2.Research progress of growth factors in the pathogenesis and developments of myopia

Myopia is one of the most popular eye diseases all over the world. The development of the current understanding of its mechanism is still limited. Many studies indicated that the growth factors closely related to eye development and myopia. Some growth factors with biological activity, such as transforming growth factor ( TGF ), fibroblast growth factor ( FGF ) and epidermal growth factor ( EGF ), have an impact on scleral thickness variation, the regulation of the development of myopia and so on, which plays a non-negligible role in the pathogenesis of myopia. In this paper, the function of various growth factors in myopia will be reviewed.
3.Morphological evaluation of lumbar dorsal root ganglion on three-dimensional magnetic resonance imaging

Objective To investigate the morphological features of normal lumbar dorsal root ganglia using a three-dimensional(3D)coronal MR imaging.Methods One hundred and fifteen volunteers were included.Ages ranged from 15 to 75 years,with a mean of 40 years.Coronal 3D fast field echo(FFE) with water selective excitation(Proset)MR examination of 1150 dorsal root gangha were underwent at nerve root levels from L1 to L5.The source coronal images were further reconstructed into a series of rotational alignment coronal images with an interval angel of 12 degree using maximum intensity projection(MIP) technique.All DRGs of bilateral spinal nerve from L1 to L5 were morphologically analyzed on the original and MIP images including qualitative evaluation of the location,signal intensity,architecture and quantitative dimensional measurement.Results There were 225,225,219,210 and 160 foraminal ganglia from L1 to L5 level,respectively.The incidence of intraspinal ganglia from L3 to L5 gradually increased with a maximum at L5 level of 29.1%(X~2=188.371,P
4.Clinical and genetic analysis of Dent' s disease in 6 Chinese children with low molecular weight proteinuria.

OBJECTIVETo analyze the clinical features and gene mutations of 6 Chinese children with Dent's disease.
METHODThe clinical and laboratory data of 6 children with Dent's disease were summarized. CLCN5 gene was analyzed using PCR amplification and DNA sequencing.
RESULTAll the six patients presented with low molecular weight proteinuria and hypercalciuria, including 3/6 hematuria, 4/6 nephrocalcinosis, 3/6 hypophosphatemia, 1/6 rickets. Six mutations of the CLCN5 gene were revealed, including L594fsX595, R637X, R467X, IVS4-2A > G, S244L and V505G. The mutation L594fsX595, IVS4-2A > G and V505G was never reported before.
CONCLUSIONLow molecular weight proteinuria and hypercalciuria were the main clinical features of the six Chinese boys with Dent's disease. Dent's disease could be associated with a Bartter-like syndrome, which make the gene diagnosis more important.

6.Application of near-infrared spectroscopy technology in extraction and concentration process of Reduning injection.

OBJECTIVETo establish a rapid quantitative analysis method for the content of chlorogenic acid and solid content in the extraction liquid concentration process during the production of Reduning injection by using the near-infrared (NIR) spectroscopy, in order to reflect the concentration state in a real-time manner and really realize the quality control of concentrating process of the extraction and concentration process.
METHODThe samples during the Jinqing extraction liquid concentration process were collected. After the removal of abnormal samples, the spectra pretreatment and the wave band selection, the quantitative calibration model between NIR spectra and chlorogenic acid HPLC analytical value and solid content was established by using PLS algorithm, and unknown samples were predicted.
RESULTThe correlation coefficients between the chlorogenic acid content and the solid content were respectively 0.992 1 and 0.994 0, and the correlation coefficients of the verification model were respectively 0.994 4 and 0.998 4, with the root mean square error of calibration (RMSEC) of 0.814 6 and 2.656 1 and the root mean square error of prediction (RMSEP) of 0.704 6 and 1.876 7 respectively, and the relative standard errors of predictions (RSEP) were 6.01% and 2.93% respectively.
CONCLUSIONThe method is simple, rapid, nondestructive, accurate and reliable, thus could be adopted for the fast monitoring of the chlorogenic acid content and the solid content during the concentration process of Reduning injection extraction liquid.

7.Evaluation of correlation between gelatin crosslinking indicators and influence factors.

AIMTo evaluate the correlation between the swelling equilibrium quantity (Seq), dissolution rate and amino group content which were used to estimate the degree of gelatin crosslinking, and the factors which influence the crosslinking, and to investigate the crosslinking mechanism of delayed dissolution of soft capsules.
METHODSThe swelling equilibrium quantity, dissolution rate and amino group content of formaldehyde-crosslinked capsule shells were determined. Gelatin structure was analyzed by means of DSC.
RESULTSA good lineal relativity was found between the swelling equilibrium quantity, dissolution rate and amino group content (r = 0.9953-0.9985). The Seq of capsule shells was influenced by high temperature, lighting and some additives. Glycine and sodium pyrosulfite might retard the decrease of Seq of capsule shells.
CONCLUSIONThe swelling equilibrium quantity, dissolution rate and amino group content exhibited the same rule in gelatin crosslinking extent. High temperature, lighting and some additives might induce gelatin crosslinking. Antioxidant such as glycine and sodium pyrosulfite might retard the crosslinking.

8.An analysis on the temporal-spatial epidemiological characteristics of HFMD based on village scale

Objective To learn the temporal-spatial distribution and clustering of hand-foot-mouth disease (HFMD)in Xiaoshan in 2014,and to provide reference and basis for prevention and control.Methods The HFMD data of Xiaoshan in 2014 was derived from the China Information System for Diseases Control and Prevention.The vector map was created by Map Info 10.0 on the background of 1∶65 000 zoning map ,extracted village and community geographical position information from Baidu map.At the village level,the spatial autocorrelation analysis and spatial scanning analysis were made using software ArcGIS10.2 and SaTScan9.2.Results In Xiaoshan,the epidemic curve of HFMD in 2014 showed two peaks,during April to July and during September to October.The global spatial autocorrelation Moran's I index was 0.442 7(P<0.001),and the global Getis-Ord G value was 0.003 3,(E(G)=0.002 1,Z(G)=11.82,P<0.001). Local autocorrelation analysis showed that the cluster state was high-high.Ningwei Street had the most hot spots.Five statistically significant HFMD clusters were identified by space-time scan statistics,the most likely cluster was located in Heshang Town,from January 6 to February 4 (RR=23.00,LLR=17.45,P<0.05).Conclusion In Xiaoshan,the major epidemic peak of HFMD in 2014 was from April to July.A positive spatial correlations was found,and the disease showed a distinct regional distribution feature and temporal-spatial clustering.The clusters were observed including the villages and communities of rural-urban continuum and in vicinity of industrial development zone in countryside.
9.Effect of microbubble cavitation on microcirculation of rat skeletal muscle.

OBJECTIVETo investigate the effect of therapeutic ultrasound-induced microbubble destruction on the microcirculation of rat skeletal muscle.
METHODSThirty SD rats were randomized into 5 groups (n=6), namely normal saline, microbubble, ultrasound, high-energy ultrasound microbubble and low-energy ultrasound microbubble groups. Before and after the treatments, the diameter and blood flow velocity in the microvessels in the skeletal muscle were measured, and the structural changes of the injured microvessels observed by electron microscopy.
RESULTSMicrobubble cavitation did not produce significant effect on the mean arterial pressure and diameter of microvessels in rat skeletal muscle (P>0.05), but the blood flow velocity was obviously lowered and blood flow volume reduced in the microvessels. The reduction of the flow velocity and blood flow volume and their subsequent recovery were associated with ultrasound energy, and in the low ultrasound energy group, the flow velocity and blood flow volume in the of venules recovered obviously after about 15 min, which, however, took approximately 1 h for the arterioles. In contrast, recovery of the flow velocity and blood flow volume in the microvessels took more than 2 h in the high ultrasound energy group. Cavitation resulted in endothelium cell rupture, widening of the endothelial interspace and entry of the red blood cells into the extravascular tissues as revealed by electron microscopy, but no rupture of the lining endothelium was observed 2 h after the treatment.
CONCLUSIONSEndothelium cell rupture induced by microbubble cavitation may affect the local microcirculation, and lower ultrasound energy exposure is associated with milder endothelial injury and more rapid recovery.

10.Study of RON mediated invasion of Raji cell line and drug-target effects.

OBJECTIVETo study the proto-oncogene RON mediated aggression of Raji cells and the inhibitory effects by monoclonal antibody Zt/f2 (2f2).
METHODSThe effects of RON ligand macrophage stimulating protein (MSP) (2.0 nmol/L) and inhibitory Zt/f2 (2F2) (2.0 nmol/L) antibody on proliferation of RON positive Raji cells after treatment for 24 and72 hours were detected by MTT method, colony formation units (CFU) of Raji cells by methylcellulose semi solid culture, Raji cells apoptosis and cell cycle analysis by AnnexinV/PI double staining, expression of RON, apoptosis-related proteins, and cyclins by Western blot.
RESULTS(1)Compared with the cell viability (1.0) and counts of CFU (103.6±7.0) in control group, Raji cells after MSP treatment had better viability (1.35±0.20) and CFU counts (133.7±10.4) (P<0.05), but worse viability (0.68±0.11) and CFU counts (66.3±6.1) after Zt/f2 (2F2) treatment (P<0.05). (2)Percentage of Raji cells apoptosis after Zt/f2 (2F2) antibody treatment (12.16±2.33)% was significantly increased than the control (2.89±1.03)% (P<0.05). The percentage of Raji cells arrested in G0/G1 phase was increased after Zt/f2 (2F2) antibody treatment as compared to the control [ (54.96 ±3.70)% vs (39.10±2.30)%, (P<0.05) ]. (3) High-level of RON phosphorylation and β-catenin expression activated by MSP could be inhibited significantly by Zt/f2 (2F2), which also up-regulated the expression of caspase-3, caspase-8, caspase-9 and PARP and down-regulated anti-apoptotic MCL-1 gene and inhibitor of apoptosis protein XIAP expression, accompanied with G1 phase protein changes accordingly.
CONCLUSIONMSP could aggravate Raji cells proliferation. Inversely, Zt/f2 (2F2) could inhibit proliferation and induce apoptosis by inhibition of RON phosphorylation and up-regulation of apoptosis related proteins.
