1.Inhibitory effects of cytotoxin IL-2-PE40 on corneal allograft rejection

Objective To examine the inhibitory effect of IL-2-PE40 on the mouse corneal allograft rejection. Methods A mouse corneal graft model was set up by using C57BL/6 mice as donors and Balb/c mice as recipients. In the treatment group, IL-2-PE40 (0.6 μg/g body weight) was intraperitoneally injected from the day of surgery every 12 h until rejection happened. In the control group, the equal volume of PS was injected intraperitoneally at the corresponding time points. The transplanted cornea was observed under slit-lamp twice a week and the transplanted corneal opacity and neovascularization were rated according to Horis grading standards. It led to the determination of rejection response. The survival of transplanted cornea was regarded to be stopped when the rejection occurred. The operated eyes were observed historically on the 10th, 15th, 25th and 35th day after the surgery, and the peripheral blood was collected for measurement of T cell subgroups and T lymphocyte colonies. Results The survival time of cornea in the treatment and control groups was (30.2±2.9) days and (15.1±2.1) days respectively. In the control group, rejection occurred on the 15th day after the surgery, CD4~+ cells started rise right after the surgery and increased most obviously on the 15th day [(63.9±4.0)%] and decreased afterwards. CD4~+ cells in the treatment group were increased slightly [(42.6±4.0)%] on the 15th day. The number of CD4~+ cells in the treatment group was obviously less than in the control group (P<0.01). No significant changes in CD8~+ cells were observed in both groups. The number of T lymphocyte colonies in the control group was increased at the beginning and dropped then. No obvious change was found in the treatment group. The number of T lymphocyte colonies in the treatment group was significantly less than in the control group (P<0.01). Conclusion IL-2-PE40 is a highly specific immunosuppressive agent. It can delay the development of corneal graft rejection and reduce the percentage of T-helper cells significantly. It also works to weaken the forming capacity of the peripheral T lymphocyte colonies.
2.Apoptosis of MDA-MB-231 cells induced by berberine alpha-hydroxy beta-decanoylethyl sulfonate.

To investigate the cell proliferation inhibition and apoptosis induced by berberine a-hydroxy f-decanoylethyl sulfonate (HB) on MDA-MB-231 cells in vitro, and the inhibitory effect of HB on the expression of poly adenosine diphosphate RNA polymerase (PARP), MTT assay was used to detect the viability of MDA-MB-231 cells and cell cycle was examined by flow cytometry. The results showed that HB could significantly inhibit the proliferation of MDA-MB-231 cells, and mildly arrested cell cycle progression at S phase. The IC50S for 24, 48 and 72 h treatment were 4.65, 1.46 and 0.75 mg.L-1 (7.55, 2.37 and 1.22 micromol.L-1), respectively. Annexin V-FITC/PI double staining assay showed that HB increased apoptotic ratio of MDA-MB-231 cells. Western blotting analysis showed the expressions of procaspase-3, procaspase-9 and PARP were decreased after HB treatment, while their fragment increased. The results suggest that HB can inhibit the growth and induce apoptosis of MDA-MB-231 cells, which may be associated with inhibition of the expression of procaspase-3, procaspase-9 and PARP.

4.Clinical value analysis of serum protein fingerprint in patients with rheumatoid arthritis associated interstitial lung disease

Objective:To screen differentially expressed proteins in serum proteomic patterns in patients with Rheumatoid Arthritis associated Interstitial Lung Disease by Surface enhanced laser desorption /ionization mass spectrometry with Protein Chip ,and evaluate the value of a preliminary study.Methods: WCX2 ( Weak cation exchange chip ) and Surface enhanced laser desorption/ionization mass spectrometry with ProteinChip were performed to test serum proteins in 19 Rheumatoid Arthritis associated Interstitial Lung Disease,15 Rheumatoid Arthritis patients and 13 health adults,Biomarker Wizard ,Biomarker Pattern and Spss13.0 software were used in combination to analyze the data.Results:The mass/charge ratio ( M/Z) value of 142 protein peaks were detected in serum , there were 4 proteins differentially expressed with statistical significance among them ( P<0.05;Student′s t-test ) , which were the proteins with M/Z 3382.59 ,3453.39 ,11886.0 ,5825.48;the proteins with M/Z 11689.4 ,2266.49 ,1020.22 ,4392.28 ,5074.38 and 2764.69 were selected to establish the optimal diagnostic model.The sensitivity was of was 90%(9/10),the specificity was of 90%(18/20).Conclusion: Surface enhanced laser desorption/ionization time of flight mass spectrum can screen out the significantly different proteins in serum of patients with Rheumatoid Arthritis associated Interstitial Lung Disease.As bomarkers , both of their sensitivity and specificity were high and determination have good prospects for clinical application .
5.Treatment of chronic mallet finger deformity with minor bone anchors and palmaris longus tendon graft.

OBJECTIVETo explore the clinical effects of minor bone anchors and palmaris longus tendon graft in treating chronic mallet fingers deformity.
METHODSFrom January 2008 to June 2013, 26 patients with chronic mallet fingers deformity were treated with minor bone anchors and palmaris longus tendon graft. There were 18 males and 8 females, aged from 18 to 52 years old with an average of (32.0±1.3) years. Among them, 8 cases caused by machine injury, 6 cases by fall injury, 6 cases by sprain from fight, 4 cases by tendon spontaneous rupture, 2 cases by knife trauma. There was no tendon attachment of extensor tendon check in 16 cases, and with 0.3 to 0.5 cm tendon attachment in 10 cases. All patients had the flexion deformity and the disability of dorsiflexion activity. During operation, the distal interphalangeal joint was fixed in 10° to 20° dorsiflexion by a Kirshner wire, the minor bone anchor was used to reconstruct the extensor tendon insertion, the palmaris longus tendon slice was transplanted the decayed area of extensor tendon insertion. Four weeks postoperatively, the Kirshner wire was removed and the plaster external fixation was used, and the patient began function exercises. Postoperative complications were observed and fingers functions were assessed according to Dargan standard.
RESULTSThe patients were followed up from 6 to 14 months with an average of (5.0±0.3) months. Wound superficial infection occurred in 2 cases, the skin pressure ulcer in 2 cases, joint activities disability in 1 case; these symptoms got improvement after symptomatic treatment. Traumatic arthritis occurred in 2 cases, 1 case was improved after treatment, and 1 case had chronic pain for a long time. No internal fixation loosening or breakage and tendon rupture were found. According to Dargan standard to evaluate the finger function, 17 cases got excellent results, 8 good, and 1 poor.
CONCLUSIONIt is an effective way to treat the chronic mallet finger deformity using minor bone anchors and palmaris longus tendon graft, and the method has advantages of reliable fixation, easy operation, satisfactory effect and less complication.

6.Effect of Electroacupuncture on the Expression of Adenosine A1Receptor in Hypothalamus and Spinal Cord of Adjuvant Arthritis Rats

Objective To observe the effect of adenosine A1 receptor on pain and electroacupuncture analgesia, and to explore the action mechanism of electroacupuncture in analgesia.Method Adjuvant arthritis rats were taken as the study subjects. 24 rats were randomized into a normal group, a model group and an electroacupuncture (EA) group, 8 rats in each group. The pain threshold was evaluated by using thermal radiation method, immunohistochemical method and real-time fluorescence quantitative PCR were adopted to observe the expression of adenosine A1 receptor in hypothalamus and spinal cord.Result One day after modeling, the pain thresholds of right hind paw in the model group and EA group were significantly changed compared to that before modeling in the same group (P<0.01). The pain thresholds of right hind paw in the model group and EA group were significantly different from that in the normal group one day after modeling (P<0.01). 7 d after modeling, the pain threshold of right hind paw in the model group was still significantly lower than that before modeling in the same group (P<0.01), and it was significantly different from that in the normal group and EA group (P<0.01). The pain threshold was significantly enhanced in the EA group 7 d after modeling, and was significantly different from that of 1 d after modeling in the same group (P<0.01). The positive cell expression was lower in the model group and was significantly different from that in the normal group (P<0.01). The expression of adenosine A1 receptor in the EA group was markedly higher than that in the model group (P<0.01). The expression of adenosine A1 receptor in hypothalamus and spinal cord of the model group was significantly lower than that of the normal group (P<0.01). The expression of adenosine A1 receptor in hypothalamus and spinal cord of EA group was markedly higher than that of the model group (P<0.01).Conclusion EA can up-regulate the expression of adenosine A1 receptor in hypothalamus and spinal cord of adjuvant arthritis rats.
7.Effects of aminophylline used in the early stage of resuscitation on adenosine and vaso-active factors

8.The role of B7-H1mRNA in immune privilege of homogeneity

Objective To clarify the role of B7-H1 in the immune privilege after corneal transplantation in homogeneity variant mice.Methods We established the experimental animal model of allograft mice by using C57BL/6 mouse as donor and Balb/c mouse as recipient.We allocaated the mice with long time survival (>50 days) corneal graft into survival group,mice with rejection occurring in 50 days into rejection group,and normal C57BL/6 mice into control group.The transplanted corneal grafts were obtained for future reference at the 8th week after transplantation in survival group,and the time of rejection in rejection group.The expression of B7-H1 mRNA was detected by using immunohistochemistry and real time quantitative PCR (RT-PCR),and the relationship between B7-H1 and the immune privilege after corneal transplantation was analyzed. Results The B7 H1 mRNA was highly expressed in epithelium and endothelium of corneal grafts both in survival and control group,in comparison to an obviously lower expression in rejection group.The relative expression level of B7-H1 mRNA was 200.0 ± 11.5 in survival group,44.7 ± 10.8 in control group,and 6.9 ± 12.0 in rejection group,respectively. There were statistically significant differences among the three groups (F=241.164,P<0.01 ).The was a significant correlation between the level of B7-H1 mRNA and occurrence rate of rejection in corneal graft (P<0.01 ).Conclusion The results suggest that the immune privilege after corneal transplantation might be mediated by B7-H1,which plays an important role in maintaining the state of corneal immune privilege.
9.Effect of octreotide combined with compound coenzyme on Th17/CD4+CD25+Foxp3+ Treg imbalance in patients with purulent appendicitis

Objective To analyse the effect of octreotide combined with compound coenzyme on Th17/CD4 +CD25 +Foxp3 + regulatory T cells imbalance in patients with purulent appendicitis.Methods 150 patients with acute suppurative appendicitis were randomly divided into experimental group and control group, 75 cases in each group.Patients of two groups underwent appendicitis laparoscopic resection, then the control group were given conventional treatment and experimental group were given subcutaneous injection of octreotide on the basis of conventional treatment.Th17, CD4 +CD25 +Foxp3 +regulation T cells ( Treg) , and Th17 /Treg balance were analyzed.ResuIts Compared with control group, Th17/Treg imbalance of experimental group recovered better: the proportion of Th17, Treg, and Th17/Treg ratio decreased (P<0.05); the expression levels of related transcription factor, RORγt and Foxp3 mRNA decreased (P<0.05); serum cytokine (IL-4, IL-6, IL-10, IL-17, IL-23 and TGF beta) and C-reactive protein (CRP) content decreased (P<0.05);serum interferon gamma (IFN-gamma) content increased (P<0.05).ConcIusion Octreotide combined with compound coenzyme could correct Th17/Treg imbalance in suppurative appendicitis patients, improve the immune function of patients and affect the prognosis of patients.
10.Several Issues Need to be Resolved in Implementing Health Management

Health management plays an important role in the control of medical costs and in maintaining people' s health.Since the beginning of health management performance in our country,the major problems were:inadequacy of govement's support;imperfect laws;irregular marketing operation;lack of feasible health management model;controversy over long-term economic benefits;lack of awareness on health in common people and so on.To solve these problems,it is necessary to strengthen government support,and enhance people's health awareness,and explore the health management model consistent with the actual situation in China.
