1.Treatment of posterior column fracture in tibial plateau injury

Objective To describe the operative procedures.fracture patterns and clinical outcomes of treatment of posterior column tibial plateau fractures.Methods From February 2005 to August 2006,36 patients with posterior column tibial plateau fracture were operated on.Twenty patients were treated with open reduction and buttress plate fixation via a posterior approach.They were 13 males and 7 females,with a mean age of 38.5(ranging from 29 to 52)years.The other 16 patients were operated on via anterior plus posterior approaches.They were 11 males and 5 females,with a mean age of 37.3(ranging from 27 to 49) yeam. Results All were followed up for an average of 14.5(ranging from 12 to 15)months.All frac- tures got united with an average radiographic bone union time of 15.7(ranging from 11 to 16)weeks and an average full weight-bearing time of 17.6(range from 13 to 21)weeks.The mean HSS(The Hospital for special surgery)score of all these patients at 12 months postoperatively were 83.4(ranging from 68 to 92). There were 1 case of inferior medial genicular vessel injury during the operation,1 wound dehiscence and 1 partial incision necrosis postoperatively which were all eventually healed with conservative treatments.Three cases of numbness within the posterior lower part of the calf occurred postoperatively.No screw toggle,slide, dislodging or fixation failure was found during follow-up.There were no significant differences both between TPA and PA on radiographies and between immediately after operation and 12 months psstoperatively. Conclusion Open reduction and internal fixation with buttress plates via a posterior or anterior plus posterior approaches is appropriate for treatment of posterior column tibial plateau fractures.
2.Progress in the study of Her2-targeted cancer therapeutic antibodies.

Tumor surface antigen human epidermal growth factor receptor 2 (Her2) is a type I receptor tyrosine kinase, which belongs to human epidermal growth factor receptor family. Her2-overexpression is associated with tumorigenesis and metastasis. Due to significant clinical effects, Her2-targeted cancer therapy especially therapeutic antibody has become the hot spot in the field of cancer treatment. Anti-Her2 antibody drugs include monoclonal antibodies, antibody-drug conjugates, bispecific antibodies and emerging "two in one" antibody. Based on structure and function of Her2, this review focuses on recent advances in active mechanisms and clinical researches of these antibodies.

4.Correlation analysis between resting energy expenditure and nutritional assessment indicators in critically ill children with mechanical ventilation

Objective:To investigate the resting energy expenditure of children with mechanical ventilation and analyze their association with anthropometric indicators (height, weight, and body mass index) and serum protein markers (pre-albumin, albumin, transferrin, and retinol).Methods:An observational study was held in Pediatric Intensive Care Unit(PICU) of Beijing Children′s Hospital, Capital Medical University, from August 2015 to April 2016.Critically ill children with mechanical ventilation were enrolled in this study.Inclusion criteria were as follows: (1)pediatric critical illness score<90 scores or meet the United States PICU admission criteria; (2)age>29 days, <18 years old; (3)mechanical ventilation>24 hours; (4) tidal volume >60 mL.Resting energy expenditure was determined by US Med Graphic Company CCM/D energy metabolism test system.German Famed Zywier intensive care bed with weight scale weighing, uniform measuring bed or meter measuring height, automatic biochemical analyzer and radioimmunoassay was adopted for the determination of albumin, pre-albumin, transferrin, retinol binding protein.SPSS 20.0 software was applied for carrying out Pearson line analysis correlation. Results:A total of 68 children were included in the study, including 36 males and 32 females, body weight (22.34±14.89) kg, height (122.23±14.89) cm, resting energy consumption (784.63±278.25) kcal(1 kcal=4.185 kJ), and resting energy consumption per kilogram of body weight (48.80±31.26) kcal.In anthropometric indicators, resting energy expenditure was significantly positively correlated with height, body weight ( r=0.764, 0.734, all P<0.05), a low positive correlation with body mass index( r=0.396, P<0.05). In serum proteomics, resting energy expenditure was negatively correlated with pre-albumin, transferrin ( r=-0.318, -0.282, all P<0.05), and a moderate negative correlation with retinol protein ( r=-0.505, P<0.05), while without correlation with albumin ( r=-0.075, P>0.05). Conclusions:The height, weight and serum proteomics of children with mechanical ventilation may affect resting energy expenditure.
5.Development and application of a surgical information acquisition system based on performance assessment

7.Biomimetic synthesis and cytocompatibility of agar-hydroxyapatite composites

BACKGROUND: Studies regarding synthesizing composite bone repair materials with organic macromolecules as mineralizers template are the hot research spot. However, reports concerning using agar as bone repair materials are few. OBJECTIVE: To synthesize a novel agar-hydroxyapatite composite material, and to evaluate its physical and chemical properties as well as cytocompatibility.METHODS: ①A certain amount of the non-nano-hydroxyapatite in hydrochloric acid solution was added into a certain amount of agar sol, and the reaction system was adjusted with PH value of 7-8. And then the precipitate was lyophilized to obtain the composite of agar-hydroxyapatite.②The third generation of rat bone marrow stromal cell (BMSC) was co-cultured with agar-hydroxyapatite. And the growth of cells was observed at days 1, 3 and 5 after culture. RESULTS AND CONCLUSION: The results of X-ray diffraction patterns, Fourier transform infrared spectrometer (FTIR), thermal analyzer, transmission electron microscope, and scanning electron microscope showed that agar could manipulate the growth of hydroxyapatite, and hydroxyapatite nanocrystals was equably formed on the agar-fibers with high porosity. The BMSC grew well in the composite and form a clear cytoskeletal at days 3 and 5 after culture. The results reveal that agar-hydroxyapatite composite has good physical and chemical properties and cytocompatibility.
8.Construction of sharing type network resource data base for clinical skills teaching

The construction of a network resource data base for clinical skill teaching aiming at sharing excellent teaching resources was completed by integrating all kinds of teaching resource including characters,eourseware,pictures,cartoons,videos and examination questions,etc.In view of the generally weak situation of the current domestic education resource data base construction,the contents,guiding ideology,principle,object,orientation,function and the management mechanism of the network resource data base construction were devised for clinical skills teaching.Suggestions for specific ideas and construction problems were proposed to promote the construction and optimization of the network resource database for clinical skills teaching.
9.Influences of antenatal administration of taurine on cerebral neurogenesis of fetal rats with intrauterine growth restriction

Objective To provide the experimental evidence for clinical application of taurine,rat model of intrauterine growth restriction (FGR) was made to investigate influence of prenatal administration of taurine on neurogenesis.Methods Fifteen pregnant rats were divided into control,FGR model and taurine groups,5 rats for each group.Rats in the control group were supplied with unlimited food and drink while the other two groups were fed by 40% food intake of the control group throughout pregnancy.Since gestational day 12,taurine (100 mg/kg) was added into diet of taurine group every day until term delivery.Brain tissues were obtained immediately after baby rats were born.Expression of proliferating cell nuclear antigen (PCNA),neuron-specific enolase (NSE) and glial fibrillary acidic protein (GFAP) of brain tissue was measured by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR) and immunohistochemistry; meanwhile,numbers of PCNA-immunoreactive cells in subventricular zone,subgranular zone and cerebral cortex were compared with ANOVA test or q test.Results Levels of PCNA mRNA and GFAP mRNA expression in FGR group were significantly higher than those of control group (PCNA mRNA:1.002±0.011 vs 0.894 ± 0.040,P<0.01; GFAP-mRNA:1.012±0.013 vs 0.913±0.012,P<0.01).Compared to FGR model group,mRNA expressions of PCNA and GFAP in taurine groups were higher (1.090±0.029,P<0.01 ; 1.034±0.011,P>0.05).There was a significant decrease in the expression of NSE mRNA in FGR group compared with control group (0.796±0.036 vs 1.582±0.057,P<0.01),while the expression in taurine group (0.933±0.027) was significantly higher than that in FGR model group (P < 0.01).PCNA immunoreactive cells were mostly distributed in subventricular zone,followed by subgranular zone and cerebral cortex.Conclusions Prenatal application of taurine could enhance neurogenesis of FGR newborn rats and improve survival of neurons to ameliorate FGR brain damage.
10.Down-regulation of HIV-1 Infection by Inhibition of the MAPK Signaling Pathway

The human immunodeficiency virus type 1(HIV-1)can interact with and exploit the host cellular machinery to replicate and propagate itself.Numerous studies have shown that the Mitogen-activated protein kinase(MAPK)signal pathway can positively regulate the replication of HIV-1,but exactly how each MAPK pathway affects HIV-1 infection and replication is not understood.In this study,we used the Extracellular signal-regulated kinase(ERK)pathway inhibitor,PD98059,the Jun N-terminal kinase(JNK)pathway inhibitor,SP600125,and the p38 pathway inhibitor,SB203580,to investigate the roles of these pathways in HIV-1replication.We found that application of PD98059 results in a strong VSV-G pseudotyped HIV-1NL4-3 luciferase reporter virus and HIV-1NL4-3 virus inhibition activity.In addition,SB203580 and SP600125 also elicited marked VSV-G pseudotyped HIV-1NL4-3 luciferase reporter virus inhibition activity but no HIV-1NL4-3 virus inhibition activity.We also found that SB203580 and SP600125 can enhance the HIV-1 inhibition activity of PD98059when cells were treated with all three MAPK pathway inhibitors in combination.Finally,we show that HIV-1virus inhibition activity of the MAPK pathway inhibitors was the result of the negative regulation of HIV-1 LTR promoter activity.
