Chinese Journal of Experimental Ophthalmology 2012;30(5):467-470
Traditionally, arsenic is thought to be a strong toxicant, especially arsenic trioxide (As2O3 ).As2O3 leads to the damage of ocular surface, such as cornea, conjunctiva etc.,and the evidence of its damage to retinal function also has been found. However, arsenic is a double-edged sword, because it possesses well- remedial effect on some eye diseases, including proliferative diabetic retinopathy, lens posterior capsular opaciffcation, keratopathy,meIanoma and bleb scarring. At present, there is still few research literature was seen about the active mechanism of arsenic agents on eye damage or therapeutic effect. Some relative studies are still in an experimental stage. This offers us the new research direction in clinical treatment of eye diseases. It is believed that arsenic will play an incalculable role on experiment and clinical treatment of eye diseases as further development and research of technology. The relationship between arsenic and eye disease is summed by consulting internal and external literature.

With the wide use of nanomaterials in biomedicine, materials, chemicals and other fields, their environmental exposure and cellular toxicity have come to the attention of researchers as a new research area in the field of nanotoxicology and nanomedicine. Researches on the toxic effects of nanomaterials have not only provided of a theoretical basis for safety evaluation of nanomaterials ,but also promoted the have applications of nanotechnology. Numerous studies have revealed cell death is closely associated with the toxicity of nanomaterials. In this paper,we elaborated on the roles of three key cell death modes in nanotoxicity, including autophagy, apoptosis, and necrosis. Furthermore, the pos?sible mechanisms involved in these three modes were explored. All this will provide reference for safety evaluation of nanomaterials.

Objective To explore the clinical features and outcomes of defo rm ities correction of old bucket handle and tilt fractures of pelvis. Methods From October 1998 to October 2002, six patients treated with open reduction, osteoto my and internal fixation for old bucket handle and tilt fractures of pelvis were analyzed. There were 3 males and 3 females aging from 18 to 51 years (mean, 27. 5 years). According to Tile’s classification, all fractures were diagnosed as t ype B2 with longitudinal stability and rotational instability, which were caused by traumatic violence of lateral compression, and associated with anterior ring injuries of ipsilateral inferior and superior rami of pubis combined symphysis separation as well as contralateral posterior ring injury of innominate fracture , dislocation of sacraoiliac joint, or sacral fracture. The incidence of associa ted injury was 100% in the patients as injured. The mean interval from the prima ry injury to the operation was an average of 6.5 months ranging from 2 to 15 mon ths. The limb shortening of buckle handle side of pelvis ranged from 2 to 3.5 cm (mean, 2.5 cm), and the rotational deformity of pelvis was more than 30? in al l patients. The tilting proximal fragment of the superior ramus and disrupted sy mphysis projected inferiorly and posteriorly into the perineum. The surgical pro cedures consisted of: 1) osteotomy through the superior pubic ramus malunion in 6 cases to correct anterior ring deformity, and through innominate osteotomy in 3 cases, direct osteotomy of sacral malunion in 1 case , and sacraoiliac fusion in 2 cases to correct the posterior ring deformity; 2) reduction by manipulati on or assisted with instrumentation; 3) fixation with reconstruction plate in 3 , reconstruction plate combined with anterior pelvic frame in 2, and reconstruct ion plate combined with lag screws in 1. Results All the patients were followed up on an average of 15.6 months (range, 3 to 45 months). The limb discrepancy ha d been completely corrected following satisfactory fracture reduction and union. According to Mears’s evaluation, the anatomical reduction of the pelvis was fo und in 5 cases and satisfactory in 1 patient. According to Majeed’s radiography evaluation, 5 patients were classified as excellent, and 1 good. One patient wi th delayed sacral nerve injury recovered without any treatment after operation. Conclusion The combination of osteotomy through anterior and posterior pelvic ri ng to correct the entire deformities in one stage is an useful technique for imp rovement of the affected limb shortening and prevention of the related complicat ions.

BACKGROUND: Reactive oxygen species (ROS), including superoxide anion (O2) and hydrogen peroxide (H2O2), have been recognized as specific second messengers in signaling cascades involved in the growth and differentiation of cells. The generation of excessive ROS initiates cardiomyocyte apoptosis. This paper is aimed to corroborate the hypothesis that excessive amounts of nicotinamide adenine dinucleotide phosphate (NADPH) oxidase (NOX) 4-dependent ROS can suppress cardiomyocyte differentiation from embryonic stem (ES) cells by induction of apoptosis.OBJECTIVE: To investigate the role of NOX4 ROS on cardiomyocyte differentiation.DESIGN: Randomized and controlled trial observation.SETTING: Beijing Institute of Geriatrics, Beijing Hospital, Key Laboratory of Geriatrics, Ministry of Health.MATERIALS: ES cells were preserved by the laboratory of Beijing Institute of Geriatrics, Beijing Hospital, Key Laboratory of Geriatrics, Ministry of Health, while other reagents were purchased from Sigma Company without specific notification.METHODS: The experiment was carried out in the Beijing Institute of Geriatrics, Beijing Hospital, Key Laboratory of Geriatrics, Ministry of Health from October 2003 to August 2005. ①Mouse ES cells were differentiated into embryoid bodies (Ebs). At day 4, Ebs were managed for one hour with different concentrations (1, 10, 100, 1 000 nmol/L) of H2O2, and generation of cardiomyocytes was observed at day 8 to analyze the effect of ROS on cardiomyocyte differentiation.② CGB8 cells transfected with pcDNA3.1 and pcDNA3.1-NOX4 respectively were adopted, while those untransfected CGB8 cells were taken as controls. A quantitative NBT (nitro blue tetrazolium) test was used to measure NOX4 ROS generation. The levels of NOX4 mRNA and MLC2v protein were assayed by RT-PCR and western blotting, respectively.③ROS scavenger N-acetylcysteine (5 mmol/L) and catalase (200 U/mL) managed for 2 hours before transcription were taken as controls to observe the apoptosis of CGB8 cell after NOX4 overexpressed and the expressions of p21, p53 and Bcl-2 of NOX4-transfected CGB8 cells simultaneously. And the apoptosis of ES cells was detected by Hoechst staining, TUNEL assay and DNA fragmentation.MAIN OUTCOME MEASURES: ①the role of ROS on cardiomyocyte differentiation.②the expression of NOX in ES cells.③ the production of ROS, differentiation of cardiomyocyte and apoptosis of ES cells after NOX4 overexpressed.④the feasibility that p53, p21 and Bcl-2 are involved in the apoptosis induced by the overexpression of NOX4.RESULTS: ①Different concentrations of ROS play different roles on cardiomyocyte differentiation. The exposure of Ebs to 1-100 nmol/L H2O2 for 2 hours at day 4 of culture leaded to an enhanced beating activity (P < 0.01 or 0.001), whereas 1 μmol/L H2O2 depressed cardiomyocyte differentiation as compared with control conditions (P < 0.001). ②NOX4 was highly expressed in ES cells, while NOX1 and NOX2 were absent and only a weak band of NOX3 was detected. The results from RT-PCR revealed that NOX4 overexpressed in CGR8 transfected with pcDNA3.1-NOX4 as compared to the control.③NBT result proved that highly-expressed NOX4 produced the excessive amounts of ROS (P < 0.05). The beating activity was remarkably reduced in NOX4-overexpressing Ebs as compared to wild-type or mock-transfected Ebs (P <0.01). Moreover, a severe reduction of MLC2v protein in NOX4-overexpressing Ebs was also observed by western blot analysis.④p21 and p53 may be responsible for the NOX4-induced apoptosis. P53-/- cells R72D27 transfected by NOX4 did not induce apoptosis. NOX4 overexpression induced ES cell apoptosis could be prevented by overexpression of Bcl-2.CONCLUSION: The findings of this paper highlight the role of NADPH oxidase NOX4 in cardiomyocyte differentiation from ES cells. P53, p21 and Bcl-2 could be involved in the apoptotic pathway.
