4.Relationship between Rhinovirus Infection and Asthma in Children

Objective To explore the correlation of rhinovirus infection and the morbidity of asthma in children.Methods The RV gene in nasopharyngeal secretion of 30 children with asthma acute exacerbation(asthma group),30 children in asthma clinical remission(remission group) and 30 healthy children (control group) were detected by reverse transcription-polymerase chain reaction(RT-PCR).The total serum IgE(T-IgE) was determined by chemiluminescence.The eosinophil(EOS) count (EOS%) of peripheral blood and lung function were also measured.Results The rhinovirus infection rate was 36.67% in asthma group and 3.33% in remission group.No rhinovirus was detected in control group.The rate of rhinovirus infections in asthma group was significantly highter than that in remission group and control group(?2=7.50,13.47 Pa0.05). FEV1% was (62.73?13.54)% in asthma rhinovirus infection and (86.42?17.78)% masthma with no rhinovirus infection.There was remarkable difference between 2 groups(F=14.553 P﹤0.05). The T-IgE was (836.32?44.801) kU/L and EOS% was 10.63?4.09 in asthma group with rhinovirus infection.The total IgE was (439.10?231.28) kU/L and EOS% was 5.04?2.64 in asthma group with no rhinovirus infection.There was significant difference between 2 groups(U=2.475,F=20.806 Pa0.05).Conclusions There is a close correlation between rhinovirus infection and acute attack of asthma(or worsening asthma) in children. Asthma exacerbations would be likely to happened easier because of rhinovirus infection in asthmatic children with high T-IgE.
5.Application of He-Ne laser combined with TDP irradiation in the treatment of herpes zoster ophthalmicus

AIM: To evaluate the effect of He-Ne laser combined with TDP irradiation in the treatment of herpes zoster ophthalmicus.METHODS: Totally 22 patients (22 eyes) with herpes zoster ophthalmicus were treated by He-Ne laser irradiation combined with TDP (Group A).Another 22 patients (22 eyes) with herpes zoster ophthalmicus (Group B) were treated by medication.The duration of the disease, the incidence of complications, and the time of pain relief were compared between the two groups.RESULTS: The cure rate of Group A was 91%, that of Group B was 73%, there was no significant difference between the two groups (x2=1.375, P>0.05).The average cure time of Group A was 8±2.22d, Group B was 12±1.88d, the difference between the two groups was statistically significant (t=5.22, P<0.05).The complication rate of Group A was 18%, Group B was 59%, the difference between the two groups was statistically significant (x2=6.86, P<0.05).The average pain relief time of Group A was 5±1.23d, Group B was 10±1.34d, the difference between the two groups was statistically significant (t=11.17, P<0.05).CONCLUSION: He-Ne laser combined with TDP irradiation can shorten the treatment time of herpes zoster ophthalmicus, shorten the course of disease, reduce the incidence of complications, relieve pain, with no significant effect on the cure rate of herpes zoster ophthalmicus.

7.Effect of dexamethasone on cerebral white matter development of premature rats and its mechanism

Objective By making models of premature animal,explores the effects of dexamethasone on the brain development of premature rats and its mechanisms.Methods Eighteen SD rats were randomly divided into high-dexamethasone(H-Dex) group,low-dexamethasone (L-Dex) group and normal saline(NS) control group,with 6 rats in each group.The pregnant rats in L-Dex group were injected with dexamethasone [0.1 mg/(kg·d)] from 16 to 18 days of pregnancy,while the pregnant rats in H-Dex group were injected with dexamethasone [0.5 mg/(kg· d)] ; the pregnant rats in NS control group were injected with 0.9% NaCl of the same volume.All of the fetal rats were received after administrating caesarean operation on the day 19 of pregnancy.Rats were sacrificed at the directed time and brain tissue was prepared.Histological feature and the water content of the brains were observed.Level of apoptosis was measured by flow cytometry.The expressions of myelin basic protein (MBP) and interleukin(IL)-1β in brain tissue homogenate were detected by ELISA.Results (1) The brain water contents of rats in H-Dex group,L-Dex group and NS control group were (85.94 ± 0.54) %,(86.08 ± 1.01) %,(86.94 ± 0.82) %.Compared with NS control group,the water contents of Dex group were lower (P < 0.05).(2) Glial cells of brain cortex in L-Dex group and H-Dex group were more mature than in NS control group,and the changes in H-Dex group was more significant.(3) The expressions of MBP in brain tissue of H-Dex group,L-Dex group and NS control group were (5.73 ± 1.06) μg/mg,(5.46 ±0.77) μg/mg and (2.42 ±0.52) μg/mg.Compared with NS control group,Dex group was higher(P <0.05).While the expressions of IL-1β in brain tissue of H-Dex group,L-Dex group and NS control group were (249.05 ± 11.29) pg/g,(257.47 ± 9.33) and (292.66 ± 21.51) pg/g.Compared with NS control group,Dex group was lower(P < 0.05).There was no significant difference between H-Dex group and L-Dex group(P > 0.05).(4) The level of apoptosis in H-Dex group,L-Dex group and NS control group were (18.07 ± 1.63) %,(6.88 ± 0.47) % and (2.00 ± 0.32) %.Compared with NS control group,the level of apoptosis in Dex group was higher(P <0.05),and H-Dex group was higher than that in L-Dex group.Conclusion (1) Using dexamethasone prophylactic could promote the development of glial cells,reduce the water content,increase the expressions of MBP,and decrease the expressions of IL-1β in brain tissues.It indicates that dexamethasone may play a major role in maturation of fetal brain.(2) Using dexamethasone prophylactic could increase the amounts of the apoptosis cells,and this effect is dose-dependent.It indicates that dexamethasone may have a negative effect on the fetal brain and suggestes that using dexamethasone in premature infant should be cautious,and if it has to,using a lower dose.
8.Determination of alliin in Allium sativum Linn by HPLC

AIM: To establish a RP HPLC method to determine the content of alliin in Allium sativum Linn. METHODS: Chromatographic separation was performed on ODS column with a mobile phase composed of methanol water (3∶7 by volume). The eluate was monitored at 214nm wavelength. RESULTS: The average recovery of sample for alliin was 98.7%, The RSD of within day was 1.1%. The RSD of between day was 1.9%. CONCLUSION: The method is simple, rapid with good suitability.
9.Study and Application of Bacterial Mercury-resistance Mechanism and Evolution

There exist a number of mercury-resistant bacterial in environment, Mer operon is involved in the resistant mechanism, MerRTPA of Mer operon encodes the proteins related to the regulation, transport and reduction of mercury ion, respectively. The toxic mercury ion is transported by MerTP from medium to cytoplasmic mercuric reductase, MerA, and deoxidized to non-toxic and volatile element mercury, Hg(0). Bacterial mercury-resistant system originated from ancient times, and evolved into the Mer operon with diversity by gene integration and insertion. Mercury-resistant bacteria highly specifically absorb mercury ion, and can be used in recovering the mercury-polluted environment as well as the genetic selective marker.
10.Verification of a method with low usage amount of arsenic trioxide for determining urinary iodine by As (Ⅲ)-Ce4+ catalytic spectrophotometry using ammonium persulfate digestion

Objective To verify a new revised method with low usage amount of arsenic trioxide for determining urinary iodine by As(Ⅲ)-Ce4+ catalytic spectrophotometry using ammonium persulfate digestion.Methods The standard curve linearity,sample detection limit,precision and accuracy of determining urinary iodine of this modified method were verified according to Determination Methods of Chemicals in Biological Materials.Results The linear correlative coefficients of the 0-300 μg/L range and 300-1200 μg/L range calibration curve were-0.9998--1.0000(n =6) and-0.9998--1.0000,respectively.The detection limit for iodine was 1.3 μg/L.The relative standard deviations were 1.5% (1.1/71.3)-2.5% (6.2/244.9) when measuring 3 urine samples with iodine concentration of 71.3-244.9 μg/L,and 0.6%(2.4/388.5)-1.7%(17.3/1018.0) when measuring 3 urine samples with iodine concentration of 388.5-1018.0 μg/L,respectively(n =6).The test results of the four urinary iodine national standard materials with iodine concentration of 73.0,206.0,556.0 and 883.0 μg,/L were all within the given value range and the average value relative deviation was 1.8% (1.3/73.0),0.4% (0.8/206.0),0.2% (1.0/556.0) and-1.6%(-13.7/883.0),respectively (n =6).The average recovery was 98.8% with a range of 93.2% (186.3/200.0)-103.4%(51.7/50.0) when measuring 3 urine samples with iodine concentration of 64.6-144.9 μg/L and 3 urine samples with iodine concentration of 346.8-574.4 μg/L,respectively.Conclusions This new modified method greatly reduces the amount of waste containing arsenic,and can directly take urine samples with high iodine concentration to digest and determine without dilution.It is performed with good standard linear curve,better precision and high accuracy,and in line with the analysis of biological samples requirements.
