1. Effects of preventive high thoracic epidural block for different periods on cerebral vasospasm in rabbits

Objective: To investigate the effects of preventive high thoracic epidural block (PHTEB) for different periods on cerebral vasospasm (CVS) following subarachnoid hemorrhage (SAH) in rabbits. Methods: T6-7 epidural space was opened and an epidural pipe was introduced to establish PHTEB model. Ropivacaine(1 g • L-1) was injected continuously through the pipe at 1 ml/h to block T1-5 sympathetic nerves. Sixty HTEB rabbits were assigned to 6 groups (n = 10 each): the normal 1 day group (N1), the normal 7 day group(N7), SAH 1 day group (S1), SAH 7 day group (S7), PHTEB plus SAH 1 day group (HS1), and PHTEB plus SAH 7 day group (HS7). CVS was developed by injecting non-anticoagulant autologous arterial blood (0.5 ml/kg) into the cisterna magna. Transcranial Doppler (TCD) ultrasonography was used to determine Vm of the basilar artery. The lesion and spasm of the basilar artery were observed under optical microscope. Results: Compared with group N1, Vm was significantly increased in group S1 (P<0.01) and increased in group HS1 (P<0.05). Compared with group N7, Vm was significantly increased in group S7 (P<0.05) and similar to that in group HS7 (P>0.05). Vm in group S1 was significantly increased compared with group S7 (P<0.05); and that in group HS1 was significantly increased compared with group HS7 (P<0.05). Pathological observation showed no obvious changes in group N1 and N7. The vessel wall was shrunk and the lumina became narrow in group S1 and S7. The pathologic changes in group HS1 and HS7 were slighter than those in group S1 and S7;and Group HS7 was slighter than group HS1. Conclusion: PHTEB can relieve CVS in SAH rabbits, and the relieving effect increases with time of PHTEB.

3.Stable replication and antigen expression of hepatitis B virus in HepG2 cells

Objective To establish the HepG2 cell lines which can stably express and replicate hepatitis 13 virus (HBV).Methods One point two X unit length of HBV genome was cloned intn SalⅠsite of the eukaryotic expression vector pREP10 to construct the recombinant plasmid pREP-HBV. Human hepatoblastoma cell HepG2 was transfected with pREP-HBV by Lipofectamine 2000 and seh,cted by bygromycin at the concentration of 250?g/mL.HBsAg and HBeAg were monitored by enzyme linked immunosorbent assay (ELISA)kits.H13V particles presemed in supernatant were ex- amincd by electronic microscopy.DNA isolated from intracellular HBV core particles was analyzed by Southbern blot using HBV-specific probe.Results The recombinant vector pREP HBV containing 1.2?unit length of HBV DNA was constructed successfully.After transfection of pREP-HBV to HepG2 cells and consistently cultured in hygromycin selective medium.5 drug-resistant cell lines, RHBV1-RHBV5.were established,and all of them could stably express HBsAgand HBeAg.South ern blot analysis revealed that HBV could replicate in all cell lines,as confirmed by the presence of replicateintermediatc DNA in intracellular HBV core particles.Clustered 42 nm Dane particles as well as 22-26 nm spherical H13sAg particles in condensed cuhure supernatant were visualized by elec tronic microsopic analysis.Conclusion HepG2 ceil lines in which HBV can replicate and express specific antigens are successfully established.Up to now,the cells have been passaged every three days for 50 times.
4.Clinical effect of early postoperation adjuvant intraperitoneal chemotherapy of locally advanced gastric cancer

0.05).The most common toxicity was neutropenia and nausea.There are no serious nausea,damage of liver and kidney functions and grade 4 neutropenia in the study group.The side-ef- fects,in the study group,such as hemorrhage,infection,medicine outleakage,catheter obstruct,catheter slough,gastric-intestinal perforation,and fever did not appear.Conclusion Although this study was conduct- ed in a small number of patients and short time,compared with the control group,the study group had a ben- eficial effect in peritoneal recurrence after curative gastrectomy for advanced gastric cancer and had less side effect and good toleration.It played a role in enhancing 1-year overall survival rate.The combination of intra- venous and intraperitoneal chemotherapy after gastrectomy may have better treatment effect on gastric cancer than that of intravenous chemotherapy alone.The randomized controlled trials are required.

6.Effects of Jianpi Qinghua Chinese herbal compound on TLR4-MyD88-dependent pathway protein expression and TNF-αin animal model of chronic atrophic gastritis(CAG) in rats

Aim To investigate Jianpi Qinghua Chinese herbal compound( JQCC) on the expressions of the rel-evant proteins of TLR4 and its downstream MyD88-de-pendent pathways, and on the inflammatory factor TNF-α in the animal model of chronic atrophic gastritis ( CAG) in rats, so as to discuss the molecular mecha-nism of JQCC in the treatment of CAG. Methods 53 Wistar rats were randomly divided into the blank con-trol group(n=8) and the CAG model group(n=45), and the animal model of CAG in rats was replicated by the “ammonia + sodium deoxycholic acid + ethanol”method. After the successful modeling was confirmed, the rest of the 40 CAG rats in the CAG model group were divided into the model group, the vitacoenzyme-tablet group, the low dose of JQCC group, the medium dose of JQCC group, the high dose of JQCC group ( each group n =8 ) . The experimental animals of all the groups were given intragastric administration of medication continuously for 30 days. Then the patho-logical histological changes were observed by HE stai-ning. The protein expressions of TLR4, MyD88, NF-КB and COX-2 were tested by Western-blot assay. And the serum TNF-α level was measured by ELISA. Results The protein expressions of TLR4 , MyD88 , NF-КB and COX-2 and the serum TNF-α level in the rats in the model group were increased evidently ( P<0. 01). Compared with the model group, the gastric mucosa lesions were improved in the low dose of JQCC group, the medium dose of JQCC group, the high dose of JQCC group, together with significant decreases of the protein expressions of TLR4 , MyD88 , NF-κB and COX-2 and the serum TNF-α level ( P <0. 05 or P <0. 01 ) . Conclusion JQCC could effectively improve the pathological and histological changes in the gastric mucosa in CAG rats, and the therapeutic mechanism might be related to the expressions of the relevant pro-teins of TLR4-MyD88-dependent pathways and the ex-pressions of anti-inflammatory cytokines.

Objective To investigate the application of two-factor theory of nursing students to improve the quality of teaching.Methods for nursing students of 2009,through the application of two-factor theory,to take effective measures to meet the health factors,and use incentives to improve nursing students to learn initiative and enthusiasm,for the nursing students of 2008,using the traditional clinical teaching methods.Results The 2008 nursing students theory test scores for (83.94 ±8.21 )points,operation test scores for (81.28 ±6.54)points,zongping scores for (82.47 ± 6.89 )points; The 2009 nursing students theory test scores for (87.17 ± 5.13 )points,operation test scores for (85.24 ± 4.61 )points,zongping scores for (86.27 ± 7.14)points,two groups of comparisons were statistically significant differences(t =-3.72,-4.14,-3.97;P <0.05).Conclusions Thetwo-factor theory can improve nursing students to learn initiative and enthusiasms to improve the quality of teaching.
9.Intravenous injection of Xuebijing attenuates acute kidney injury in rats with paraquat intoxication

BACKGROUND: The study aimed to investigate the therapeutic benefits of intravenous Xuebijing on acute kidney injury (AKI) in rats with paraquat intoxication. METHODS: Male Sprague-Dawley rats were randomly divided equally into three groups:sham group (n=8), paraquat group (n=8) and Xuebijing-treated group (n=8) using a random number table. The rats were intraperitoneally injected with 50 mg/kg of paraquat. One hour after paraquat administration, the rats were treated intravenously with Xuebijing (8 mL/kg). At 12 hours after paraquat administration, serum was collected to evaluate kidney function, then the rats were sacrificed and kidney samples were immediately harvested. AKI scores were evaluated by renal histopathology and pro-inflammatory cytokines mRNA levels in kidney were assayed using real-time RT-PCR. RESULTS: Serum urea nitrogen, creatinine and AKI scores were significantly higher in the paraquat group, compared with the sham group (P<0.05, respectively). Moreover, interleukin (IL)-1β, IL-6 and TNF-α mRNA levels were significantly higher in the paraquat group (P<0.01, respectively). However, intravenous Xuebijing significantly decreased serum urea nitrogen, creatinine, AKI scores and IL-1β, IL-6 and TNF-α mRNA levels, compared with the paraquat group (P<0.05, respectively). CONCLUSION: Intravenous Xuebijing attenuates AKI fol owing paraquat poisoning by suppressing inflammatory response.
10.Application of colonoscopic tattooing with autologous blood for localization of colorectal lesions in laparoscopic surgery

Thirteen patients with colorectal lesions underwent laparoscopic surgery from January to December 2019. Before surgery, 5.0 ml autogenous blood was injected under colonoscopy into the inferior margin and opposite sides of the lesion for localization. The operation time,success rate,complications,location efficiency and postoperative pathology were evaluated. The autologous blood tattooing was easily applied for all patients without complication. At laparoscopic surgery,the lesions of all patients were clearly visualized except one obese patients with rectal tumors, because the tumor was located below the retroperitoneal fold. No blood diffusion and leakage,and local inflammatory responses were observed. The surgical margins of all samples were tumor negative. Preoperative tattooing with autologous blood is recommended as an easy,safe and economical procedure for colonoscopic surgery in patients with colorectal lesions.
