1.?-Lactamase Genes of Multidrug Resistant Acinetobacter baumannii Isolates

OBJECTIVE To study the 16 genotypes of ?-lactamase in multidrug-resistant Acinetobacter baumannii isolated from inpatients,to know the state of multidrug resistance of A.baumannii in Urumqi.METHODS Bacterial strains were identified by system of API,and antibiotic susceptibility test was detected by K-B and microdilution according CLSI.PCR was used to determine the genotypes of ?-lactamase and compare with that in GenBank.RESULTS The antibiotic resistance rates to cefataxime and tetracycline of the isolates were the highest,(100%).Resistance rate to cefoperazone/sulbactam was the lowest(5 TEM(25%)strains and 10 OXA-23 strains(50%)from 20 strains multidrug resistant A.baumannii).CONCLUSIONS There is a high percentage of multidrug resistance production of A.baumannii in Urumqi.The ?-lactamase is focused on TEM and OXA-23;it is deserved to make further augmentation of epidemiology surveillance on ?-lactamase.

'99 Rehabilitation Exercise Program after THA includes myodynamic exercise,range of motion,activity of daily living discipline.The patients begin to do this program as soon as they finish operation.And the whole time is two weeks.The program includes clinostatism and sitting position, standing position and walking exercise.The patients who have done this program can restore affected extremity function quickly, reduce postoperatives and shorten hospitalization dates.The program is feasible in clinic.

5.Effects of FLAG regimen and IA regimen for acute myeloblastic leukaemia cells on P-glycoprotein-positive and P-glycoprotein-negative

Objective To explore in vitro effects and the mechanism for FLAG regimen compared with IA regimen in P-glycoprotein-positive and -negative acute myeloblastic leukemia(AML) cell lines. Methods The expression of P-glycoprotein in K562 and K562/A02 cells were analyzed by flow cytometry. The effects of FLAG and IA on the proliferation of K562 and K562/A02 cells were detected by MTT assay. The Ara-CTP and Ara-C levels in those cells were measured by HPLC,the gene expression of hENT1 in K562 and K562/A02 cells was detected by real-time PCR. Results The positive rates of P- glycoprotein were (1.32±0.24)% in K562 cell and (97.66±3.77)% in K562/A02 cell, respectively. The expression of P- glycoprotein had no change after treated with FLAG or IA. The cytotoxicity to K562 of IA was better than FLAG [(84.41±9.33) % v.s (73.17±13.20)%, P<0.05], and the cytotoxicity to K562/A02 of FLAG was better than IA [(70.55±11.32)% v.s (48.46±12.81)%, P<0.01]. Ara-C and Ara-CTP accumulation and hENT1 expression in AML cells treated with FLAG were higher than that treated with IA. Conclusion P-glycoprotein-positive AML cells are more sensitive to FLAG regimen than IA regimen. The biochemical modulation of Ara-CTP and Ara-C may be the major mechanism.
6.In vitro experience with tissue-engineered heart valve leaflets

Objective: The experiment was designed to evaluate the in vitro creation of tissue engineered heart valve leaflets. Methods: Aortic artery was harvested from 6 hybrid young pigs. The endothelial and myofibroblast cells were isolated and cultured to get enough cells for seeding. The myofibroblast and endothelial cells were seeded on the scaffolds sequentially. The cellular scaffolds were cultured 28 days and sent for scanning electron microscopy (SEM), histological and hydroxyproline examination. Results: SEM showed that a large amount of cells attached on the scaffolds. The cell had covered a large area of scaffold with a lot of matrix synthesized. The HE stain showed large amount of cells growing on the scaffolds and some cells were found in the central part of scaffolds. Gomori stain showed that there was elastin synthesized on the scaffolds. Hydroxyproline content is (10.93?2.89) ?g/g (scaffolds). Conclusion: With vascular cell, it is possible to seed them onto biodegradable scaffolds and create a viable tissue-engineered heart valve leaflets in vitro.
7.Establishment and analysis of serum two-dimensional gel electrophoresis profiles of myasthenia gravis patients with spleen and kidney deficiency syndrome

OBJECTIVE: The purpose of this study was to establish two-dimensional gel electrophoresis (2-DE) profiles of serum of myasthenia gravis patients, and to identify the differential proteomic expressions between normal persons and myasthenia gravis patients with spleen and kidney deficiency syndrome. METHODS: Samples of serum protein were extracted by repeated freeze-thaw method and separated by two-dimensional electrophoresis. Differential proteomic expressions between the myasthenia gravis patients and the normal control persons were identified by two-dimensional polyacrylamide gel electrophoresis, silver staining, image-master 2-DE software analysis, peptide mass fingerprinting based on matrix assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF-MS), BioWorks and NCBI software database searching. RESULTS: The two-dimensional polyacrylamide gel electrophoresis profiles of serum proteins were successfully established by 2-DE. Twenty-one of the significant differential proteins were selected and identified by MALDI-TOF-MS. Eight of them were finally identified. CONCLUSIONS: The 2-DE profiles of serum proteins were established and the differential proteomic expressions were identified by proteome technique in our study. This can be an experimental basis for further research of the pathogenesis and treatment of myasthenia gravis.

Henoch-Schonlein purpura(HSP)is the most common vasculitic disease affecting children,which is a multisystem immunoglobulin A mediated vasculitis with a self-limited course affecting the skin,joints,gastrointestinal tract and kidneys.Antiphospholipid syndrome(APS)is a systemic autoimmune disorder characterized by a combination of arterial or venous thrombosis and recurrent fetal loss,accompanied by elevated titers of antiphospholipid antibodies.Both HSP and APS are multisytem diseases and are thought to result from immunologically mediated processes.It was known that APS can associated with HSP,but the relationship between the 2 diseases is unclear.The literature on HSP associated with APS was reviewed and the possible mechanism was analyzed.

10.Combined Effect of Atrazine and Chlorotoluron on Mice Testis,a Morphological Research

Objective To study the combined effect of chlorotoluron and atrazine on the testis of mice. Methods Kunming mice were divided into groups and treated with chlorotoluron and atrazine alone or in combination by gavage at different doses for 25 consecutive days. Microscope and electron microscope were used to observe the morphological changes. Results The herbicides used alone or in combination, at all the test doses, caused the morphological changes in degrees in the testis. Under the light microscope, seminiferous epithelium arrayed loosely and disorderedly, spermatogenic cell shed, and layers lessened compared with the control group. Under the electron microscope, mitochondria in the seminiferous epithelium appeared vacuolated, karyotheca swelled, bent and the function of sustentacular cell declined. Compared with alone use groups, the pathological changes were more serious in combination use groups. Conclusion Chlorotoluron and atrazine can produce a combined toxic effect on the testis of mice.
