2. Protective effect of intracerebral transplantation of human mesenchymal stem cells on hypoxic-ischemic brain damage in rats

Objective: To explore the migration,differentiation and the therapeutic effect of intracerebral transplantation of human mesenchymal stem cells (hMSCs) after hypoxic-ischemic brain damage in rats, and to test whether three anti-hMSCs monoclonal antibodies prepared by our group can detect hMSCs in rat brains. Methods: hMSCs were isolated and purified by density gradient centrifugation and adherence to culture flask. Cells of passage 3-5 were prelabeled with bromodeoxyuridine (BrdU) for 72 h before transplantation. Animal models of hypoxic-ischemic brain damage (HIBD) were built with 1 month old Wistar rats. Three days after hypoxia-ischemia, HIBD rats in hMSCs-treated group (n=18) received intracerebral transplantation of 5×105 hMSCs. In control group (n=18) HIBD rats received phosphate buffered saline of the same volume. In sham-operated group (n=6) and HIBD group (n=6), rats did not receive any transplantation. Rats in hMSCs-treated group and control group were allowed to survive for 0 day,3 days, 1 weeks,2 weeks (all n=3) and 4 weeks (n=6, after transplantation). Rats in sham-operated group and HIBD group were allowed to survive for 31 days (4 weeks after transplantation). Rats of all the 4 groups received behavior test (alternative electro-stimulus Y-maze) 4 weeks after transplantation before sacrificed; their brains were sectioned for H-E staining. The brains from all rats were also prepared for immunohistochemistry analysis to detect the distribution of hMSCs. Results: Immunohistochemistry results showed that hMSCs migrated mainly along the ventricular system 1 week later,and a few hMSCs migrated along the corpus callosum to the opposite side. Four weeks after transplantaion, hMSCs migrated to the parenchyma and distributed throughout the cerebra. Behavior test showed that the total errors (TE) in hMSCs-treated group (TE= 5.00±2.82) were significantly less than those in the control group (TE = 12.67±3.72, P < 0.05). H-E staining showed that the brain damage in the hMSCs-treated group was slighter than that of the control group and HIBD group. Conclusion: After transplanted in the brain of rats,hMSCs can survive and migrate in the central nervous system (CNS). hMSCs can promote tissue repair and functional recovery of rat brain, suggesting that hMSCs might be a possible treatment for hypoxic-ischemic brain damage.
3.Effect of TPh on cell HepG-2 cell cycle, apoptosis and related gene expression of hepatocellular carcinoma

Objective To investigate the effect of different concentrations of TPh on hepatoma cells , cell viability and and its possible mechanisms of anti-tumor.Methods The inhibitory rate of hepatoma cells and cell viability on different concentrations of TPh and time were measured by MTT assay;The morphological changes of apoptosis were observed by Hoechst 33258 straining; Cell cycle distribution was evaluated by flow cytometry (FCM).The protein expression of Bcl-2 and Bax were detected by Western blot analysis.Results MTT assay showed that TPh inhibited the proliferation of HepG-2 cells in a dose-dependent manner, and the inhibitory rate increased with the increase of concentration.The inhibitory rate was 50.9% (P<0.01).The results of Hoechst 33258 staining showed that the fluorescence intensity of hepatocellular carcinoma cells was light blue in fluorescence microscopy and bright blue fluorescence in apoptotic hepatocarcinoma cells, and the apoptosis of hepatocarcinoma cells increased with the increase of drug concentration.The percentage of cells in G0/G1 phase increased with the increase of cell cycle, and the ratio of cells in S phase was decreased in G2/M phase compared with blank control group (P<0.05);Western blot results showed that compared with the blank control group, TPh inhibited the proliferation of Bcl-2 cells in a concentration-dependent manner ( P <0.05 ) , and the number of apoptotic cells increased with the increase of drug concentration (P<0.05), and the ratio of Bcl-2/Bax in the TPh group decreased significantly (P<0.05), and the expression of Bax gene increased. Conclusion TPh inhibits cell proliferation, promotes apoptosis and induces HepG-2 to block G0/G1 phase.Its mechanism may increase the expression of Bax and decrease Bcl-2 protein expression.
4.Effect of Infrared Radiation and Magnetic Fields on Synovitis of Rabbit with Knee Osteoarthritis

ObjectiveTo investigate the effects of infrared radiation and magnetic fields on the synovitis of rabbit with experimental knee osteoarthritis (OA).MethodsTwenty-four New Zealand grown rabbits were duplicated for OA model of the knee by extended fixation technique, and after 6 weeks, all rabbits were unchained from fixation. The rabbits were randomly divided into four groups: control group (no treatment), infrared group (treated with infrared radiation ), magnet-therapy group (treated with magnetic fields ) and the combined therapy group (treated with infrared radiation and magnetic fields). At the ends of the first, second and third week after treatment, 2 rabbits in each group were taken to measure the general change and histopathology of their synovium.ResultsCompared with other groups, the severity of synovitis (include the synovium line cells proliferation, the invading of granulation and blood vessels in the underlayer of synovium, the infiltration of inflammatory cells) declined significantly in combined therapy group. Scores of Ayral's index of combined group were significantly lower than other groups ( P<0.05), and there was no significant difference between infrared group and magnet-therapy group ( P>0.05).ConclusionThe combination of infrared radiation and magnetic fields can efficiently suppress the synovitis of OA and alleviate the tissue's destructions.
5.Study of Bupropion Hydrochloride Tablet and Fluoxetine in Treatment of Depression Multicenter Clinical Trial

Safety analysis suggested that no significant differences were found in symptoms and frequency of side effects between the two groups.Conclusion Bupropion hydrochloride tablet is an effective and safe antidepressant. It has similar effect and safety compared with fluoxetine in the treatment of depression.
6.Protective effect and mechanism of MST1 inhibition on kidney tissue in diabetic rats induced by streptozotocin

Objective To investigate the protective effect and mechanism of MST1 inhibition on kidney tissue in diabetic rats,and to find a new therapeutic target for diabetic nephropathy.Methods Total of 54 male SD rats enrolled in this study were divided into 3 groups including normal control (group A,n=18),MST1 inhibition group (Group B,n=18) and diabetes group (group C,n=18).Diabetes was induced by a single streptozotocin (STZ,50 mg/kg) injection in group B and group C.rats in group B received lentiviral vector contain Mst1 interference RNA (shRNA) and the rats in group C received empty vector.The end of 4th,8th and 12th week after modeling were considered as time points in this study.At each time point,the level of 24 hours urine protein (24-HUP),blood glucose and serum creatinine were examined.Pathological changes were observed with HE stain; Injury of podocyte and glomerular basement membrane (GBM) were examined with transmission electron microscope (TEM).The intensity and location of MST1 in kidney tissue were detected by immunohistochemistry.The level of MST1,Phosphorylated-MST1,nephrin,Caspase-3 and FasL were detected by western bloting.Results (1) At the starting point,there were no significant differences among groups in terms of weight,activity,eating and drinking.Since the end of 72nd hour after modeling,the levels of glucose in both group B and group C,compared to those in group A,significantly increased (P < 0.05).There was no significant difference between group B and group C for glucose level at each time point (P > 0.05); the level of 24-HUP increased significantly since the end of 4th week after modeling,and the level in group C was higher than its counterpart in group B at the same point (P < 0.05); (2) There was no significant pathological lesion observed in group A.Without obvious K-W nodular changes,mesangial proliferation was observed in group B and group C.It was shown by TEM that podocyte fusion and thickening of the GBM could be found in group B and group C.The pathological change in group B was better than that in group C; (3) Compared to group A,it was shown by western blot that the levels of MST1,Phosphorylated-MST1,Caspase-3 and FasL in group B and group C were significantly higher (P < 0.05),and the levels of nephrin in group B and group C were significantly lower (P < 0.05) since the end of 4th week after modeling.Meanwhile,the levels of MST1,Phosphorylated-MST1,Caspase-3 and FasL in group B were significantly lower than that in group C at each time point (P < 0.05),the level of nephrin in group B was significantly higher than the one in group C; (4) It was shown by immunohistochemistry that there was low MST1 expression in normal condition,especially in cytoplasm of tubular epithelial cells.The level of MST1 in group B and group C significantly increased after modeling,and the change could be the same as Western blot shown.Conclusions MST1 pathway could be involved in kidney injury induced by diabetes.MST1 inhibition could alleviate the kidney injury in STZ-induced diabetes animal model.
7.Isolation and Identification of Antimicrobial Activity of Symbiotic and Epiphyte Microorganisms on Marine Organisms

The 125 strains of the symbiotic and epiphyte microorganisms were isolated from marine organisms (Sea cucumber, Sea urchin, Sea anemone, Sea actinia, Ulra, Sargassum, Undaria). Among them,21 strains of bacteria,8 strains of actinomycetes and 2 strains of fungi have shown to have antagonistic activity on bacterial or fungal growth. In the 21 strains of bacteria, 7 strains belong to Bacillus sp., 11 strains belong to Vibro sp., and 3 strains belong to Pseudomonas sp.. In the 8 strains of actinomycetes, 5 strains belong to Streptomyces sp., 3 strains belong to Micromonospora sp.. 2 strains of fungi belong to Penicillum sp..
8.Epidemiological study of human brucekkosis in Qian'an county of Hebei province from 2007 to 2010

ObjectiveTo analyze the prevalence and epidemic trend of human brucellosis between 2007 -2010 in Qian'an county,and to provide a basis for future prevention and control measures. Methods Epidemiological data of brucellosis between 2007 - 2010 were obtained from the national disease surveillance report on management information system, population data were from the national disease surveillance information management system for basic information reporting system, and descriptive epidemiological methods was used for statistical analysis. Results A total of 86 cases were infected with brucellosis in Qian'an county from 2007 to 2010, with 68 cases of male and 18 cases of female, and male to female ratio was 3.78 : 1. Patients were mainly in the age of 20 - 55, with 40 - < 45 year-old group the highest. All patients were local residents. Most professional were livestock acquisition, processing and aquaculture personnel. The majority of patients lived in Jianchang town,with 51 cases, accounting for 59.30%. Thirteen patients were reported in 2007, incidence was 0.0201‰(13/ 647 983). Thirty-nine patients were reported in 2010, and incidence was 0.0563‰(39/657 380). There was a fluctuations increase from 2007 to 2010. In 2007 brucellosis occurred in only two townships(towns), which spread to eight townships (towns) and urban areas in 2010. ConclusionsThe epidemic of human brucellosis in Qian'an is in a spreading trend. We recommend the government to strengthen the quarantine of livestock, and the infected livestock should be timely treated. Strengthen the prevention and control in Jianchangying. At the same time increase the brucellosis propaganda, and enhance self-protection awareness of the occupational groups.
9.Effect of Comprehensive Therapy on Forearm Extensor Myotenositis: 72 Cases Report

ObjectiveTo observe the effect of comprehensive therapy on forearm extensor myotenositis.Methods72 cases were divided into two groups: a control group of 36 cases who were given routine treatment,and an experiment group of 36 cases who were given thermotherapy,computerized medium-frequency electrotherapy,physiotherapy,and ADL instruction,etc.After two courses,a simple grading score(for forearms) was used to assess the effect.ResultsOf the control group,22 cases were cured,10 remarkably effective,4 effective;of the experiment,30 cured,4 remarkably effective,2 effective(u=2.04, P<0.05).The difference of average score for forearms before and after the treatment were(6.58±3.17) points for the control and(8.19±3.55) for the experiment(t=2.03,P<0.05).The average days of cure were(5.60±2.54) d for the experiment group,shorter than those for the control(7.00±2.27) d(t=2.05,P<0.05).ConclusionComprehensive therapy is effective on forearm extensor myotenositis.

To review the development and application of powdering technique on oily drug of the traditional Chinese medicine. There have been numerous methods of powdering technique on oily drug, such as preparing complexation, microcapsule, adsorption by adsorbent, solid lipid nanoparticles, etc. And beta-Cyclodextrin complexation is the most usually operated. Powdering techniques have broad prospects in the pharmaceutical field, but more efforts should be made to improve oily drug of the traditional Chinese medicine.
