Protein degradation mediated by ubiquitin-proteasome pathway is involved in many biological processes.SCF complex is a sort of important E3 ubiquitin-ligase enzyme.Fbx4 protein is one of F-box protein family members,an important component of substrate-specific ScF Fbx4/αβ-crystallin ligase,and a subunit of SCF complex.Inactivation of Fbx4 protein or alteration of its expression level will influence activity of SCF Fbx4/αβ-crystallin ligase,thus will influence expression level of Cyclin D1.And Fbx4 protein is also an important regulatory factor for telomere maintenance.

OBJECTIVE:To investigate the use of antibiotics in our hospital.METHODS:The data regarding the use of antibiotics retrieved in July 2006 from 184 discharged histories were analyzed retrospectively.RESULTS:The application rate of antibiotics in our hospital reached 77.3%,of which,34.6% were used in combination of two kinds,and 32.6% were used in combination of three kinds.Only 5.9% of the specimens had been sent for pathologic examination.CONCLUSION:The application rate of antibiotics in the inpatients of our hospital as well as the postoperative use of antibiotics were all on the high side,while the rate of specimens sent for pathologic examination was on the low side,therefore,it is urgent to strengthen control on the rational use and standardized management of antibiotics.

Purpose To investigate the responsiveness of intracellular oxygen free radical and calcium on acrolein exposure and acrolein-induced cardiomyocytes apoptosis. Methods The viable adult mice cardiac myocytes were isolated by modified langendorff methods. We have examined the intracellular oxygen free radical and calcium concentration using DCF and Fura-2 AM, and the cardiomyocytes viability with WST assay. Are evaluated the DNA ladder pattern and cell apoptotic morphology on the adult mice cardiomyocytes that are exposed to acrolein. Results Our results show that acrolein can increase markedly the intracellular oxygen free radical and calcium concentration, that reach 12 fold and twofold respectively compared to the resting value when the cells were exposed to 1 μmol/L of acrolein. Moreover, the injury induced by acrolein in cardiac myocytes is concentration-dependent. The cardiomyocytes viability treated with 25, 50, 100 μmol/L of acrolein respectively were significantly lower compared to controls (P ＜ 0.01 ). DNA ladder pattern and apoptotic morphological changes were found after being exposed to acrolein in the adult mice cardiomyocytes. Conclusion It is concluded that acrolein induces adult mice cardiomyocytes apoptosis, and it may be due to the increased intracellular oxygen free radicals and calcium concentration.

Objective To discuss the diagnosis,differential diagnosis and treatment of obstructive jaundice in infants.Methods Infants with obstructive jaundice were examined by ECT to obtain dynamic images of the liver,biliary tract and intestine to find out the position and nature of the obstruction.Results Eight patients with delayed nuclide imagining were diagnosed as obstructive jaundice of cholestasis.Five of which with aggravated jaundice were cured by biliary irrigation and 3 by medication.For 16 patients with congenital biliary atresia,10 were exterior hepatic types and 6 interior hepatic types.They had the nuclide image of liver and biliary tract.Ten cases of the exterior hepatic types and four cases of the interior types received the Kasai operation.And bile was drained after the surgery.Seven patients survived and 4 of interior heaptic types died postoperatively.Due to bile fistula,3 patients died after the hepato-porto-jejunostomy for internal drainage.Two patients of interior hepatic types who had no operation died of the hepatic failure.Conclusion The nature and position of obstructive jaundice could be identified by nuclide imaging.Young patients with jaundice of cholestasis could be treated by antiinflamation,liver function protection,Hymecromone and Dexamethasone for two weeks.Once the serum bilirubin rises,operation would be indicated.In the case of congenital biliary atresia,operation should be done earlier on the basis of medication.

Objective To compare the therapeutic effects of 1470nm diode laser vaporization prostatectomy and bipolar transurethral plasmakinetic prostatectomy (TUPKP) for treatment of benign prostatic hyperplasia (BPH).Methods From June 2015-February 2016 a total of 95 patients diagnosed with BPH were randomly divided into 2 group:52 patients were treated with TUPKP while 43 patients with 1470 diode laser.All patients were followed up with mean operative time,intra-operative blood loss,postoperative hospital stay,postoperative catheterization time,postoperative complications,international prostate symptom score (IPSS),score of life quality (QoL),maximum flow rate (Qmax),post-void residual (PVR) before and after surgery.Results Compared with data of preoperation,postoperative IPSS,QoL,Qmax,PVR in 6 months revealed significant improvement in both of two groups.IPSS of TUPKP group and 1470 group respectively dropped to 6.3 ± 2.6 and 6.7 ± 2.4.In TUPKP group,PVR reduced to 23.1 ± 20.9ml and in 1470 group decreased to 24.3 ± 19.9ml.While Qmax increased to 18.5 ± 3.1 ml/s and 18.7 ± 2.8ml/s respectively in TUPKP and 1470 group.In TUPKP group,operation time were 60.1 ± 14.9min,significantly less than that in 1470 group of 69.3 ± 12.9min (P ＜ 0.05).Compared with catheterization time in 1470 group of 33.9 ± 9.4h,the time in TUPKP group of 73.9 ± 37.6h was shorter with significant difference (P ＜ 0.05).While Curative effect of two groups of showed no significant difference (P ＞ 0.05).Conclusion The clinical curative effect of two operation methods for patients with BPH showed no significant difference.1470 group had longer operation time,while TUPKP group had less intraoperative bleeding and postoperative recovery.

Objective To investigate the characteristics of human immunodeficiency virus(HIV-1)V3 loop amino acid mutations among AIDS patients in Shenzhen. Methods Fragments of HIV-1 env gene were amplified by RT-nested-polymerase chain reaction(PCR)from plasma of the AIDS patients in Shenzhen.The PCR products were then directly sequenced and the sequences were aligned,translated and analyzed. Results Sequence analysis showed that there were B and CRF01-AE HIV-1 subtypes in Shenzhen.The V3 loop terra peptide GPGQ accounted for about 48%,GPGR about 36%,other V3 loop terra peptide forms were 16%;the V3 loop central motif in the majority(76.9%)of the CRF01-AE strains was GPGQ,the majority(75%)of the B strains was GPGR.We also found some unusual terra peptide compositions:DQDR and DQGQ on the tip of V3 loop,some amino acid mutations at specific of V3 loop associated with SI phenotype were very common in AIDS patients(80%). Conclusions The V3 loop amino acid mutations were very common in AIDS patients,the V3 loop central motif in the majority was GPGQ and GPGR in Shenzhen,the mutations shows the change of HIV-1 phenotype and predicts that disease progress to AIDS.

Objective To investigate the MRI features and the diagnostic value of uterine leiomyoma.Methods All cases were examinated by MRI and ultrasound.Retrospective analysis of MRI manifestations of 71 uterine leiomyomas in 32 patients in combination with the results of operation and pathology was carried out and was compared with ultrasound findings.Results The MRI features of the tumors were:intermediate or slightly low signal intensity on T 1WI and low signal intensity on T 2WI,patch hyperintense areas were seen in degenerated leiomyoma on T 2WI,there were clear margins in all tumors and capsules in most tumors(55/71).MRI was superior to sonography in showing 

Objective To observe the changes of the content of ATP,lactate and the activity of phosphofructokinase(PFK) in perihemotoma region in rabbit brain after intracerebral hemorrhage. Methods(The rabbits) were divided into 3 groups: normal group(N),hemorrhage group (H) and the sham group(S). The H and S groups were subdivided into 1 h, 6 h, 12 h, 24 h, 48 h and 72 h groups. A model of cerebral hemorrhage in rabbit with autologous blood infusion was set. The contents of ATP, lactate and water in perihemotoma region were measured and the metabolite concentration in edematous brain regions were corrected for dilution. The activity of phosphofructokinase was also determined.Results There was obvious edema in perihemotoma region at 1 hour postinfusion and getting more serious at 72 hours. ATP was moderately decreased at 1 hour, and remained in a similar level until 12 hours. There appeared another evident decrease in 24 hours [(13.29?2.92) ?g/g, about 58% of N group], and no obvious changes from 24 to 72 hours. The content of lactate increased in 1 hour and reached the peak at 12 hours [(21.01?0.18) ?mol/g]. Until 72 hours , the level of lactate [(12.89?0.25) ?mol/g] in hemorrhage group reach higher than in the normal and sham group. The activity of phosphofructokinase was decreased at 1 hour [(3.98?0.02) U/mg] and remained in a decreasing tendency to 72 hours. Conclusion The failure of energy metabolism occurs in 24 hours postinfusion,coming late somewhat to the brain edema. The result might be related to the changes of some enzymes, maybe a key role in energy metabolism.

Objective To study the content of IL-8 in the peripheral blood and the expression of its mRNA in liver biopsy tissue of patients with hepatocirrhosis.Methods The patients with typical hepatocirrhosis resulted from HBV infection were selected.The IL-8 concentrations in peripheral blood of the Datients were evaluated with ELISA.The content of IL-8 mRNA in liver biopsy was measured by real-time PCR.The level of IL-8 mRNA was surrogated by lg cDNA/lg GAPDH.Reslllts The levels of IL-8 in the serum and its mRNA in hepatic tissue of patients with hepatoeirrhosis were(43 1.39±97.39)μg/L and 1.3647±0.2203,respectively,which were significantly higher than those of control group.MoreoCer,the levels of IL-8 in the serum and IL-8 mRNA in hepatic tissue of patients with positive HBV-DNA were (502.43-4-102.24)μg/L and 1.51424±0.2245,respectively,and both ofwhich were higher than those of patients without HBV-DNA replication(P<0.0l,P<0.05).And the levels of IL-8 in the selqlm and its mRNA in hepatic tissue of patients with higher ALT.AST also increased significantly.compared with those in control group(P<0.01).Conclusion The levels of IL-8 in the peripheral blood and the IL-8 mRNA in liver biopsy tissue of patients with hepatocirrhosis increase.which are related to the degree of HBV-DNA replication and the damage of hepatocytes.The IL-8 plays a key role in local inflammatory infihration of liver and the progression of post-hepatitis cirrhosis.
