume of cerebral infarction.Results Western blot showed that at day 7 and 14 after cerebral ischemia-reperfusion,expression levels of PPARκ (all P＜0.001) and NF-κB p65 (all P＜0.001) in the cerebral ischemia-reperfusion group were significantly higher than those in the sham operation group,the expression levels of PPARκ (all P ＜0.05) in the vinpocetine group were significantly higher than those in the cerebral ischemia-reperfusion group,but the expression levels of NF-κB p65 (all P ＜0.05) were significantly lower than those in the cerebral ischemia-reperfusion group.Reverse transcription polymerase chain reaction showed that COX-2 mRNA expression levels were upregulated significantly at day 7 and 14 after cerebral ischemia-reperfusion compared with the sham operation group (all P ＜ 0.001),the expression levels of COX-2 mRNA in the vinpocetine group were significantly downregulated compared with the cerebral ischemia-reperfusion group (all P＜ 0.05).The infarct volumes at day 7 (134.308± 9.954 mm3vs.185.543 ± 9.100 mm3;q=10.659,P＜0.001) and at day 14 (137.865 ± 9.094 mm3vs.183.210±4.368 mm3;q=11.166,P＜0.001) in the vinpocetine group were significantly less than those in the cerebral ischemia-reperfusion group.Conclusions Vimpocetine significantly reduces infarct vohme after focal cerebral ischemia-reperfusion,its mechanism may be associated with upreguhtion of PPARγexpression and downreguhtion of the expressions of NF-κB p65 and COX-2.

Objective:To explore the judging value of brain natriuretic peptide (BNP) on patient's condition of male patients with senile degenerated heart valvular disease (SDHVD) complicated heart failure (HF) .Methods :A total of 67 SDHVD + HF male patients were regarded as SHDVD group ,another 43 male HF patients ,who accepted treatment during the same period with corresponding age and without valvular heart disease ,were enrolled as pure HF group .Cardiac function indexes and plasma BNP level were measured and compared between two groups ,the correlation between BNP and left ventricular end -diastolic dimension (LVEDd) was analyzed ,and the cutoff point of BNP level for judging severity of SDHVD + HF was also calculated .Results:Compared with pure HF group , there were significant rise in LVEDd [ (49.79 ± 4.31) mm vs .(53.04 ± 7.10) mm] and plasma BNP level [ (214.43 ± 237.71) pg/ml vs . (682.06 ± 981.15 ) pg/ml ] , and significant reduction in left ventricular ejection fraction [LVEF ,(64.84 ± 5.83)% vs .(58.24 ± 7.99)% ] , P<0.05 or <0.01;percentage of moderate -severe heart dys‐function (NYHA class Ⅲ + Ⅳ) in SDHVD group was significantly higher than that of pure HF group (56.72% vs . 37.21% ) ,χ2 =3.988 , P=0.046. Spearman correlation analysis indicated that BNP level was positively correlated with LVEDd (r=0.588 ,P= 0.001) .The area under ROC (AUC) of BNP was 0.975 ,and the critical point of BNP level judging severity of male SDHVD complicated HF was 312 pg/ml ,and its sensitivity and specificity was 86.8% and 96.6% respectively .Conclusion:The SDHVD patient's condition is more severe compared with patients without valvular heart disease ,and BNP level is help to judging patient's condition .

Objective To value the clinical application of diffusion tensor imaging (DTI) in cerebral diseases. Methods Six volunteers and 6 patients (including 3 patients with ischemic stroke and 3 patients with glioma) were examined by DTI and T1weighted, T2weighted MR scan. All data were processed with DtiStudio software to show the white matter fiber tracts. The fractional anisotropy (FA) of the diffusion tensor were measured between the affected and the unaffected side. Results The white matter fiber tract could be observed clearly on the FA map. The pyramidal tract with different degree disruption could be showed in 3 patients with ischemic stroke. Compression, displacement, infiltration or destruction of pyramidal tract, corpus callosum or internal capsule and external capsule could be seen in 3 patients with glioma, and FA was significantly reduced on the affected side as compared to the unaffected side. Conclusion Diffusion tensor imaging is useful in observing the damage and displacement of the white matter fiber tracts in vivo, beneficial to the surgical plan for patients and prognosing recovery of function.
