1.Preparation and anti-cancer activity in vitro of curcumin loaded mesoporous silica nanoparticle.

This paper is to prepare curcumin (Cur) loaded mesoporous silica nanoparticle (Cur-MSN), evaluate its release behavior and anti-cancer activity in vitro. Mesoporous silica nanoparticle (MSN) was prepared by polymerization method and Cur-MSN was obtained using solvent evaporation method and impregnation centrifugation method. The preparation method was optimized using entrapment efficiency (EE) and loading efficiency (LE) as indexes. Cur-MSN was characterized with scanning electron microscope and its particle size and zeta potential were determined. Finally, in vitro release behavior in 0.2% SDS solution and its cell-killing effect on HeLa cells were also evaluated. The Cur-MSN prepared with process optimization method was round and uniform and exhibited typical mesoporous characterization. The mean particle size and Zeta potential of Cur-MSN were 75.8 nm and -30.1 mV, respectively. EE and LE of three batches of Cur-MSN were (72.55 ± 2.01)% and (16.21 ± 1.12)%, respectively. In vitro release behavior of Cur-MSN showed a sustained release profile with 83.5% cumulative release within 96 h. The killing effect of Cur-MSN on HeLa cells was dose-dependent with IC50 of 19.40 mg x L(-1), which was similar to that of Cur.

2.Application of superparamagnetic nanoparticles for cancer treatment

BACKGROUND: In recent years, nanoparticles has been rapidly developing in tumor hyperthermia, genophore research, and targeted drug therapy, particularly nanoparticle containing drug delivery systems will become another breach in tumor therapy.OBJECTIVE: To summarize the application and mechanism of superparamagnetic nanoparticles for cancer treatment in the medical field.METHODS: A computer-based online search was conducted in Medline for English language publications containing the key words of "superparamagnetic, nanoparticles, targeting" from January 2000 to October 2009. Relevant articles were also searched from CNKI with the same key words in Chinese from January 2005 to October 2009.RESULTS AND CONCLUSION: A total of 123 articles about targeting role of magnetic nanoparticles were included, and there were 24 in Chinese and 108 in English. Articles published earlier, duplicated, and similarly were excluded, and 30 references were finally included. Superparamagnetic nanoparticles characterized by targeting role under external magnetic field, and crystal of ferroso-ferric oxide did not has toxicity to cells. As a gene carrier and drug carrier, superparamagnetic nanoparticles were widely used in medical research and they also provided novel evidences for cancer treatment. By an external magnetic field, how to avoid a comprehensive system of phagocytic endothelial phagocytosis and prevent the course of treatment such as drug-induced thrombus is still inadequate.
3.The detection of viral and atypical pathogens in children with acute respiratory infection

Objective To investigete the viral and atypical pathogens in children with acute respiratory infection (ARI) .Methods A total of 17 048 serum samples were collected from children ,who were intended to diagnose as ARI .Indirect immunofluores-cence assays were performed to detect IgM antibodies against 9 common pathogens ,including mycoplasma pneumonia(MP) ,influen-za virus B(FluB) ,respiratory syncytial virus(RSV) ,parainfluenza virus(PIV) ,adenovirus(ADV) ,influenza A virus(FluA) ,legio-nella pneumophila(LPN) ,coxiella burnetii(COX) ,chamydophila pneumonia(CPn) .Results The total detection rate of the 9 patho-gens was 60 .24% (10 269/17 048) .The detection rate of MP was 47 .78% (8 146/17 048) which was the highest ,followed by FluB which was 30 .87% (5 262/17 048) .The detection rate of MP kept at a high level through the whole year ;FluB appeared a little popular in the summer ,winter and spring ;RSV had higher detection rate in winter and spring ;ADV had higher detection rate in spring ;FluA appeared a little popular in spring and summer ;PIV and LPN didn′t show any seasonal change .There were significant differences in the detection rates of ADV ,CPn ,FluA ,FluB ,LPN ,MP ,PIV and RSV among different age groups (P< 0 .05) .In mixed infection with 2 kinds of pathogens ,MP+FluB was the most common pattern which accounted for 39 .58% of mixed infec-tion .In mixed infection with 3 kinds of pathogens ,MP+FluB+RSV was the most common pattern which accounted for 13 .68% .In mixed infection with 4 kinds of pathogens ,MP+ FluB+ RSV+ PIV was the most common pattern which accounted for 3 .84% . Other mixed infection pattern was rare .Conclusion The most common pathogens causing ARI in children of Wuhan were MP ,FluB and RSV .The detection rate of each pathogen displayed specific seasonal variations and variations among different age groups . Mixed infections were more common than single infections ,and the most common pattern was MP+ FluB .
4.Duodenal fistula caused by surgical procedures of the distal common bile duct

Objective To investigate the etiology of and therapy for duodenal fistula caused by distal common bile duct operation. Methods An retrospective analysis of 19 cases of duodenal fistula after distal common bile duct operation was made.All cases received surgical management after the fistula was identified.Surgical procedures included abdominal abscess drainage in 2 cases, gastrostomy and jejunostomy combined with abscess drainage in 11 cases, gastrostomy and jejunostomy combined with duodenal repair?abscess drainage in 3 cases,gastrojejunostomy with pyloric exclusion combined jejunostomy in 1 case, repair of fistula with padicled intestinal seromuscular patch combined with gastrostomy and jejunostomy in 1 case, and repair of fistula with padicled intestinal seromuscular patch combined with gastrostomy and jejunostomy plus sphincteroplasty in one case. Results Among 19 cases,15 were cured. Two died of gastrointestinal bleeding, one of severe retroperitoneal infection,one of severe pancreatitis. ConclusionsEarly diagnosis is of great importance for the treatment of duodenal injury. Drainage is the mainstay for the treatment of iatrogenic duodenal fistula, while enough enteral nutrition should be given to accelerate spontaneous closure of the fistula.
5.Analysis of Psychological and Cognitive State of Patients with Tension Headache

ObjectiveTo explore the possible pathogeny of tension headache(TH) as well as some emotional and cognitive function impediment.Methods53 cases with tension headache were given coronal CT or MRI examination and cervix X-ray examination.Self-Rating Anxiety Scale(SAS),Self-Rating Depression Scale(SDS) and Mini-Mental State Examination(MMSE) were used to assess their emotion and cognition.ResultsThere were 14 cases of magnitude mental events induced anxiety and/or depression,23 cases of work tensions induced anxiety,14 cases of work tensions induced depression,and 12 cases of lightly cognitive impediment.ConclusionTension headache patients usually related with continued convulsion of cervical muscle induced by hard work and life.Most of them represent more or less emotional impediment,part of them represent lightly cognitive impediment.
6.Study on effect of astragali radix polysaccharides in improving learning and memory functions in aged rats and its mechanism.

To observe the effect of Astragali Radix polysaccharides (APS) on the learning and memory functions of aged rats, in order to explore its mechanism for improving the learning and memory functions. Natural aging female SD rats were selected in the animal model and randomly divided into the control group, the APS low-dose group (50 mg x kg(-1)), the APS high-dose group (150 mg x kg(-1)) and the piracetam-treated group (560 mg x kg(-1)). They were orally administered with the corresponding drugs for consecutively 60 days. Besides, a young control group was set. The learning and memory functions of the rats were tested by the open-field test and the Morris water maze task. The Western-blot method was used to observe the levels of relevant neural plasticity protein N-methyl-D-aspartate receptor (NMDA receptor) in hippocampus, calcium/calmodulin dependent protein kinase II (CaMK II), protein kinase (PKA), the phosphorylation level of CAMP response element binding protein (CREB) and the protein expression of brain derived neurotrophic factor(BDNF). In this study, the authors found that the learning and memory functions and the hippocampus neural plasticity protein expression of the aged rat group were much lower than that of the young control group (P < 0.01). Compared with the aged rat group, the APS group showed the significant improvement in the impaired learning and memory functions of aged rats and the up-regulation in the hippocampus neural plasticity protein expression. The results showed that APS may improve the learning and memory functions of aged rats by increasing the expressions of relevant neural plasticity proteins.

7.Effects of low molecular weight heparin on newborn mouse liver cells congenitally infected with human cytomegalovirus

Objective To explore the effects of low molecular weight heparin (LMWH) on the virological and pathological changes of newborn mouse liver congenitally infected with human cytomegalovirus (HCMV).Methods Sixty healthy pure line clean level BALB/c mice which were about 10 weeks old (half were female) were divided into five groups (six pairs in each group).The mice in LMWH intervention group and positive control group were intraperitoneally inoculated with 6.0 lg tissue culture infective dose50 (TCID50) of HCMV AD169; those in blank control group were intraperitoneally injected with 0.5 mL dulbecco's modified Eagles medium (DMEM) ; then all the mice were paired to mate.The pregnant mice in LMWH intervention group Ⅰ were subcutaneously injected with LMWH 1000 U/kg daily for 10 days; those in group Ⅱ were subcutaneously injected with LMWH 1000 U/kg daily for 5 days and their newborn mice were subcutaneously injected with LMWH 1000 U/kg daily for 5 days; those in group Ⅲ were subcutaneously injected with LMWH 1000 U/kg daily for 10 days in their newborn mice.All these newborn mice were sacrificed at day 10 of birth.The liver was removed for virus isolation,dry-wet weight determination,pathology examination and quantitative fluorescence-polymerase chain reaction (PCR) detection.The comparison among groups was done by analysis of variance.Results HCMV was isolated from the supernatant of liver tissue homogenate in 10-day positive control newborn mice,while HCMV was isolated in 24-day newborn mice of the other three groups of LMWH intervention.Pathology confirmed that positive control liver tissue had inflammatory changed,liver cell inflammatory swelling degeneration,vacuoles degeneration,specific HCMV inclusion body in nuclear,and portion of liver cell necrosis,while liver pathological results of LMWH intervention group showed mild liver cell inflammatory changes and slight cell inflammatory swelling degeneration,which were similar to the blank control group.The moisture of liver tissue contents in LMWH intervention group decreased more obviously than positive control group.The HCMV DNA loads in 50 mg liver tissues of LMWH intervention groups Ⅰ,Ⅱand Ⅲ were (3.26±0.43),(3.26±0.41) and (3.32±0.51) lg copy,respectively,which were significantly lower than that of positive control group [(7.38 ± 0.53) lg copy; F =314.620,P0.01],while there were no significant differences among LMWH intervention groups (P>0.05).Conclusion LMWH intrauterine and postnatal interventions can significantly reduce HCMV DNA replication in hepatocytes,and relieve inflammatory changes in liver tissue.
8.A clinical study of 32 cases with HELLP syndrome

Objective To analyze the clinical presentation and maternal-neonatal outcome of HELLP syndrome.Methods 32 cases with HELLP syndrome from January 2003 to January 2009 were analyzed retrospectively.Results The incidence of HELLP syndrome in preeclampsia was 3.7%.Complete HELLP were 17 cases,partial HELLP were 15 cases.Two pregnant women died(one case of intracranial hemorrhage,one case of acute left cardiac failure),the mortality was 6.3%.Five newborns died,the mortality was 15.6%.The main materal complications were DIC(8 cases),placental abruption(2 cases),intracranial hemorrhage(2 cases).The main perinatal complications were preterm labor(20 cases),newborn asphysia(17 cases),SGA(15 cases).Conclusion HELLP syndrome is one of the most serious obstetric complications,early diagnosis,early therapy,controlling hypertension and termination of pregnancy in time are most important for treatment.
9.Detecting rs12979860 polymorphisms of IL28B gene by AS-PCR in the patients with hepatitis C

Objective To evaluate the value of detection of interleukin 28B (IL28B) rs12979860 by allele-specific PCR (AS-PCR) for the prediction of antiviral treatment hepatitis C patients.Methods One hundred seventy-four blood samples were random collected from hospitalized patients with hepatitis C,who came from department of infectious diseases,Renmin Hospital of Wuhan University from May 2011 to May 2012.Two pairs of specific primers were designed for rs12979860 gene polymorphisms,and one mutated base was introduced to the second or third site of the end of 3' with the reverse primer.rs12979860 gene polymorphism of 30 cases with hepatitis C was detected by AS-PCR,and gene sequencing was further used to verify the consistency of the two methods in parallel.Then,the frequency distribution of different rs12979860 genotypes with 174 cases were analyzed by the AS-PCR method in the population.Results The genotype CC,CT or TT of rs12979860 with 30 cases could be well identified by both AS-PCR and gene sequencing,and the coincidence rate was 100% (x2 =60.0,P < 0.01).Compared to gene sequencing,both of the sensitivity and specificity of AS-PCR were 100%.Compared to the control (CC genotype),TT genotype detection sensitivity by AS-PCR was 10-5,while sequencing sensitivity was 2 × 10-1.rs12979860 polymorphism in the TT,CC and CT genotype distribution in the Chinese population frequencies were 3.45% (6/174),13.2% (23/174) and 83.3% (145/174),respectively.Conclusion AS-PCR can quickly,accurate,reliable,economic and efficiently detect IL28B rs12979860 gene polymorphism of hepatitis C in patients,which could predict the effect of antiviral therapy on patients with hepatitis C.
10.Antibiotics resistance profile and ERIC-PCR fingerprinting of Pseudomonas aeruginosa isolates resisting to imipenem

Objective To explore antibiotics resistance profiles and DNA fingerprints of Pseudomonas aeruginosa isolates resistant to imipenem (IRPA). Methods DNA fingerprints of 56 strains isolated from ICU (intensive care unit) were constructed by ERIC-PCR (enterobacter repetitive intergenic consensus-PCR). MICs (minimal inhibitory concentrations) were determined by agar dilution method.Results 33 genotypes were got from 56 strains by ERIC-PCR. Of 8 frequently used antibiotics, 5 of them showed resistance rate higher than 50%. Conclusion It is high time to pay attention to multi-drug resistance of IRPA. The exist of prevalence of IRPA clone in ICU advise us to control of IRPA in hospital effectively.
